Andean Potato is a major crop for farmers in the Andes and represents an important gene pool for potato improvement. We present the population structure and genetic diversity of 88 Andigena accessions collected in Northwestern Argentina based on functional markers (25 SSR) distributed along 12 chromosomes. Polymorphic information content ranged from 0.40 to 0.87. A Bayesian approach, a Principal Coordinate Analysis and a Cluster Analysis revealed the presence of: I) a major group containing most of the Andean accessions and II) a smaller group including the out-group cv. Spunta and the sequenced genotype DM. Several group specific alleles were detected. AMOVA showed that 81% of the variability was within each group. Eleven of the SSRs analyzed are linked or within genes reported to regulate traits of nutritional and industrial interest. Additionally, the allelic variant of a photoperiod dependent tuberization regulator gene, StCDF1.2, was exclusively detected in accessions clustered in group II.  相似文献

The species Ooencyrtus telenomicida (Vassiliev) (Hymenoptera: Encyrtidae) is currently being considered as a candidate for augmentative biological control agent (BCA) against several pests, including Halyomorpha halys (Stål) (Heteroptera: Pentatomidae). Protocols for mass production of a BCA need fundamental information on its biological attributes. Here we tested a possible laboratory host for the rearing of O. telenomicida: the common bug Graphosoma lineatum L. (Heteroptera: Pentatomidae). At tested conditions, O. telenomicida biological parameters were: rm 0.154, offspring production 84.07 specimens/female, sex ratio (% females) 71%, juvenile development time about 16 days, population doubling time 4.52 days, emergence rate about 90%.  相似文献

BACKGROUND

Maize technologies expressing Bacillus thuringiensis (Bt) insecticidal proteins are widely used in Argentina to control sugarcane borer (Diatraea saccharalis Fabricius). Unexpected D. saccharalis damage was observed to Bt maize events TC1507 (expressing Cry1F) and MON 89034 × MON 88017 (expressing Cry1A.105 and Cry2Ab2) in an isolated area of San Luis Province. Diatraea saccharalis larvae were sampled from MON 89034 × MON 88017 fields in the area to generate a resistant strain (RR), which was subsequently characterized in plant and diet bioassays.

RESULTS

Survivorship of the RR strain was high on TC1507 leaf tissue, intermediate on MON 89034 × MON 88017, and low on MON 810 (expressing Cry1Ab). The RR strain had high resistance to Cry1A.105 (186.74‐fold) and no resistance to Cry2Ab2 in diet bioassays. These results indicate resistance to Cry1F and Cry1A.105 (and likely cross‐resistance between them) but not to Cry1Ab or Cry2Ab2. Resistance to MON 89034 × MON 88017 was functionally recessive. Reviews of grower records suggest that resistance initially evolved to Cry1F, conferring cross‐resistance to Cry1A.105, with low refuge compliance as the primary cause. A mitigation plan was implemented in San Luis that included technology rotation, field monitoring, and grower education on best management practices (BMPs) including refuges.

CONCLUSION

In the affected area, the resistance to Cry1F and Cry1A.105 is being managed effectively through use of MON 89034 × MON 88017 and MON 810 in combination with BMPs, and no spread of resistance to other regions has been observed. © 2017 The Authors. Prunus necrotic ringspot Ilarvirus (PNRSV) is a pollen and seed-borne ilarvirus affecting most Prunus spp. The location of the virus in infected nectarine pollen grains was investigated by molecular hybridisation, RT-PCR and in situ hybridisation. The first two approaches revealed an internal location of the virus. In situ hybridisation demonstrated the virus in the bicellular pollen grain, where it was present in the cytoplasm of the vegetative cell but not in the generative cell. This result seems to indicate that the sperm cells, formed by the mitosis of the generative cell, are not involved in virus transmission to seed. Other possible mechanisms are discussed.  相似文献

BackgroundBasal serum cortisol (BSC) ≥2 μg/dL (>55 nmol/L) has high sensitivity but low specificity for hypoadrenocorticism (HA).ObjectiveTo determine whether the urinary corticoid:creatinine ratio (UCCR) can be used to differentiate dogs with HA from healthy dogs and those with diseases mimicking HA (DMHA).AnimalsNineteen healthy dogs, 18 dogs with DMHA, and 10 dogs with HA.MethodsRetrospective study. The UCCR was determined on urine samples from healthy dogs, dogs with DMHA, and dogs with HA. The diagnostic performance of the UCCR was assessed based on receiver operating characteristics (ROC) curves, calculating the area under the ROC curve.ResultsThe UCCR was significantly lower in dogs with HA (0.65 × 10−6; range, 0.33‐1.22 × 10−6) as compared to healthy dogs (3.38 × 10−6; range, 1.11‐17.32 × 10−6) and those with DMHA (10.28 × 10−6; range, 2.46‐78.65 × 10−6) (P < .0001). There was no overlap between dogs with HA and dogs with DMHA. In contrast, 1 healthy dog had a UCCR value in the range of dogs with HA. The area under the ROC curve was 0.99. A UCCR cut‐off value of <1.4 yielded 100% sensitivity and 97.3% specificity in diagnosing HA.Conclusions and Clinical ImportanceThe UCCR seems to be a valuable and reliable screening test for HA in dogs. The greatest advantage of this test is the need for only a single urine sample.  相似文献

The determination of stomatal ozone fluxes is essential to assess the potential damage to plants due to ozone uptake. This parameter is not accessible directly with measurements, but can be deduced through algorithms using observational data. Total ozone fluxes and water vapour fluxes are generally used. Water vapour fluxes give an indication on stomatal aperture, which is the controlling factor of ozone uptake by vegetation. In this work, a series of observations made during the growing season over an onion field are used to show the equivalence of two algorithms found in the literature to derive ozone stomatal fluxes and both based on the similarity between ozone stomatal fluxes and water vapour stomatal fluxes. One of these algorithms uses the Penman-Monteith approach, where the water vapour pressure deficit is calculated using air temperatures; the second calculates, with another formulation, the water vapour deficit from the leaf temperature. The two approaches lead to the same results if applied properly, as shown in this work, both theoretically and numerically.  相似文献

With the objective of finding floral markers for the determination of the botanical origin of acacia (robinia) honey, the phytochemicals present in nectar collected from Robinia pseudacacia flowers were analyzed by high-performance liquid chromatography-tandem mass spectrometry. Eight flavonoid glycosides were detected and characterized as kaempferol combinations with rhamnose and hexose. Acacia honey produced in the same location where the nectar was collected contained nectar-derived kaempferol rhamnosides. This is the first time that flavonoid glycosides have been found as honey constituents. Differences in the stability of nectar flavonoids during honey elaboration and ripening in the hive were shown to be due to hydrolytic enzymatic activity and to oxidation probably related to hydrogen peroxide (glucose-oxidase) activity. Acacia honeys contained propolis-derived flavonoid aglycones (468-4348 microg/100 g) and hydroxycinnamic acid derivatives (281-3249 microg/100 g). In addition, nectar-derived kaempferol glycosides were detected in all of the acacia honey samples analyzed (100-800 microg/100 g). These flavonoids were not detected in any of the different honey samples analyzed previously from different floral origins other than acacia. Finding flavonoid glycosides in honey related to floral origin is particularly relevant as it considerably enlarges the number of possible suitable markers to be used for the determination of the floral origin of honeys.  相似文献
