Oak leaves have a high concentration of ellagitannins. These phytochemicals can be beneficial or poisonous to animals. Beef cattle are often intoxicated by oak leaf consumption, particularly after suffering feed restriction. The severity of the poisoning has recently been associated with the ruminal microbiota, as different bacterial populations were found in animals that tolerated oak leaves and in those that showed clinical and pathological signs of toxicity. Intoxication has previously been linked to the production of phenolic metabolites, particularly catechol, phloroglucinol, and resorcinol. This suggested that the microbial metabolism of ellagitannins could also be associated with its tolerance or intoxication in different animals. Therefore, it is essential to understand the metabolism of ellagitannins in cattle. Here we show that ellagitannins are metabolized in the cattle rumen to urolithins. Different urolithins were detected in ruminal fluid, feces, urine, and plasma. Oak leaf ellagitannins declined as they were converted to urolithins, mainly isourolithin A and urolithin B, by the ruminal and fecal microbiota. Urolithin aglycons were observed in rumen and feces, and glucuronide and sulfate derivatives were detected in plasma and urine. Sulfate derivatives were the main metabolites detected in plasma, while glucuronide derivatives were the main ones in urine. The main urolithins produced in cattle were isourolithin A and urolithin B. This is a relevant difference from the monogastric mammals studied previously in which urolithin A was the main metabolite produced. Low molecular weight phenolics of the benzoic, phenylacetic, and phenylpropionic groups and metabolites such as catechol, resorcinol, and related compounds were also detected. There was a large variability in the kinetics of production of these metabolites in individual animals, although they produced similar metabolites in all cases. This large variability could be associated with the large variability in the rumen and intestine microbiota that has previously been observed. Further studies are needed to demonstrate if the efficiency in the metabolism of ellagitannins by the microbiota could explain the differences observed in susceptibility to intoxication by the different animals.  相似文献

Three different solventless sample preparation techniques based on microextraction, membrane extraction, and headspace extraction have been developed and optimized for determination of trihalomethanes in drinking water by gas chromatography electron capture detector and mass spectrometry detection. The techniques employed were headspace (HS) solid-phase microextraction, hollow fiber liquid-phase microextraction (HFLPME) and HS extraction. All techniques used were optimized with different experimental designs in order to select the most relevant variables which significantly affect the different processes. The different analytical figures of merit such as limit of detection (LOD), limit of quantification, reproducibility, accuracy, and linear dynamic range were obtained. The new HFLPME method applied used a hollow fiber membrane of polypropylene and the optimized variables were extraction time, extraction temperature, and salting-out effect. The software MODDE 6.0 was used and its design was one central composite on face with a total of 17 runs. The best conditions for the HFLPME were 20 min, 40°C, and 10% NaCl, respectively. The LODs ranged from 0.018 μg·L−1 (for CHClBr2) to 0.049 μg·L−1 (for CHBr3), being this technique the most sensitive one among those studied. Finally, after having optimized the sample preparation techniques and chromatographic conditions, several water samples were taken in two different water treatment plants in Spain (Zaragoza) and Colombia (Viterbo, Caldas). In our search for quorum-sensing (QS) disrupting molecules, 75 fungal isolates were recovered from reef organisms (endophytes), saline lakes and mangrove rhizosphere. Their QS inhibitory activity was evaluated in Chromobacterium violaceum CVO26. Four strains of endophytic fungi stood out for their potent activity at concentrations from 500 to 50 μg mL−1. The molecular characterization, based on the internal transcribed spacer (ITS) region sequences (ITS1, 5.8S and ITS2) between the rRNA of 18S and 28S, identified these strains as belonging to four genera: Sarocladium (LAEE06), Fusarium (LAEE13), Epicoccum (LAEE14), and Khuskia (LAEE21). Interestingly, three came from coral species and two of them came from the same organism, the coral Diploria strigosa. Metabolic profiles obtained by Liquid Chromatography-High Resolution Mass Spectrometry (LC-HRMS) suggest that a combination of fungal secondary metabolites and fatty acids could be the responsible for the observed activities. The LC-HRMS analysis also revealed the presence of potentially new secondary metabolites. This is, to the best of our knowledge, the first report of QS inhibition by marine endophytic fungi.  相似文献

