Microbial degradation of 3-(3,4-dichlorophenyl)-1,1-dimethylurea (diuron) and mineralization of 4-chloro-2-methylphenoxyacetic acid (MCPA) were studied in soil samples taken from the ballast layers of three Swedish railway embankments. The degradation of diuron followed first-order kinetics and half-lives ranged between 122 and 365 days. The half-lives correlated strongly with microbial biomass estimated by substrate-induced respiration (SIR; R=-0.85; p<0.05) and with the amount of organic matter measured as loss on ignition (R=-0.87; p<0.05). Accumulation of the metabolites 1-(3,4-dichlorophenyl)-3-methyl urea (DCPMU) and 1-(3,4-dichlorophenyl) urea (DCPU) was observed in all samples and these were only detectably degraded in the sample with the highest SIR. Addition of ground lucerne straw to the ballast samples stimulated microbial activity and led to increased formation of metabolites, but further transformation of DCPMU and DCPU was not enhanced. Mineralization of MCPA followed growth-linked kinetics and the time for 50% mineralization was 44.5±7.1 days in samples of previously untreated ballast. In samples of ballast that had been previously treated with the herbicide formulation MCPA 750, the time for 50% mineralization was reduced to 13.7±11.3 days. The number of MCPA degraders, quantified using an MPN technique, was clearly increased but highly variable. An average yield of 0.18 cells pg−1 of MCPA was estimated from the kinetic data. The yield estimates correlated with the amount of nitrogen in the ballast, indicating that mineralization of MCPA was nitrogen-limited in the railway embankments studied. This has practical implications for weed control using herbicides on railways.  相似文献

Soil tillage has the potential to influence water flow and solute transport through the soil by cutting continuous macropores which connect the soil surface to the subsoil. Tillage also affects soil organic carbon sequestration which may lead to different sorption and degradation properties depending on the long-term tillage practices. The objective of this study was to quantify the differences in sorption, degradation and leaching of the herbicides bentazone and isoproturon between conventional tillage (CT) and reduced tillage (RT) under Swedish conditions. Three sites, Ultuna (silty clay), Säby (loam), and Lönnstorp (sandy loam moraine till), where replicate plots had been under either CT or RT for at least 9 years, were included in the study. A higher organic carbon content had developed in the top 5 cm of RT plots compared to the 10–20 cm depth and CT plots since the establishment of the experimental treatments. Adsorption and degradation were studied in laboratory experiments and solute transport was studied in undisturbed column experiments using non-reactive tracers and herbicides. The results from the column experiments were not significantly different between tillage treatments for Säby and Lönnstorp. For Ultuna, RT resulted in a more pronounced preferential tracer transport pattern and isoproturon leaching was twelve times larger compared to CT columns. This indicates that the tillage treatment had affected the macropore connectivity only at Ultuna. Freundlich adsorption coefficients for both bentazone and isoproturon were larger (though not always significantly) in the top 5 cm of RT soil compared to the 10–20 cm depth and to CT, reflecting the higher organic carbon content. The degradation rate was also generally larger (though not always significantly) in the top 5 cm of RT soil. These results show that RT has the potential to reduce pesticide leaching. However, any such reduction may be counter-balanced by enhanced preferential flow for soils where RT results in improved macropore connectivity.  相似文献

