The effect of L-carnitine supplementation during pregnancy and lactation on the reproductive performance of sows was studied in two separate trials over three reproductive cycles. Both trials were identical in design and conduct but were performed with different animals. The trials comprised of a total of 127 sows (trial 1) and 100 sows (trial 2) which were divided into control and treatment groups. All animals were fed individually and received basic feed mixtures with low native carnitine concentrations. The rations of the sows in the treated group were supplemented with 125 mg L-carnitine per head and day during pregnancy and 250 mg L-carnitine per head and day during lactation. The animals of the control group received identical feed mixtures in identical amounts, but without the L-carnitine supplement. In the first trial, 212 litters were produced and evaluated for number and body weight of the animals, in the second trial, 173 litters were produced. L-carnitine supplementation significantly increased body weight gains of the sows between day 1 and day 85 of weaning. The number of born piglets, stillborn piglets and piglets fit for rearing was not influenced by dietary L-carnitine supplementation. However, L-carnitine supplementation significantly increased the weights of piglets and litters at birth, weight gains of litters during suckling and weights of litters at weaning. These effects of L-carnitine were seen in both trials; they were independent of the age of the sows and remained over three reproductive cycles in which the sows where continuously treated with L-carnitine. Overall, the study shows that dietary supplementation with L-carnitine during pregnancy and lactation improves the reproductive performance of sows over several reproductive cycles, independent of the age of the sows.  相似文献

Three experiments were conducted to determine the requirement of tryptophan for piglets. In the first two experiments dose-response relationships between the dietary tryptophan concentration and performance criteria of piglets were examined in order to determine the dietary tryptophan concentration required for maximum performance. In the first experiment dietary tryptophan levels ranged from 1.09 to 2.32 g per kg, in the second experiment they ranged from 1.70 to 2.60 g per kg. In both experiments a close correlation was observed between the dietary tryptophan concentration and piglet performance criteria (feed intake, daily gains, feed efficiency). In the first experiment, regression analysis using a non-linear model revealed that the optima, defined as 95% of the asymptotic response in the model used, for feed intake and daily gains were achieved at tryptophan levels in excess of the highest concentration of 2.32 g per kg feed. The performance level of the piglets in this experiment was generally very low, however. In the second experiment feed intake, the optimum tryptophan concentration, defined as 95% of the asymptotic response, for daily gains and feed efficiency were achieved within a small range between 2.07 and 2.14 g tryptophan per kg feed, corresponding to 1.84 to 1.91 g precaecal digestible tryptophan per kg feed or 0.153 to 0.159 g tryptophan per MJ ME. These results suggest that the tryptophan concentration for maximum performance of piglets is probably higher than has been implied in numerous studies to date. The third experiment was set up to investigate the effect of the reduced feed intake and the effect of an inadequate tryptophan supply per se on the animals' growth. Here a two-factorial experimental design was used also by varying the energy density of the diet (13 vs. 14 MJ ME per kg feed). In addition to tryptophan deficient groups (1.5 g tryptophan per kg feed), this experiment contained conventional control groups (2.6 g tryptophan per kg feed, ad libitum feeding) and pair-fed control groups (2.6 g tryptophan per kg feed, feed intake identical to that of the tryptophan deficient group). The energy density had no significant effect on the animals' performance and increasing the energy density of the diet did not significantly affect feed and energy intake or daily gains of the tryptophan deficient animals. Feed intake, daily gains and feed efficiency of the tryptophan deficient groups were markedly poorer (by 30, 35 and 10%) than in the ad libitum control groups. When compared with the pair-fed control groups, on the other hand, the performance of the tryptophan deficient groups in terms of daily gains, feed conversion and energy efficiency was only slightly and not significant lower by 1, 4 and 3%, respectively. These results demonstrate conclusively that the growth depression in tryptophan deficiency is almost entirely due to the marked reduction in feed intake rather than to a direct limitation of protein accretion caused by an inadequate tryptophan supply.  相似文献