Reasons for performing study: No studies have been reported on the effects of enoximone in anaesthetised colic horses. Objective: To examine whether enoximone improves cardiovascular function and reduces dobutamine requirement in anaesthetised colic horses. Methods: Forty‐eight mature colic horses were enrolled in this prospective, randomised clinical trial. After sedation (xylazine 0.7 mg/kg bwt) and induction (midazolam 0.06 mg/kg bwt, ketamine 2.2 mg/kg bwt), anaesthesia was maintained with isoflurane in oxygen and a lidocaine constant rate infusion (1.5 mg/kg bwt, 2 mg/kg/h). Horses were ventilated (PaCO2<8.00 kPa). If hypotension occurred, dobutamine and/or colloids were administered. Ten minutes after skin incision, horses randomly received an i.v. bolus of enoximone (0.5 mg/kg bwt) or saline. Monitoring included respiratory and arterial blood gases, heart rate (HR), arterial pressure and cardiac index (CI). Systemic vascular resistance (SVR), stroke index (SI) and oxygen delivery index (DO2I) were calculated. For each variable, changes between baseline and T10 within each treatment group and/or colic type (small intestines, large intestines or mixed) were analysed and compared between treatments in a fixed effects model. Differences between treatments until T30 were investigated using a mixed model (α= 0.05). Results: Ten minutes after enoximone treatment, CI (P = 0.0010), HR (P = 0.0033) and DO2I (P = 0.0007) were higher and SVR lower (P = 0.0043) than at baseline. The changes in CI, HR and SVR were significantly different from those after saline treatment. During the first 30 min after enoximone treatment, DO2I (P = 0.0224) and HR (P = 0.0003) were higher than after saline administration. Because the difference in HR between treatments was much clearer in large intestine colic cases, an interaction was detected between treatment and colic type in both analyses (P = 0.0076 and 0.0038, respectively). Conclusions: Enoximone produced significant, but short lasting, cardiovascular effects in colic horses. Potential relevance: Enoximone's cardiovascular effects in colic horses were of shorter duration than in healthy ponies.  相似文献

Cystic echinococcosis is the most prevalent zoonosis in Tierra del Fuego province, Argentina, with important economic, productive and public health consequences. The present work was performed to determine the ovine prevalence in Tierra del Fuego, Argentina, as well as to evaluate the quality of diagnostic systems in slaughterhouses. Moreover, genetic analyses to characterize the strain of Echinococcus granulosus involved in the region were done. The first actions to perform a diagnosis of the epidemiological situation of hydatidosis in Tierra del Fuego were done between 1976 and 1977. A canine prevalence of 80% and an ovine prevalence of 55% results were obtained. Since 1979 the control program of Hydatidosis of Tierra del Fuego was implemented. It was based on semiannual canine anthelmintic treatment with praziquantel at dose of 5mg/kg, and complemented with sanitary education and canine and ovine epidemiological surveillance. During May 1997-January 1999: 5,916 sheep coming from 20 farms of the programmatic area were evaluated. In the lamb category, hydatid cysts were not found. In the adults category, 62 infected animals were found (3.2%). The ovine prevalence was 1.1% and there was 100% of coincidence between diagnosis in the slaughterhouse, re-inspection in the laboratory and histopathological study. The marked decrease in the prevalence observed for sheep infection evidenced a destabilization of the biological cycle of the parasite. This could be explained by the application of a control program with uninterrupted systematic actions. Polymerase chain reaction-ribosomal ITS-1 DNA (rDNA) restriction fragment length polymorphism (PCR-RFLP) analysis and partial sequencing of the mitochondrial cytochrome c oxidase subunit 1 (CO1) gene were used to characterize E. granulosus isolates collected from different regions of Tierra del Fuego to determine which genotypes occurred in this region. The results revealed the presence of the G1 genotype (sheep-dog strain). This is the first time that a molecular analysis was performed for the E. granulosus isolates from Tierra del Fuego.  相似文献