The impact of individual amino acids (AA) on gut hormone secretion and appetite regulation in pigs remains largely unknown. The aim of the present study was to determine the effect of the 20 proteinogenic AA on the release of the anorexigenic hormones cholecystokinin (CCK) and glucagon-like peptide 1 (GLP-1) in postweaning pigs. Six 25-d-old male piglets (Domestic Landrace × Large White; body weight = 6.94 ± 0.29 kg) were humanely killed for the collection of intestinal segments from the duodenum, jejunum, and ileum. Tissue samples from the three intestinal segments were used to determine which of the regions were more relevant for the analysis of gut peptides. Only the segments with the highest CCK and GLP-1 secretion and expression levels were evaluated with the 20 individual AA. Tissue segments were cut open, cleaned, and stripped of their muscle layer before identical circular samples were collected and incubated in 24-well plates for 1 h (37 °C, 5% v/v CO2). The culture broth consisted of a glucose-free KRB buffer containing no added AA (control) or with the addition of 10 mM of 1 of the 20 proteinogenic AA. Following incubation, tissues and supernatant were collected for gene expression and secretion analysis of CCK and GLP-1 levels. CCK secretion and mRNA expression were higher (P < 0.05) in duodenum when compared with proximal jejunum or ileum, whereas GLP-1/proglucagon levels were higher in ileum vs. duodenum (P < 0.05) and jejunum (P < 0.05, for GLP-1 only) in postweaning pigs. Based on these results, the effect of AA on CCK and GLP-1 secretion was studied in the duodenum and ileum, respectively. None of the AA tested stimulated both anorexigenic hormones. Of all the essential AA, Ile, Leu, Met, and Trp significantly (P < 0.05) stimulated GLP-1 from the ileum, while only Phe stimulated CCK from the duodenum. Of the nonessential AA, amide AA (Gln and Asn) caused the release of CCK, while Glu and Arg increased the release of GLP-1 from the ileum. Interpreting the results in the context of the digestion and absorption dynamics, non-bound AA are quickly absorbed and have their effect on gut peptide secretion limited to the proximal small intestine (i.e., duodenum), thus, mainly CCK. In contrast, protein-bound AA would only stimulate CCK release from the duodenum through feedback mechanisms (such as through GLP-1 secreted mainly in the ileum).  相似文献

An improved understanding of how precipitation patterns control pesticide leaching from structured soils prone to macropore flow could lead to practical mitigation strategies that would help farmers minimize losses by optimizing application timings. A sensitivity analysis of the macropore flow model MACRO was therefore carried out to examine the influence of antecedent soil water content and precipitation patterns on pesticide leaching to drainage systems and groundwater. One thousand model runs were executed (20 four-year weather data series, 50 application dates per season) for both autumn and spring applications of a hypothetical moderately sorbed and quickly degraded herbicide for one of three national scenarios for pesticide risk assessment in Sweden (Näsbygård, a loamy moraine soil in Scania, southern Sweden). Rapid and direct transport of pesticides in macropores to drainage systems and shallow groundwater was predicted to occur rather infrequently in spring (in 4 of the 20 years) and even more rarely in autumn. For autumn applications, the soil water deficit at application (SWDtot) and medium-term precipitation (30–90 days after application) were the two most sensitive variables controlling pesticide leaching. For spring applications, total leaching was most closely linked to rainfall the following winter, while short-term precipitation (5 days after application) and the antecedent soil water deficit were the two most important predictors for maximum pesticide concentrations in drainflow. The potential for reducing leaching by restricting applications to periods of low risk was investigated. The results showed that avoiding applications on wet soil in autumn could potentially reduce total pesticide losses by a factor of two to three. Similarly, the risk of acute toxicological effects in surface waters following pesticide applications in spring could be reduced by a factor of 2–3 by avoiding application when 5-day weather forecasts predict precipitation >10 mm.  相似文献

African trypanosomes cause human sleeping sickness and livestock trypanosomiasis in sub-Saharan Africa. We present the sequence and analysis of the 11 megabase-sized chromosomes of Trypanosoma brucei. The 26-megabase genome contains 9068 predicted genes, including approximately 900 pseudogenes and approximately 1700 T. brucei-specific genes. Large subtelomeric arrays contain an archive of 806 variant surface glycoprotein (VSG) genes used by the parasite to evade the mammalian immune system. Most VSG genes are pseudogenes, which may be used to generate expressed mosaic genes by ectopic recombination. Comparisons of the cytoskeleton and endocytic trafficking systems with those of humans and other eukaryotic organisms reveal major differences. A comparison of metabolic pathways encoded by the genomes of T. brucei, T. cruzi, and Leishmania major reveals the least overall metabolic capability in T. brucei and the greatest in L. major. Horizontal transfer of genes of bacterial origin has contributed to some of the metabolic differences in these parasites, and a number of novel potential drug targets have been identified.  相似文献

