The present study investigated the effect of outdoor rearing and genetic background on behaviour and meat quality traits in heavyweight pigs. Large White and Duroc-sired pigs were reared in groups of 3 castrated males and 3 females, conventionally or in fields, from April to October, in a study with two replicates. Compared to indoor pigs, outdoor pigs were more active and showed a larger range of behaviour suggesting improved welfare from a behavioural point of view. During 5 h of isolation, outdoor pigs were less active and had lower heart rates, and in another test, they reacted less to a non-familiar object (traffic cone). Pigs were slaughtered at 150 kg live weight, half of each treatment group after mixing, short transport, and overnight lairage and half immediately following short transport. Outdoor pigs were less aggressive during pre-slaughter mixing. Ante- and post-mortem glycogen content of the Longissimus lumborum (LL), Semimembranonus (SM) and Semispinalis capitis (SC) muscles depended on slaughter conditions, sire breed, rearing conditions, gender and year of experimentation, sometimes influencing ultimate pH. Effects of rearing on muscle glycogen content and post-mortem pH were stronger during the second year of rearing. Effect of slaughter conditions on glycogen content and ultimate pH depended on fighting levels during pre-slaughter mixing. Outdoor rearing increased muscle redness. Drip and cooking loss were higher in Large White than Duroc-sired pigs. Drip and cooking loss were correlated with early post-mortem and ultimate pH. Thawing loss was correlated with early post-mortem temperature and ultimate pH. Finally, reactivity to isolation had a predictive value as pigs more active during isolation had less skin damage due to fighting during pre-slaughter mixing. In conclusion, despite large effects of year of experimentation, outdoor rearing, sire breed, and slaughter conditions influenced behaviour and muscle characteristics.  相似文献

This study examined the effect of treating mares with equine pituitary extract (EPE) in combination with human chorionic gonadotropin (hCG; EPE/hCG) on the recovery rate of immature oocytes by ovum pick-up (OPU) and on oocyte morphology. Ten mares were subjected to each of two treatments in a random sequence: superstimulated with EPE (25 mg, twice daily) and treated with hCG (2,500 IU) or control (no exogenous treatment). The cytoplasmic morphology of oocytes recovered was evaluated through transmission electron microscopy. Follicular fluid was collected at aspiration for progesterone analysis, which was performed by radioimmunoassay. The EPE/hCG did not increase the oocyte recovery rate from immature follicles when compared with the controls (15.5% and 16.7%, respectively). A significantly higher oocyte recovery rate per mare was observed (70% versus 50%). However, precocious granulosa cell expansion was observed with EPE/hCG treatment in contrast to the control (64.4% and 33% of follicles with expanded cumulus, respectively), and increased intrafollicular progesterone concentration was also seen (158.80 ng/mL versus 82.05 ng/mL). The ultrastructural analysis of oocytes from both groups showed morphologic features related to immaturity. Numerous vesicles containing cortical granules were found, distributed in clusters into the cytoplasm, and junctional complexes were still seen between oocyte and granulosa cells. In conclusion, EPE/hCG treatment induced some follicular modifications, but the recovery rate was not increased. All oocytes examined presented signs of immaturity.  相似文献

This study quantitatively investigated the analgesic action of a low-dose constant-rate-infusion (CRI) of racemic ketamine (as a 0.5 mg kg−1 bolus and at a dose rate of 10 μg kg−1 min−1) in conscious dogs using a nociceptive withdrawal reflex (NWR) and with enantioselective measurement of plasma levels of ketamine and norketamine. Withdrawal reflexes evoked by transcutaneous single and repeated electrical stimulation (10 pulses, 5 Hz) of the digital plantar nerve were recorded from the biceps femoris muscle using surface electromyography.Ketamine did not affect NWR thresholds or the recruitment curves after a single nociceptive stimulation. Temporal summation (as evaluated by repeated stimuli) and the evoked behavioural response scores were however reduced compared to baseline demonstrating the antinociceptive activity of ketamine correlated with the peak plasma concentrations. Thereafter the plasma levels at pseudo-steady-state did not modulate temporal summation. Based on these experimental findings low-dose ketamine CRI cannot be recommended for use as a sole analgesic in the dog.  相似文献

Impaired insulin sensitivity is increasingly recognised in cats, but sequences of genes involved in insulin-signalling are largely undetermined in this species. In this study, extended feline mRNA sequences were determined for the adiponectin, glucose transporter-1 (GLUT1), GLUT4, peroxisome proliferative activated receptor-gamma1 (PPARgamma1), PPARgamma2, plasminogen activator inhibitor-1 (PAI-1), monocyte chemoattractant protein-1 (MCP-1) and insulin receptor genes. Conserved dog-specific primers identified from human-dog mRNA alignments were used to amplify feline cDNA in the polymerase chain reaction (PCR). The feline sequences determined by this method were used to design feline-specific primers suitable for real-time PCR for quantification of gene expression in insulin sensitive tissues of healthy cats. Partial sequences of feline mRNAs had 86-95% identity with dog and human genes. Expression of adiponectin, GLUT1, GLUT4, PPARgamma1, PPARgamma2, PAI-1 and insulin receptor mRNA was detected and quantified in subcutaneous and visceral fat and skeletal muscle, whereas MCP-1 mRNA was detected in adipose tissue but not in skeletal muscle. Further characterisation of genes related to glucose metabolism in cats will provide additional insights into insulin-signalling mechanisms in this species.  相似文献

