We evaluated the humoral and cellular immune responses in pigs immunized intranasally with crude rhoptry proteins of Toxoplasma gondii plus Quil-A. The experiment used 13 mixed-breed pigs divided into the following three groups: G1 (vaccinated-challenged, n=6), which received the rhoptry vaccine (200(g/dose); G2 (adjuvant-challenged, n=4), which received PBS plus Quil-A; and G3 (unvaccinated-challenged, n=3), which was the control group. The treatments were performed intranasally at days 0, 21, and 42. Three pigs from G1 produced IgG and IgM antibody levels above the cut-off in the ELISA on the challenge day. Partial protection was observed in G1 at the chronic phase of infection when compared with G3. The preventable fractions were 41.6% and 6.5%, in G1 and G2, respectively. The results of this study suggest that rhoptry proteins plus Quil-A stimulated humoral, local, and systemic immune responses, which were able to partially protect the brain from cyst formation.  相似文献

Background: The Sysmex XT‐2000iV is a hematology analyzer that combines laser and impedance technology. Its usefulness for determining cell counts in canine and feline intracavitary effusions has not yet been studied. Objectives: The objectives of this study were to evaluate the analytical performance of the Sysmex XT‐2000iV for cell counts in effusions from dogs and cats, and to assess correlation with an impedance counter and concordance with diagnoses based on cytologic findings. Methods: Effusions (43 pleural, 23 peritoneal, 6 pericardial) were analyzed from 32 dogs and 34 cats. Total nucleated cell count (TNCC), HCT, and RBC count were determined on the Sysmex and compared with those obtained on an impedance counter (Hemat 8, SEAC). Imprecision, linearity, and limit of detection were determined for the Sysmex. An algorithm was designed using quantitative and qualitative data from the Sysmex to classify the effusions and the results were compared with diagnoses based on cytologic findings. Results: Intra‐assay and interassay coefficients of variation on the Sysmex were variable. Linearity of TNCC was ≥0.993 for dogs and cats, with the exception of effusions from cats with feline infectious peritonitis, which had delta (Δ) TNC values >3.0. In comparison with the Hemat 8, a proportional error was found for TNCC on the Sysmex. Effusion classification based on the algorithm was concordant with that obtained by cytologic examination in 43/72 (60%) samples. Discordant results usually were due to the misclassification of cells with similar morphology (such as mesothelial and carcinoma cells) in Sysmex scattergrams. Conclusion: The Sysmex XT‐2000iV provides a precise and accurate TNCC and has moderate concordance with cytologic findings for classifying canine and feline effusions. Although microscopic examination of effusions is necessary to achieve an accurate diagnosis, the Sysmex can provide preliminary information that may be helpful to cytopathologists.  相似文献

Polyunsaturated fatty acids are important membrane components that influence membrane integrity and fluidity. In the present study, the effect of oral supplementation for 60 days with essential fatty acids (omega 3, 6 and 9) and vitamin E on canine semen quality was evaluated. Sixteen dogs were selected for the experiment; eight were used as the control group and eight received the fatty acid supplemented diet for 60 days. Semen samples were taken every 15 days during the entire experimental period and were analyzed for volume (ml), motility (%), vigour (0–5), concentration (×106/ml), morphology of spermatozoa (%), plasma membrane integrity (%; using the hyposmotic swelling test) and thermoresistance (motility and vigour after 4 h at 38°C). We concluded that, daily supplementation with omega 3, omega 6 and omega 9 fatty acids, together with vitamin E, for a period of 60 days, significantly increased the semen volume of the treated group after 15 days of supplementation; the vigour and concentration of spermatozoa were superior after the first month of supplementation, while the percentage of morphologically abnormal spermatozoa decreased and the cells were protected against thermal stress.  相似文献

ObjectiveTo assess the cardiorespiratory and hypnotic-sparing effects of ketamine co-induction with target-controlled infusion of propofol in dogs.Study designProspective, randomized, blinded clinical study.AnimalsNinety healthy dogs (ASA grades I/II). Mean body mass 30.5 ± SD 8.6 kg and mean age 4.2 ± 2.6 years.MethodsAll dogs received pre-anaesthetic medication with acepromazine (0.03 mg kg?1) and morphine (0.2 mg kg?1) administered intramuscularly 30 minutes prior to induction of anaesthesia. Heart rate and respiratory rate were recorded prior to pre-medication. Animals were allocated into three different groups: Group 1 (control) received 0.9% NaCl, group 2, 0.25 mg kg?1 ketamine and group 3, 0.5 mg kg?1 ketamine, intravenously 1 minute prior to induction of anaesthesia, which was accomplished using a propofol target-controlled infusion system. The target propofol concentration was gradually increased until endotracheal intubation was possible and the target concentration at intubation was recorded. Heart rate, respiratory rate and noninvasive blood pressure were recorded immediately prior to induction, at successful intubation and at 3 and 5 minutes post-intubation. The quality of induction was graded according to the amount of muscle twitching and paddling observed. Data were analysed using a combination of chi-squared tests, Fisher's exact tests, Kruskal–Wallis, and anova with significance assumed at p< 0.05.ResultsThere were no significant differences between groups in the blood propofol targets required to achieve endotracheal intubation, nor with respect to heart rate, noninvasive blood pressure or quality of induction. Compared with the other groups, the incidence of post-induction apnoea was significantly higher in group 3, but despite this dogs in this group had higher respiratory rates overall.Conclusions and clinical relevanceUnder the conditions of this study, ketamine does not seem to be a useful agent for co-induction of anaesthesia with propofol in dogs.  相似文献

