Molecular markers (PCR–RFLP and microsatellite) were used to investigate the genetic background of the only brown trout (Salmo trutta m fario) broodstock in Hungary which due to the hydrogeography of the country should theoretically belong to the Danubian lineage. PCR–RFLP (mitochondrial DNA control region and lactate dehydrogenase and somatolactin genes) as well as microsatellite (BFRO002, OMM1064, Ssa408uos, SsoSL417, SsoSL438) markers were used to distinguish between Danubian and Atlantic lineages of brown trout in the Lillafüred broodstock. Altogether 435 fish were tagged during the experimental period. Due to mortalities, fin clips were collected from 401 individuals. According to the genetic analysis of the mitochondrial DNA, the Danubian haplotype is present in only one individual (0.2 %) of the broodstock. Analysis of the nuclear markers revealed that alleles characteristic of both the Atlantic and the Danubian lineages are found in the population. However, Atlantic alleles dominate throughout the broodstock which is in Hardy–Weinberg equilibrium according to the investigated markers. Results indicate that the original broodstock that was introduced to the farm following its construction in 1933 was of the Atlantic lineage. Although later fish from a local stream were collected and added to the broodstock, the number of these was limited and they were almost exclusively males. Fish from this farm that are stocked by anglers can have a significant genetic impact on trout populations of natural streams. 相似文献
Soil organic carbon (SOC) pool has the potential to mitigate or enhance climate change by either acting as a sink,or a source of atmospheric carbon dioxide (CO2) and also plays a fundamental role in the health and proper functioning of soils to sustain life on Earth.As such,the objective of this study was to investigate the applicability of a novel evolutionary genetic optimization-based adaptive neuro-fuzzy inference system (ANFIS-EG) in predicting and mapping the spatial patterns of SOC stocks in the Eastern Mau Forest Reserve,Kenya.Field measurements and auxiliary data reflecting the soil-forming factors were used to design an ANFIS-EG model,which was then implemented to predict and map the areal differentiation of SOC stocks in the Eastern Mau Forest Reserve.This was achieved with a reasonable level of uncertainty (i.e.,root mean square error of 15.07 Mg C ha-1),hence demonstrating the applicability of the ANFIS-EG in SOC mapping studies.There is potential for improving the model performance,as indicated by the current ratio of performance to deviation (1.6).The mnapping also revealed marginally higher SOC stocks in the forested ecosystems (i.e.,an average of 109.78 Mg C ha-1) than in the agro-ecosystems (i.e.,an average of 95.9 Mg C ha-1). 相似文献
The effect of heavy metals on the motility parameters of common carp sperm was investigated. In vitro test systems are widespread in ecotoxicology, and fish sperm can be a suitable model. For this reason, studies had been carried out in this topic; however, the published methods are not standard in several aspects (donor species, measured endpoint, etc.). In this study, a previously published toxicology-aimed sperm analysis protocol was tested to examine the effect of heavy metals (arsenic, cadmium, chromium, copper, mercury, nickel, zinc,) on common carp sperm. According to our results, PMOT is the most sensitive of the investigated parameters: dose-response was observed in case of each metal at low concentrations, already after 30 min of exposure. VCL was less sensitive: lower effects were observed at the same concentrations compared to PMOT. Among the examined parameters, LIN was the least affected: a dose-response was observed only in case of arsenic and mercury. The same sensitivity of motility parameters was observed on zebrafish sperm previously. Moreover, we found that PMOT, VCL, and LIN of common carp sperm were affected at the same concentrations as it had been observed in zebrafish, when the identical analytical protocol was applied. The only exception was As3+, where common carp sperm proved to be more sensitive: lower concentrations already reduced its motility parameters. Consequently, PMOT of common carp sperm is an accurate and fast bioindicator of aquatic pollution. 相似文献
Deficient trace elements concentration in soils, forages, and animals have been reported in several areas of Balkan region. Main challenge in overcoming low productivity of forage and animal production in this region is the lack of data on the nutritional status of the pastures and soils. This study examined the nutrient and pseudo total concentration of trace elements in soil and herbage plants, and the potential deficiency or excess with regards to crop and livestock production. Soil and plant samples from 100 sampling points were collected in the mountainous grasslands of Manja?a (between longitudes 16°56′ and 17°8′ east; and latitudes 44°33′ and 44°42′ north) and Vla?i? (between longitudes 17°14′ and 17°29′ east; and latitudes 44°25′ and 44°37′ north). Soil samples were analysed for soil texture, pseudo total concentration (5?ml HNO3) of trace elements (TE), pH, soil organic carbon (SOC), while plant samples were analysed for TE. The soil pH varied from strongly acidic to moderately alkaline. The concentration of SOC varied from 0.5% to 12.3%. Most of the soil samples were finer-textured soils high in silt content. The average concentrations of Na, P, Zn, Se, Cu, Co, and B were low in both soil and herbage plants. Plant K, Ca, Mg, Mo, and Mn concentrations were sufficiently high to meet the requirements of grazing animals, while Fe concentrations was elevated in certain areas. High levels of Mo were found in both soil and plants. The results suggest that imbalances observed in natural pastures of Manja?a and Vla?i? area, caused by low soil TE concentration and other soil and plant properties, could contribute to poor animal nutrition. 相似文献