We analyze the impact of grazing on dung beetle diversity at the Barranca de Metztitlán Biosphere Reserve, a xeric ecosystem in central Mexico with a long history of use by humans. We compared the community structure, as well as the alpha and beta diversity between two cover conditions (open and closed vegetation) that represent the impact of grazing within a habitat, and between habitat types (submountainous and crassicaule scrublands). From 576 samples we collected 75,605 dung beetles belonging to 20 taxa. While mean species richness and diversity were different between habitat types, cumulative species richness was not. The effects of grazing on vegetation structure influenced the cumulative species richness and diversity of dung beetles in the submountainous scrubland, where grazing has created land mosaics of a grassland matrix with scrubland patches. This was not the case in the crassicaule scrubland where the impact of grazing is not as evident. Beta diversity significantly responds to the effects of grazing on habitat conditions. We discuss the ecological factors that may promote these responses by landscape diversity components. We also identify the species that could act as useful indicators to monitor the effect of land management on biodiversity. Our results indicate cattle farming maintains a diversified land mosaic, and these areas support more diverse dung beetle ensembles than homogeneous areas of closed, shrubby vegetation cover. Thus, controlled grazing activity could certainly favour the conservation of dung beetle biodiversity and improve ecosystem functioning by maintaining dung decomposition rates.  相似文献

The stir bar sorptive extraction (SBSE) technique is used to determine volatile constituents in Monastrell grapes. The method proposed involves a maceration step lasting 2 h and a sorption time of 6 h. The coefficients of variation obtained were <4% in the case of the volatile compounds and <2% for the internal standard. The behavior of the volatiles during grape ripening was studied, and 34 compounds were determined, among which geranyl acetone, geranyl butyrate, farnesol, and a bicycloterpene, tentatively identified as 6-methylene[3.1.0]byciclohexane, were recorded for the first time in this variety. The highest terpene and norisoprenoid contents together with the lowest values of C6 compounds were observed during the fifth week of ripening, meaning that this would be the optimal time for harvesting in terms of volatile compounds.  相似文献

A quantitative laboratory assessment of the different C and N forms in soil humus fractions was carried out by incubation of a mineral substrate after the addition of (15)N-labeled compost. The experimental design included (i) preparation of the (15)N-labeled organic matter (city refuse compost, 640 g kg(-1) wheat straw and K(15)NO(3) composted for 80 days), (ii) a further 80 day incubation of a mixture of the labeled compost with a mineral soil (32 g kg(-1)), (iii) measurement of stable isotope ratios, and (iv) isolation and structural comparison by (13)C and (15)N cross-polarization, magic-angle spinning nuclear magnetic resonance (NMR) of different organic fractions, i.e., soluble, colloidal (humic and fulvic type), and particulate (free organic matter and humin), from both the compost and the compost-treated soil. The results showed that the amide forms dominated in all of the newly formed N compounds, but an increased amount of alkali insoluble organic fractions was observed after incubation of the soil. The analysis of the insoluble, particulate fractions shows that nonextractable amides constitute the major pool of newly formed N compounds. The particulate soil fraction isolated by flotation in CHBr(3)-MeOH contained 16.8% of the total soil N and 26% of the (15)N. The (13)C NMR spectra showed that the fulvic acid-like fraction (7.6% of the soil N, 8.8% of (15)N) consisted almost completely of a C=O-containing carbohydrate material, whereas the humic acid-like fraction (20.3% of the total soil N, 8.6% of (15)N) resembled an oxidized lignoproteic fraction containing the most significant aromatic domain. The water soluble fraction was, in both soil and compost, the one with the highest isotopic abundance of (15)N (96%), but the (15)N NMR spectrum revealed minor amounts of soluble mineral N in this fraction and the remainder consisting of amide compounds.  相似文献

This study was performed to identify the environmental factors that control soil organic carbon (SOC) accumulation in the Basque Country (Northern Spain) and to evaluate the impact of land use change on SOC budget in this territory. A total of 30 samples of the organomineral horizon were taken under representative vegetation types including native forest, coniferous plantations, scrub, pasture and cultures. The edaphic variables measured were organic C content, pH, total N, available P, exchangeable H+ and Al3+, cation exchange capacity (CEC), texture, and carbonates. The substitution of the original forest with a pasture led to a significant increase in organic C content of the topsoil in all cases studied. Temperature was the main climatic factor affecting the organic C levels in soil, being the two variables negatively correlated. The CEC and texture were other key factors controlling the organic C content, which increased with CEC and decreased with total sand. Average organic C content of the organomineral horizon did not differ significantly (P <0.05) with base content of original substrate nor climate type, but it was more sensitive to climate change in the Atlantic soils than in the Mediterranean ones, where edaphic variables such as CEC and texture were important in regulating C sequestration.  相似文献
