This paper reports the observed formation of a secondary corpus luteum (CL) in the presence of the cyclic corpus luteum, on the ovaries of a cow after ultrasound-guided follicular aspiration for oocyte recovery. The secondary structure, although smaller and lighter (4.97 g vs. 6.02 g) than the natural one, had the typical macroscopic appearance of a corpus luteum. Histological examination of the structure using electron microscopy revealed typical structural features of a natural CL. Mean tissue progesterone concentration was significantly lower in the secondary CL (31.15 +/- 3.11 compared with 58.29 +/- 6.32 micrograms/g tissue of the cyclic CL) and oestradiol-17 beta significantly higher than in the natural CL (108 +/- 11.6 compared with 74.2 +/- 7.81 pg/g tissue). P450scc and P450(17 alpha) mRNA was detected in both structures while P450arom and full-length mRNA FSH receptor were detected only in the secondary structure.  相似文献

Five fast-neutron-derived mutants were isolated from the wheat line Hobbit 'sib' that show enhanced field resistance towards Puccinia striiformis f.sp. tritici , the causal agent of yellow rust. Subsequent testing showed the yellow rust resistance phenotypes to differ between mutants, to be expressed at different growth stages and, in some cases, to show an isolate interaction. Three mutants, I3-48, I3-49 and I3-54, exhibited an enhanced yellow rust resistance phenotype from the third seedling leaf onwards, while mutants I3-27 and I3-30 did not show an altered yellow rust phenotype until later growth stages. Additional resistance for brown rust (causal agent Puccinia triticina ) was identified in mutants I3-27, I3-30, I3-48 and I3-49, and for powdery mildew caused by Blumeria graminis f.sp. tritici in mutants I3-27, I3-30, I3-48 and I3-54, although in some cases the resistance was isolate-specific.  相似文献

The carboxyterminal processing protease of D1 protein (CtpA) is predicted to be an excellent target for the discovery of a general broad-spectrum herbicide. Directed and random screening of compounds against recombinant spinach CtpA (rCtpA) has led to the discovery of five different chemical classes of inhibitors. Lead compounds from each inhibitor class were investigated for their in vitro effects on the activities of both recombinant and native spinach CtpA. All of the lead compounds have Ki values of less than 50 μM when tested against rCtpA, and all except one showed competitive inhibition. Results from partially purified native CtpA from spinach were similar to those from the recombinant form of the enzyme, thus validating the use of rCtpA in the inhibitor screen. Compounds from three of the classes of CtpA inhibitors show in vivo herbicidal activity against Arabidopsis thaliana when applied either by addition to growth media or by spraying the leaves. Transgenic Arabidopsis plants which over-express CtpA showed greater resistance to the compounds than wild-type plants providing evidence that these inhibitors are directly acting against CtpA.  相似文献

ABSTRACT The effect of activated charcoal as an amendment to water agar medium on teliospore germination was analyzed for two species of wheat-infecting bunts, Tilletia controversa and T. tritici, and two related wild-grass infecting species, T. bromi and T. fusca. Final percentages of teliospore germination, area under the germination progress curves (AUGPC), and a standardized AUGPC (SAUGPC) on carbon agar and water agar were compared among strains. Carbon agar (CA) significantly increased the final germination percentage of teliospores, AUGPC, and SAUGPC when compared with water agar (WA) for all taxa under study. Additionally, CA reduced significantly the incubation (i.e., lag) period when compared with WA for teliospores of T. bromi, T. controversa, and T. fusca. Bovine serum albumin and polyvinyl pyrrolidone were used as alternative chemical adsorbent amendments to WA to establish the role of activated charcoal in the medium. Only media amended with bovine serum albumin and activated charcoal improved the final germination percentage of all taxa. Polyvinyl pyrrolidone was not significantly better than water agar.  相似文献

Among the 57 monoclonal antibodies analyzed within the T-cell group of the Second International Swine CD Workshop, one mAb fell within cluster T14a that included the CD6 standard a38b2 (No. 175). The new mAb MIL8 (No. 082) and a38b2 both precipitated from activated T-cells a 150 kDa monomeric protein. Staining patterns on the various cell types were similar. There was no inhibition of binding of either mAb to peripheral blood T-cells with the opposite mAb. The new mAb, MIL8, reacts with a separate epitope on porcine wCD6.  相似文献