The objective of the study was to test the feasibility of coexistence between genetically modified (GM) and non‐GM maize under real‐life agronomical conditions. GM hybrid maize with the event MON810 (Bt maize) was drilled at 30 sites in fields surrounded by near isogenic conventional maize, although only 27 sites could be finally evaluated. Field sizes of Bt maize varied between 0.3 and 23 ha, and the flowering period of the Bt and conventional maize was synchronous. At some sites, different planting dates of GM and non‐GM maize or an earlier ripening conventional maize were tested in additional strips to obtain altered flowering and thereby reduce cross‐pollination. The overlapping of flowering periods was successfully avoided only at two sites where non‐GM maize was planted 25 or 28 days later. During harvest, samples were taken from the conventional maize in strips at distances of 0–10, 20–30, and 50–60 m to the Bt maize fields to assess the GM DNA content as a function of distance. Sampled materials included chaffed plant material intended for silage (18 sites), grains (eight sites), or crushed husks and cobs (one site). Wind effects were taken into account by sampling in all four compass directions. Quantitative PCR was used to detect the event specific MON810 DNA sequence in sampled materials. The analysis was conducted by two certified independent diagnostic testing companies selected in a pre‐test. Taking averages over all compass directions and the two laboratories no samples collected beyond 10 m had levels of GM above the threshold of 0.9 %. In conclusion, the data indicate that coexistence of GM and conventional maize is possible under real‐life large‐scale agronomical conditions. Levels of GM DNA in harvested grain resulting from outcrossing can be managed to levels below 0.9 % by simply planting 20 m of conventional maize as a pollen barrier between adjacent fields.  相似文献

Wheat cultivars carrying the 1BL.1RStranslocation were crossed with newly synthesised octoploid triticale lines involving four rye genotypes having ο-secalin banding patterns different from each other and from that of the 1BL.1RS translocation. Homologous recombination was expected between the short arm of the 1R chromosomes of the rye genotypes and the 1RS arm of the 1BL.1RSwheat/rye translocation. Seven sequence-specific PCR-based markers:Xiag95, RMS13, Bmac0213, GPI, Xpsr960, 5Sand SCM9, and ο-secalinproteins were used to detect recombination events in the BC1F2 generation. Segregation analysis demonstrated that a barley SSR marker (Bmac0213) locus was present on the 1RS chromosome arm. Of 834plants tested in four different BC1F2 populations, 246individuals were found to carry recombined1BL.1RS translocation chromosomes. Genetic linkage analysis was performed on the eight markers in the four different mapping populations. The physical positions of the markers are discussed. A long‐term field experiment, conducted since 1962 in Gumpenstein (Austria) on a Dystric Cambisol, was used for the present investigation. We combined a physical fractionation procedure with the determination of natural abundance of 13C and FT‐IR spectroscopy to study the influence of fertilizer amendments (organic manure and mineral fertilizers) and management practices (fallow vs. cropped) on changes in organic carbon (OC) associated with different particle‐size fractions. The OC content in bulk soil decreased or was not affected by slurry+straw, PK, and NPK treatments in both fallow and cropped plots after 28 and 38 yr of treatment. However, OC in plots receiving organic manures increased depending on the quality of the organic manures applied. The ranking among the different treatments under both fallow and cropped plots was: animal manure (liquid) > animal manure (solid) > cattle slurry = slurry+straw = PK = NPK. Results showed that the two types of management practices, fallow (non‐tilled) vs. cropped (tilled) had effects on OC concentrations. Comparing the OC contribution of particle‐size fractions to the total OC amount revealed the following ranking: silt > clay > fine sand > coarse sand except in the plots receiving solid or liquid animal manure. Size fractions within treatments showed larger variations of 13C abundances than bulk samples between treatments. The natural abundances of 13C increased especially in cropped (and tilled) plots. It was shown by cluster analysis that FT‐IR spectra differentiated between the different treatments originating from different land management practices. The present study revealed that below‐ground C deposition by agricultural plants can hardly compensate the C losses due to tillage.  相似文献