The hydroalcoholic extract of Equisetum arvense (HAE) tested at the doses of 200 and 400 mg/kg showed a significant activity on the open-field, enhanced the number of falls in the rota-rod reducing the time of permanence in the bar and increased the sleeping time (46% and 74%) in the barbiturate-induced sleeping time. In the pentylenetetrazole-seizure, it increased the first convulsion latency, diminished the severity of convulsions, reduced the percentage of animals which developed convulsion (50% and 25%) and protected animals from death. On the contrary, in the elevated plus maze, the doses 50, 100 and 150 mg/kg did not affect the evaluated parameters. Thus, HAE presented anticonvulsant and sedative effects. Phytochemical analysis detected the presence of tannins, saponins, sterols and flavonoids.  相似文献

Molecular epidemiological analysis of bat rabies viruses in Brazil   总被引:9,自引:0,他引:9  
A molecular epidemiological analysis was performed in 19 rabies viruses (RVs) isolated from haematophagous, frugivorous and insectivorous bats, in Sao Paulo, Brazil. The authors carried out RT-PCR for amplification of the RV nucleoprotein (N) gene, and determined 1,335 nucleotide sequences of N gene by direct sequencing method. Phylogenetic analysis, which was based on the N gene of Brazilian RV isolates identified presently and previously, revealed that RVs isolated from bats were genetically divided into four lineages had a tendency to depend on the host bat species. The first lineage consisted mainly of haematophagous bat (Desmodus rotundus) isolates, including frugivorous bat (Artibeus spp.) isolates. Other three lineages consisted of insectivorous bat isolates; mainly Eptesicus spp., Molossus spp. and Nyctinomops spp. isolates, respectively. These results indicate a possibility of that there are bat species-specific RV variants in Brazil.  相似文献

This study was performed to validate previous equations and to develop and evaluate new regression equations for predicting lamb carcass fabrication yields using outputs from a lamb vision system-hot carcass component (LVS-HCC) and the lamb vision system-chilled carcass LM imaging component (LVS-CCC). Lamb carcasses (n = 149) were selected after slaughter, imaged hot using the LVS-HCC, and chilled for 24 to 48 h at -3 to 1 degrees C. Chilled carcasses yield grades (YG) were assigned on-line by USDA graders and by expert USDA grading supervisors with unlimited time and access to the carcasses. Before fabrication, carcasses were ribbed between the 12th and 13th ribs and imaged using the LVS-CCC. Carcasses were fabricated into bone-in subprimal/primal cuts. Yields calculated included 1) saleable meat yield (SMY); 2) subprimal yield (SPY); and 3) fat yield (FY). On-line (whole-number) USDA YG accounted for 59, 58, and 64%; expert (whole-number) USDA YG explained 59, 59, and 65%; and expert (nearest-tenth) USDA YG accounted for 60, 60, and 67% of the observed variation in SMY, SPY, and FY, respectively. The best prediction equation developed in this trial using LVS-HCC output and hot carcass weight as independent variables explained 68, 62, and 74% of the variation in SMY, SPY, and FY, respectively. Addition of output from LVS-CCC improved predictive accuracy of the equations; the combined output equations explained 72 and 66% of the variability in SMY and SPY, respectively. Accuracy and repeatability of measurement of LM area made with the LVS-CCC also was assessed, and results suggested that use of LVS-CCC provided reasonably accurate (R2 = 0.59) and highly repeatable (repeatability = 0.98) measurements of LM area. Compared with USDA YG, use of the dual-component lamb vision system to predict cut yields of lamb carcasses improved accuracy and precision, suggesting that this system could have an application as an objective means for pricing carcasses in a value-based marketing system.  相似文献

The aim of this study was to determine whether feed supplementation with nanoencapsulated Ph2Se2 would improve the growth performance of silver catfish and would maintain or improve the muscle antioxidant system. Final mean weight (p = .031), weight gain (p = .014) and specific growth rates (p = .045) were significantly higher in supplemented fish than in the control group. Muscle reactive oxygen species (p = .0001) levels were significantly lower in supplemented fish than in the control group. Also, muscle hydrogen peroxide (p = .0001) and lipid peroxidation (p = .0001) levels were significantly lower in supplemented fish than in the control group. Muscle antioxidant capacity against peroxyl radical (p = .00001) levels were significantly higher in fish supplemented with both Ph2Se2 forms (free or nanocapsules) than in the control group, while muscle superoxide dismutase (p = .001) and catalase (p = .0001) activities were significantly higher in supplemented fish than in the control group. Finally, selenium (Se) deposition (p = .00001) was significantly higher in the muscle of fish supplemented with free and nanoencapsulated forms compared with the control; and deposition levels were significantly higher in fish supplemented with Ph2Se2‐loaded in nanocapsules compared with the free form. Based on this evidence, Ph2Se2‐loaded in nanocapsules improves silver catfish performance and antioxidant/oxidant status. The greater content of Se (1.22 µg/g muscle) in the muscle of fish fed with dietary Ph2Se2‐loaded nanocapsules indicates that it may be considered a nutraceutical for fish.  相似文献