Whole-genome sequencing of the protozoan pathogen Trypanosoma cruzi revealed that the diploid genome contains a predicted 22,570 proteins encoded by genes, of which 12,570 represent allelic pairs. Over 50% of the genome consists of repeated sequences, such as retrotransposons and genes for large families of surface molecules, which include trans-sialidases, mucins, gp63s, and a large novel family (>1300 copies) of mucin-associated surface protein (MASP) genes. Analyses of the T. cruzi, T. brucei, and Leishmania major (Tritryp) genomes imply differences from other eukaryotes in DNA repair and initiation of replication and reflect their unusual mitochondrial DNA. Although the Tritryp lack several classes of signaling molecules, their kinomes contain a large and diverse set of protein kinases and phosphatases; their size and diversity imply previously unknown interactions and regulatory processes, which may be targets for intervention.  相似文献

Parasitic nematodes that cause elephantiasis and river blindness threaten hundreds of millions of people in the developing world. We have sequenced the approximately 90 megabase (Mb) genome of the human filarial parasite Brugia malayi and predict approximately 11,500 protein coding genes in 71 Mb of robustly assembled sequence. Comparative analysis with the free-living, model nematode Caenorhabditis elegans revealed that, despite these genes having maintained little conservation of local synteny during approximately 350 million years of evolution, they largely remain in linkage on chromosomal units. More than 100 conserved operons were identified. Analysis of the predicted proteome provides evidence for adaptations of B. malayi to niches in its human and vector hosts and insights into the molecular basis of a mutualistic relationship with its Wolbachia endosymbiont. These findings offer a foundation for rational drug design.  相似文献

This experimental study investigated differences in perceived restorativeness, mood, attention capacity and physiological reactions when visiting city and forest environments. Twenty female patients diagnosed with exhaustion disorder visited three different forest environments and one city environment in randomized order. They performed a standardized 90-min test procedure in each of these environments. Evaluation of the environments and psychological effects in mood were studied with self-administered questionnaires. Attention capacity was studied with Necker Cube Pattern Control task. Physiological responses were measured with regularly scheduled controls of heart rate and blood pressure, and a single test of heart rate recovery. Visits to the forest environments were perceived as significantly more restorative, enhancing mood and attention capacity compared to the city. This also applies to the results of heart rate and to some extent to the results of the diastolic blood pressure. The results from this experimental study support our hypothesis that short visits to forest environments enhance both psychological and physiological recovery and that visits to forest environments are likely to be beneficial when suffering from exhaustion disorder.  相似文献

Abstract

Organic dairy production will be based entirely on organically grown feeds as of 2008, and thereby a demand is created to find suitable feeds that are possible to grow locally. The aim of this experiment was to investigate whether milk yield and milk composition were maintained when cows were fed a 100% organic diet, with around 15% of dietary DM from cold-pressed organic rapeseed cake, compared to a 95% organic diet, in which a conventional protein source was used. Two experimental groups included 20 cows each and the study was carried out during two indoor periods. For cows fed the rapeseed cake diet, milk yield increased by 3.0 kg/day, but contents of milk fat and milk protein of multiparous cows decreased, resulting in similar yields of energy-corrected milk in early lactation. Milk from cows fed rapeseed cake had increased content of unsaturated fatty acids, including conjugated linoleic acid. In conclusion, it is possible to maintain milk production of dairy cows fed 100% organic feedstuffs using cold-pressed rapeseed cake as the main protein source compared to a 95% organic ration.  相似文献