It has been shown that some dietary plant proteins beneficially influence lipid metabolism in animals. The effect of pea protein in this respect however has not yet been investigated. Therefore, we studied the effect of purified pea protein on the lipid metabolism in rats. Twenty-four rats received diets with either 200 g/kg of casein or purified pea protein for 16 days. Concentrations of triacylglycerols in liver, plasma and lipoproteins did not differ between both groups of rats. However, rats fed the pea protein diet had a lower concentration of total cholesterol in the liver and the very low density lipoproteins (VLDL) fraction than rats fed the casein diet (p < 0.05); cholesterol concentration in plasma, low density lipoproteins (LDL) and high density lipoproteins (HDL) did not differ between both groups. Rats fed pea protein moreover had an increased mRNA concentration of cholesterol-7α-hydroxylase in the liver and an increased amount of bile acids excreted via faeces compared with rats fed casein (p < 0.05). Concomitantly, mRNA concentrations of sterol regulatory element-binding protein (SREBP)-2 and its target genes 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase and LDL receptor in the liver were increased in rats fed pea protein (p < 0.05). The data of this study suggests that pea protein stimulates formation and excretion of bile acids, which leads to a reduced hepatic cholesterol concentration and a reduced secretion of cholesterol via VLDL. An increased gene expression of SREBP-2 and its target genes HMG-CoA reductase and LDL receptor may be a means to compensate for the increased loss of cholesterol for bile acid synthesis.  相似文献

Investigations by Robinson [Robinson, D.L., 2007. Days to calving in artificially inseminated cattle: comparison of potential traits. Livestock Science 110, 174–180] concluded that the most useful trait for assessing fertility of artificially inseminated (AI) beef cows is AI days to calving (AIDC), a trait that mimics days to calving for naturally mated cows. Various fixed and random effects were fitted to AIDC to determine the best way of modelling lactation status of the cow, the effect of service sire, using smaller contemporary groups and lowering the penalty value for non-calvers. Fitting the time interval between calving and the start of mating either as a 10-level factor or a cubic spline function explained considerably more variation than fitting the standard 2-level factor (wet or dry). Estimated permanent environmental effects of the cow were considerably reduced. This suggests that, if a cow calves late in the season (less than 60 days before she is inseminated), her fertility is reduced. Models should therefore account for this effect. If fitted, service sire explained 1.6% of phenotypic variation, compared to a much larger sire × contemporary group interaction (3% of phenotypic variation). It is therefore important to account for sire × contemporary group interactions. When the fertility of service sires is not being evaluated, service sires could be incorporated into the definition of contemporary groups. Ideally, breeders should be encouraged to formally record contemporary (or mating) groups. Reducing the size of contemporary groups (inferred from the data) by limiting the time interval between first and last inseminations from 120 to 60 days had only a marginal effect as did reducing the penalty for non-calvers from 21 to 10 days.  相似文献