The present study used a proteomic data set obtained from a 2 × 2 × 2 factorial experiment on longissimus muscle of 24 pigs to identify single or couples of proteins allowing correct classification of pigs according to gender, sire breed, or rearing environment. An actin isoform, a myosin light chain 2 isoform and cytochrome Bc1 allowed each, correct classification of all pigs according to gender. Peroxiredoxin 6 allowed correct classification of 23 pigs according to indoor or outdoor rearing environment, but only if gender was also taken into account. Heat shock protein 73 allowed correct classification of 21 pigs according to sire breed, Duroc or Large White. Results show that proteins may be used as biomarkers for traceability or genetic research. Relationships between treatment factors and intracellular regulatory mechanisms associated with these proteins are discussed.  相似文献

A laboratory procedure for the analysis of the oxygen-18/oxygen-16 isotope ratios of ethanol derived from sugars and fruit juices by pyrolysis-isotope ratio mass spectrometry (IRMS) has been applied to the study of isotopic fractionation induced by the isotope effects of fermentation and distillation. For both processes, an experimental model has been established to describe and explain the observed fractionation phenomena. It is shown that reproducible results can be obtained when appropriate analytical conditions are used. Moreover, the ability of ethanol to act as a reliable indicator of the (18)O/(16)O ratio of sugars in orange juice (and therefore to be used as an internal reference for detecting water addition) is demonstrated both in theory and in practice.  相似文献

Soil management systems may negatively affect the quality of the soil. Policymakers and farmers need scientific information to make appropriate land management decisions. Conventional (CT) and zero tillage (ZT) are two common soil management systems. Comparative field studies under controlled conditions are required to determine the impact of these systems on soil quality and yields. The research presented studied plant and soil physical and chemical characteristics as affected by different agricultural management practices, i.e. ZT and CT, cropped with continuous wheat or maize in monoculture (M) or in a yearly rotation (R) of these two crops, either with residue retention (+r) or without residues retention (?r), in an experimental field in the Transvolcanic Belt of Mexico after 14 years. The dominant factors defining soil quality were organic C, total N, moisture, aggregate stability, mechanical resistance, pH and EC. The principal component combining the variables organic C, total N, aggregate stability and moisture content showed the highest correlations with final yield (R = 0.85 for wheat and 0.87 for maize).After 14 years of continuous practice, ZTM + r and ZTR + r had the best soil quality and produced the highest wheat and maize yields of average 2001–2004 (6683 and 7672 kg ha?1 and 5085 and 5667 kg ha?1, respectively). Removing the residues, i.e. treatments ZTM ? r with maize (average 2001–2004: 1388 kg ha?1) and ZTR ? r and CTR ? r with wheat (average 2001–2004: 3949 and 5121 kg ha?1), gave the lowest yields and less favourable soil physical and chemical characteristics compared to the other practices. It was found that zero tillage with residue retention is a feasible management technology for farmers producing maize and wheat in the agro-ecological zone studied, resulting in a better soil quality and higher yields than with the conventional farmer practice (maize monoculture, conventional tillage and residue removal).  相似文献

In vertebrates, chemosensitivity of nutrients occurs through the activation of taste receptors coupled with G-protein subunits, including α-transducin (Gαtran) and α-gustducin (Gαgust). This study was aimed at characterising the cells expressing Gαtran immunoreactivity throughout the mucosa of the sea bass gastrointestinal tract. Gαtran immunoreactive cells were mainly found in the stomach, and a lower number of immunopositive cells were detected in the intestine. Some Gαtran immunoreactive cells in the stomach contained Gαgust immunoreactivity. Gastric Gαtran immunoreactive cells co-expressed ghrelin, obestatin and 5-hydroxytryptamine immunoreactivity. In contrast, Gαtran immunopositive cells did not contain somatostatin, gastrin/cholecystokinin, glucagon-like peptide-1, substance P or calcitonin gene-related peptide immunoreactivity in any investigated segments of the sea bass gastrointestinal tract. Specificity of Gαtran and Gαgust antisera was determined by Western blot analysis, which identified two bands at the theoretical molecular weight of ~45 and ~40 kDa, respectively, in sea bass gut tissue as well as in positive tissue, and by immunoblocking with the respective peptide, which prevented immunostaining. The results of the present study provide a molecular and morphological basis for a role of taste-related molecules in chemosensing in the sea bass gastrointestinal tract.  相似文献