Based on an analysis of their reactivity with porcine peripheral blood lymphocytes (PBL), only three of the 57 mAbs assigned to the T cell/activation marker group were grouped into cluster T9 along with the two wCD8 workshop standard mAbs 76-2-11 (CD8a) and 11/295/33 (CD8b). Their placement was verified through the use of two-color cytofluorometry which established that all three mAbs (STH101, #090; UCP1H12-2, #139; and PG164A, #051) bind exclusively to CD8+ cells. Moreover, like the CD8 standard mAbs, these three mAbs reacted with two proteins with a MW of 33 and 35 kDa from lymphocyte lysates and were, thus, given the wCD8 designation. Because the mAb STH101 inhibited the binding of mAb 76-2-11 but not of 11/295/33, it was given the wCD8a designation. The reactivity of the other two new mAbs in the T9 cluster with the various subsets of CD8+ lymphocytes were distinct from that of the other members in this cluster including the standards. Although the characteristic porcine CD8 staining pattern consisting of CD8low and CD8high cells was obtained with the mAb UCP1H12-2, a wider gap between the fluorescence intensity of the CD8low and CD8high lymphocytes was observed. In contrast, the mAb PG164A, not only exclusively reacted with CD4/CD8high lymphocytes, but it also failed to recognize CD4/CD8 double positive lymphocytes. It was concluded that this mAb is specific for a previously unrecognized CD8 epitope, and was, thus, given the wCD8c designation. A very similar reactivity pattern to that of PG164A was observed for two other mAbs (STH106, #094; and SwNL554.1, #009). Although these two mAbs were not originally positioned in the T cell subgroup because of their reactivity and their ability to inhibit the binding of PG164A, they were given the wCD8c designation. Overall, five new wCD8 mAbs were identified. Although the molecular basis for the differences in PBL recognition by these mAbs is not yet understood, they will be important in defining the role of CD8+ lymphocyte subsets in health and disease.  相似文献

Among the 57 monoclonal antibodies (mAb) analyzed within the T-cell group from the Second Swine CD Workshop, six mAb fell within clusters T10 and T11 (No. 088, STH164; No. 148, FY1A3; No. 149, FY2C1; No. 150, FY1H2; No. 151, FY2A11; No. 169, BB23-8E6). The mAb within these two groups gave a similar appearance on flow cytometry and stained all peripheral blood T-cells as defined by CD4 and wCD8 staining. All six mAb precipitated a 24 kDa protein. On the basis of inhibition analyses performed as part of the workshop and from published data, the mAb define at least three epitopes. There is only minimal stimulation of resting peripheral lymphocytes, but four of the mAb produce strong stimulation in the presence of PMA. With the exception of STH164, all have been shown to react with CD3-transfected COS cells. The new mAb, therefore, react with three epitopes on porcine CD3 designated CD3a (BB23-8E6, FY2A11), CD3b (FY1A3, FY2C1), and CD3c (FY1H2). mAb STH164 appears to be reactive with another epitope, however, since its reactivity with CD3 has not been confirmed it is designated as wCD3.  相似文献

Forty-five soil samples were collected from aquaculture areas in 23 provinces of Thailand to include six soil orders and wide variation in physical and chemical properties. Soil-water microcosms were prepared containing 5 g of soil and 150 mL of distilled water. Microcosms were held on an oscillating table shaker (150 rpm) for 1 wk at 25 C in the dark. Water pH and concentrations of dissolved nutrients, total alkalinity, and total hardness were measured. Differences in properties within soil orders caused wide variation in composition of solutions and differences in concentrations of dissolved substances and pH were not related to soil order. Regression analyses revealed significant correlations between concentrations of soil nutrients extractable in dilute acid (0.05 N HCI plus O.025 N H2SO4) or in neutral, 1 N ammonium acetate and aqueous concentrations. Regression coefficients usually were higher for dilute-acid extractable nutrients than for ammonium acetate extractable ones. Regression coeficients based on dilute-acid extractable nutrients follow: soluble reactive phosphorus ( r = 0.816); calcium ( r = 0.685); magnesium ( r = 0.470); potassium ( r = 0.959); sodium ( r = 0.977); manganese ( r = 0.462); boron ( r = 0.399). The correlation between soil and solution iron was not significant and aqueous concentrations of copper and zinc were below detection limit. Hardness was correlated with soil carbon ( r = 0.710) and soil pH was a good predictor of alkalinity ( r = 0.877). Soil pH and aqueous pH were highly correlated (r = 0.939). Findings suggest that soil characteristics can be used to predict pH and concentrations of several dissolved substances in soil-water systems under aerobic conditions.  相似文献

Thirty-two pigs (1 d old) were used to determine if exogenous glucagon and(or) free fatty acids (FFA oleic acid) would enhance gluconeogenesis and glucose homeostasis during fasting. Pigs were acquired at birth, fitted with an indwelling arterial cannula (via umbilicus) and fasted 24 h to deplete liver glycogen. A jugular cannula was inserted nonsurgically 8 to 10 h before initiation of a primed-continuous infusion consisting of control (excipient), glucagon (Glu), oleic acid (FFA), or both glucagon and oleic acid (Glu-FFA). Plasma Glu averaged 395 pg/ml preinfusion and was similar across treatments. The concentration increased fivefold (P less than .05) by 80 min for Glu and Glu-FFA pigs and remained constant thereafter (160 min: 2,379, 2,258 pg/ml; 240 min: 2,355, 2,274 pg/ml, respectively). Glucagon infusion did not alter plasma glucose after 240 min of infusion (control, 50 vs Glu, 51 mg/dl); however, Glu-FFA effected an increase (60 mg/dl, P less than .10). In contrast, pigs infused with FFA alone had a lower glucose concentration (40 mg/dl, P less than .10). Rate of glucose synthesis was determined using liver slices, acquired immediately postinfusion, with alanine and lactate as substrate (7.5 mM). The rate of synthesis was not altered by Glu or Glu-FFA infusion (2.91, 2.43 mumol glucose X g-1 X h-1 vs 2.91 for control). In contrast, exogenous FFA reduced synthesis to 1.85 mumol glucose X g-1 X h-1 (P less than .05) with lactate as substrate.
