Serological investigation to detect foot-and-mouth disease (FMD) virus circulation in the domestic small ruminant population of India was conducted. A total of 4407 and 4035 serum samples from sheep and goats, respectively were collected at random covering majority of the states across the country during 2010–2012. These samples were analyzed for antibodies against the non-structural proteins (NSP) of FMD virus in an indirect 3AB NSP ELISA and against the structural proteins (SP) in a liquid phase blocking (LPB) ELISA. A total of 20.35% sheep and 13.60% goats were found to be positive for 3AB NSP antibodies providing a serological evidence of extensive viral activity. In LPB ELISA, only 4.54% sheep and 6.27% goats were found to have protective antibody (log10 titre of ≥1.8) against all three serotype strains in the vaccine, which correlates with “no or sparse vaccination” scenario in these species in the country. Hence, to check silent amplification and dissemination of virus in a mixed farming set up, small ruminants may be brought under the ambit of routine vaccination and surveillance programmes.  相似文献

The pharmacokinetics of albendazole were studied in sheep and goats following single and divided doseadministration at nematocidal and flukicidal dose rates. The disposition curves of the metabolites indicated increased uptake of the drug both in sheep and goats at divided dose schedules compared to single dose administration (P<0.05). The increased bioavailability of benzimidazole anthelmintics in divided dose schedules could improve their efficacy and help in extending their lives.  相似文献

In vitro addition of the drugs tetramisole (TMS) and levamisole (LMS) caused an inhibition of the specific activities of acid phosphatase and Mg(++)-dependent adenosine triphosphatase. The inhibition was non-competitive in nature. No significant inhibition was caused by TMS in the activity of glucose-6-phosphatase, but LMS inhibited the enzyme in a non-competitive manner. In India, systematic genotype replacement has been observed for serotype A foot-and-mouth disease virus. After a decade of co-circulation of genotypes VI and VII, genotype VII emerged as the single dominant genotype since 2001. To derive possible explanations for such epochal evolution dynamics, in vitro intergenotype growth competition experiments involving both co- and superinfection regimes were conducted. Coinfection of BHK-21 cells demonstrated abrupt loss in the genotype VI viral load with commensurate increase in the load of genotype VII as measured by the genotype differentiating ELISA, RT-PCR and real-time RT-PCR. The superinfection dynamics was shaped by temporal spacing of infection, where the invading genotype VII took more number of passages than coinfection to eventually overtake the resident genotype VI. It was speculated that such superior replicative fitness of genotype VII could have been a possible factor for the ultimate dominance of genotype VII in nature.  相似文献

L-(U-14C) aspartic acid, L-(U-14C) alanine and L-(U-14C) leucine uptake by Ascaridia galli was found to be a non-linear function of time and limiting substrate concentration. The uptake was rapid initially but achieved steady state thereafter, possibly owing to the saturation of transport loci. Linear transformations of substrate saturation kinetics by Lineweaver-Burk plots of L-(U-14C) aspartic acid, L-(U-14C) alanine and L-(U-14C) leucine gave Kt values of 4.76, 3.03 and 2.0 mM and Jmax of 5.0, 3.57 and 2.08 m moles/100 mg dry weight/2 min, respectively. D1-tetramisole and 1-tetramisole (levamisole) inhibited the uptake of amino acids. The uptaken amino acids were readily metabolized into different tissue fractions. D1-tetramisole and levamisole significantly inhibited the incorporation of the three amino acids into the nematode's total protein, RNA and lipid fractions in an in vitro incubation system.  相似文献

Adult Ascaridia galli, an intestinal nematode parasite of fowl, reveals a large variety of complex lipids such as phospholipids containing choline, ethanolamine, inositol, serine and glycerol. Lysophospholipid species, vinyl ether phospholipid (plasmalogen), neutral acylglycerols, cholesterol and non-esterified fatty acids are also present. Sugar-containing lipids, such as cerebrosides, sulphatides and gangliosides are abundantly present. Female parasites contain more lipids, particularly acylglycerols and phospholipids. Acylglycerols, phosphatidyl choline, phosphatidyl ethanolamine and glycolipids incorporate a large amount of radiolabelled precursor substrate in A. galli. The presence of important enzymes of lipid biosynthesis like glucose-6-phosphate dehydrogenase, malate dehydrogenase and hydroxymethyl glutaryl-CoA reductase as well as an enzyme of lipid ester hydrolysis, triacylglycerol lipase is detected in the parasite. These enzymes show subcellular distribution patterns and Michaelis-Menten kinetic characteristics comparable with that from rat liver homogenate. Studies on the uptake of labelled precursor molecules for lipid biosynthesis, glucose, acetate and palmitate show that the parasites can take up the isotopes readily in a time-dependent manner, showing substrate saturation kinetics, dependence upon Na ions, and can be inhibited by the presence of the bile salts sodium cholate and sodium deoxycholate. The substrate affinity constant (Kt) and maximum apparent velocity of glucose uptake in A. galli were found to be 9.09 mM and 26.67 mM per 100 mg tissue dry weight per min at 37 degrees C.  相似文献

14C-glucose uptake by adult Trichuris globulosa was found to be a non-linear function of time and limiting substrate concentration. The uptake was a two component process, an initial rapid burst, followed by a lower steady state. Linear transformation of the substrate saturation kinetics by Eadie plot gave a Kt value of 5 X 10(3) microM and Jmax of 5 mumol mg-1 dry weight min-1. Presence of the benzimidazole drugs, thiabendazole and fenbendazole, markedly inhibited the uptake process at concentrations in which the motility of the parasites was not affected. T. globulosa was found to possess the full complement of enzyme systems of glycolysis, Krebs cycle, phosphatases, transaminases and other NAD- and NADP-dependent enzymes. Thiabendazole and fenbendazole lowered the Vmax of most of these enzymes and affected the substrate-affinity constant (Km) in some of them. The mode of action of these drugs was discussed in the light of the data obtained.  相似文献

Sulphadimidine, amprolium, halofuginone and chloroquine phosphate were administered to buffalo calves 10 days after experimental infection with Eimeria bareillyi. Animals given sulphadimidine or amprolium remained clinically normal and shed only a few oocysts in their faeces. Halofuginone was found partially effective and chloroquine phosphate completely ineffective in preventing faecal oocyst discharge and intestinal lesions. Sulphadimidine and amprolium treated calves gained weight, but chloroquine treated calves suffered progressive weight loss similar to that of infected untreated controls. No significant alterations of haematological values were observed either in the treated calves or in the untreated controls.  相似文献