Seeds enriched with zinc (Zn) are ususally associated with better germination, more vigorous seedlings and higher yields. However, agronomic benefits of high‐Zn seeds were not studied under diverse agro‐climatic field conditions. This study investigated effects of low‐Zn and high‐Zn seeds (biofortified by foliar Zn fertilization of maternal plants under field conditions) of wheat (Tritcum aestivum L.), rice (Oryza sativa L.), and common bean (Phaseolus vulgaris L.) on seedling density, grain yield and grain Zn concentration in 31 field locations over two years in six countries. Experimental treatments were: (1) low‐Zn seeds and no soil Zn fertilization (control treatment), (2) low‐Zn seeds + soil Zn fertilization, and (3) Zn‐biofortified seeds and no soil Zn fertilization. The wheat experiments were established in China, India, Pakistan, and Zambia, the rice experiments in China, India and Thailand, and the common bean experiment in Brazil. When compared to the control treatment, soil Zn fertilization increased wheat grain yield in all six locations in India, two locations in Pakistan and one location in China. Zinc‐biofortified seeds also increased wheat grain yield in all four locations in Pakistan and four locations in India compared to the control treatment. Across all countries over 2 years, Zn‐biofortified wheat seeds increased plant population by 26.8% and grain yield by 5.37%. In rice, soil Zn fertilization increased paddy yield in all four locations in India and one location in Thailand. Across all countries, paddy yield increase was 8.2% by soil Zn fertilization and 5.3% by Zn‐biofortified seeds when compared to the control treatment. In common bean, soil Zn application as well as Zn‐biofortified seed increased grain yield in one location in Brazil. Effects of soil Zn fertilization and high‐Zn seed on grain Zn density were generally low. This study, at 31 field locations in six countries over two years, revealed that the seeds biofortfied with Zn enhanced crop productivity at many locations with different soil and environmental conditions. As high‐Zn grains are a by‐product of Zn biofortification, use of Zn‐enriched grains as seed in the next cropping season can contribute to enhance crop productivity in a cost‐effective manner. 相似文献
Long-term microscale field experiments established at four sites in the Czech Republic (since 1996) were used for an assessment of the severity of the wide-scale contamination of treated soils by persistent organic pollutants (POPs) and pharmaceuticals and personal care products (PPCPs) as a result of the long-term regular pollutant load via sewage sludge and farmyard manure applications and for an estimation of the potential environmental risk caused by long-term pollutant inputs to agricultural soils.
Material and methods
The experimental plots were treated as follows: (i) NPK mineral fertilization (NPK); (ii) sewage sludge (SS1); (iii) sewage sludge applied at three times the rate compared to SS1 (SS2); (iv) farmyard manure (FYM); and (v) untreated control. Except for antibiotics, which showed results for all analyses below the detection limit, all groups of the analyzed compounds showed measurable contents in the treated soils.
Results and discussion
Among the POPs, the levels of organochlorine pesticides (OCPs) were detected at two locations at levels exceeding the Czech preventive values for their contents in agricultural soils (7.5 μg/kg for dichlorodiphenyl trichloroethane family, 20 μg/kg for hexachlorbenzene, and 10 μg/kg for α-hexachlorocyclohexane), regardless of the treatment. Similarly, elevated contents of polycyclic aromatic hydrocarbons (PAHs) were occasionally observed without any substantial relationship to the fertilizer application scenario. Thus, these contaminants are connected with atmospheric deposition in the given areas (PAHs) and their long-term stability (OCPs) even several decades after their ban; the role of the fertilizer composition was negligible.
Conclusions
The levels of per- and polyfluoroalkylated substances, brominated flame retardants, and synthetic musk compounds tended to increase in the sewage sludge–treated plots, indicating that sewage sludge can contribute to the abundance of these compounds in soil, although the contaminant levels determined do not represent a direct environmental risk. The levels of these contaminants in sewage sludge and sludge-treated soils should be regularly monitored in further research.
Among the multiple functions, which have been identified for the neonatal Fc receptor (FcRn), we study its role in the IgG transport in the mammary gland during the colostrum formation. For this reason, we have obtained several mammary gland biopsies from a pregnant sheep around parturition. The presence of the FcRn heavy chain mRNA was detected exclusively in the acinar and ductal epithelial cell by in situ hybridization (ISH). We detected strong signal in samples harvested 24 and 10 days prepartum; however, in samples we collected postpartum was barely detectable. Immunohistochemistry confirmed our ISH data. The cytoplasm of the epithelial cells of the acini and ducts in the mammary gland biopsies stained homogeneously before parturition, although a remarkable difference was observed in the pattern after lambing. The signal indicated uneven distribution of the FcRn alpha chain in the epithelial cells 1 and 5 days postpartum, since the apical sides of the epithelial cells were highlighted. The presence of the FcRn in the acinar and ductal epithelial cells and the obvious change of its distribution before and after parturition suggest that FcRn plays an important role in the IgG transport during colostrum formation. FcRn expression was also found in the lamb duodenal crypt epithelial cells, which have been previously demonstrated to secrete IgG1 in newborn ruminants, suggesting secretory role of the FcRn in ruminant epithelial cells. 相似文献

Antioxidant properties of marjoram (Origanum majorana L.) herb and extracts obtained with ethanol, n-hexane, and supercritical CO2 extraction are presented. Individual antioxidants, ursolic acid, carnosic acid, and carnosol, were quantified with high-performance liquid chromatography. The effects of different parameters (temperature and pressure) of high-pressure extraction on the yield of carnosol were studied. Furthermore, two marjoram herbs from Hungary and Egypt were compared measuring hydrogen-donating abilities with 1,1-diphenyl-2-picrylhydrazyl by spectrophotometric and the total scavenger capacities by chemiluminometric methods from the aqueous extracts of the herbs. The antioxidant activities of the solvent extracts were performed using the Rancimat method. The Egyptian herb and its extracts possessed better antioxidant activities than Hungarian ones. Applying supercritical CO2 extraction, the highest value of carnosol was obtained at 400 bar and 60 degrees C. 相似文献