Fibroblast growth factor 21 (FGF21) has been identified as a novel hormonal factor involved in the regulation of metabolic adaptations during energy deprivation. The present study aimed to investigate the expression of the FGF21 gene in the liver of dairy cows during the transition from pregnancy to lactation. Therefore, the relative mRNA abundance of FGF21 in liver biopsy samples of 20 dairy cows in late pregnancy (3 weeks pre‐partum) and early lactation (1, 5, 14 weeks post‐partum) was determined. It was observed that hepatic mRNA abundance of FGF21 at 1 week post‐partum was dramatically increased (110‐fold) compared to 3 weeks pre‐partum (p < 0.001). With progress of lactation, mRNA concentration of FGF21 was declining; nevertheless, mRNA abundance at 5 and 14 weeks post‐partum remained 25‐ and 10‐fold increased compared to 3 weeks pre‐partum (p < 0.001). Using a gene array technique, it was found that many genes involved in fatty acid oxidation, gluconeogenesis and ketogenesis were up‐regulated during early lactation compared to late pregnancy. Moreover, there were positive linear correlations between hepatic mRNA concentration of FGF21 and mRNA concentrations of genes involved in ketogenesis as well as carnitine synthesis and carnitine uptake at various time‐points during lactation, indicating that FGF21 could play a role in ketogenesis and carnitine metabolism in the liver of dairy cows (p < 0.05). In overall, the present study shows that expression of the FGF21 gene is strongly up‐regulated during the transition period. It is assumed that the up‐regulation of FGF21 might play an important role in the adaptation of liver metabolism during early lactation in dairy cows such as in other species.  相似文献

This study was performed to assess the effects of potato protein and fish protein on concentrations of lipids in plasma and lipoproteins and the expression of genes involved in lipid metabolism in pigs used as an animal model. Therefore, 27 young male pigs with an average body weight of 22 kg were fed diets supplemented with protein extracted from potatoes (containing 849 g protein/kg dry matter), Alaska Pollack fillet as a source of fish protein (containing 926 g crude protein/kg dry matter) or casein which was used as control, for 3 weeks. Diets were formulated to supply identical amounts of each protein to the pigs by the three protein sources, namely 116 g/day in first week and 150 g/day in the second and third week. Pigs fed potato protein had lower concentrations of cholesterol in plasma and LDL than pigs fed casein (p < 0.05); no effect was observed on concentrations of HDL cholesterol and triglycerides. Pigs fed fish protein had lower cholesterol concentrations in plasma, LDL and HDL, and lower triglyceride concentrations in triglyceride-rich lipoproteins than pigs fed casein (p < 0.05). mRNA concentrations of genes involved in bile acid synthesis and cholesterol uptake were higher in pigs fed fish protein than in pigs fed casein (p < 0.05); no effect on these genes was observed in pigs fed potato protein. Expression of genes involved in lipogenesis and fatty acid oxidation was not altered by fish protein. In conclusion, this study shows that fish protein and potato protein lower plasma cholesterol concentrations in pigs. The hypocholesterolaemic effect of fish protein might be in part caused by a stimulation of bile acid synthesis; the reason for the hypocholesterolaemic effect of potato protein requires further elucidation.  相似文献

Bioactive peptides with a variety of effects have been described from several nutritive proteins. They exhibit antimicrobial, blood-pressure lowering, antithrombotic, immunomodulatory, and cholesterol-modulating effects. In this study, we have examined whether peptides derived from food proteins might influence bile acid synthesis. A reporter gene cell line that carries a cholesterol 7alpha-hydroxylase promoter fragment fused to firefly luciferase ( cyp7a-luc) was used to screen for nutritive peptides affecting cyp7a expression, the enzyme catalyzing the rate-limiting step in bile acid synthesis. Proteolytic hydrolysates were prepared from soy protein and bovine casein with pepsin, trypsin, chymotrypsin, and elastase and size fractionated using ultrafiltration. Several bioactive hydrolysates could be identified that inhibited luciferase expression. Also, an activation of kinase (AKT, ERK, p38-MAPK) signaling could be observed. Selected hydrolysates were further fractionated by reversed-phase HPLC. Bioactive HPLC-fractions were obtained from casein but not from soy hydrolysates; however, activity could not be recovered in single peak fractions. Peptides in such fractions were identified by mass spectrometry. Five selected peptides from alpha S1-casein present in active fractions were synthesized, but none of these showed activity in the cyp7a-luc screening system. However, two of them activated MAP-kinase signaling similar to the hydrolysates, which suggests, that these peptides are involved in cyp7a regulation by the casein hydrolysates.  相似文献
