To learn more about lignin formation in response to wounding in trees, we adopted two complementary approaches: (1) microscopic and histochemical studies of the wound response in 3.5‐month‐old Eucalyptus gunnii plantlets and (2) biochemical investigations of hydroxycinnamyl alcohol dehydrogenase activities in wounded 6‐year‐old, field‐grown E. gunnii trees. The first approach revealed that a barrier zone was formed in response to wounding in both ground tissues (cortex barrier and pith reaction zone) and vascular tissues. The barrier zone was barely detectable after 24 h but well‐developed 7 days after wounding. Microscopic analyses indicated that the barrier zone was formed by the reinforcement of cell walls with ‘lignin‐like material’ in both ground tissues and vascular tissue, and that, in addition, the lumen of certain xylem cells (vessels and fibres) were blocked by the deposition of polymeric phenolic material. Histochemical characterization revealed that the lignin‐like material (‘defence lignin’) deposited in ground tissue cell walls and xylem cell blockages was poor in syringyl (S‐type) lignin units and therefore differed from the usual mixed guaiacyl–syringyl (G–S) lignin unit composition of E. gunnii developmental lignin. In contrast, S‐type lignin appeared to be deposited in the cell walls of immature developing secondary xylem cells at a stage when the cell walls of comparable cells from unwounded control stems contained lignin poor in syringyl units. The second approach indicated that two different types of cinnamyl alcohol dehydrogenase activity are induced, and apparently regulated differentially, in response to wounding in E. gunnii trees. Coniferyl alcohol dehydrogenase activity was induced immediately and continued to increase throughout the first 15 days of the 17‐day experimental period, while sinapyl alcohol dehydrogenase activity was first detected at 8 days after wounding and continued to increase throughout the experimental period. The biological roles of the two alcohol dehydrogenase activities are discussed in relation to the formation of defence lignin versus developmental lignin in trees. 相似文献

Samples of tissue from the central nervous system (cns), the lymphoreticular system (lrs) and the rectal mucosa of a large number of scrapie-exposed sheep, with and without signs of clinical disease, were examined immunohistochemically for evidence of disease-associated prion protein (PrP(d)). The rectal mucosa has received almost no attention so far in scrapie diagnosis, despite its abundant rectoanal mucosa-associated lymphoid tissue, and its accessibility. The scrapie-confirmed cases included 244 with clinical disease, of which 237 (97.1 per cent) were positive in the rectal mucosa, and 121 apparently healthy sheep, of which 104 (86 per cent) were positive in the rectal mucosa. PrP(d) was detected in 86.4 to 91.5 per cent of the other lrs tissues of the healthy sheep examined and in 77.7 per cent of their cns tissues. The stage of infection, therefore, affected the probability of a positive result in the rectal mucosa, whereas the breed, PrP genotype, age and sex had little or no independent effect. Accumulations of PrP(d) were observed in the rectal mucosa and other lrs tissues of vrq/arr sheep with preclinical and clinical scrapie, albeit with a lower frequency and magnitude than in sheep of other PrP genotypes. Western immunoblotting analyses of samples of rectal mucosa gave the characteristic PrP glycoprofile, with a sensitivity similar to that of immunohistochemistry. 相似文献

The effect of prebreeding short-term protein supplementation level (PL) and body condition (BC) on fertility rate (FERT,%),
uterine pH and embryonic mortality (EMORT,%) in sheep, was evaluated. Multiparous Rambouillet ewes at low BC (LC; n = 6, 62.7 ± 1.7 kg) or high BC (HC; n = 6, 71.9 ± 1.7 kg), received. within BC, one of two levels of ruminally undegradable protein: low (LP, 14 g/ewe per day)
and high (HP, 30 g/ewe per day). Once the animals were euthanized, corpus luteum number (CLN), as an indicator of ovulation
rate, was registered and uterine horns were irrigated to recover embryonic tissue plus associated membranes as well as to
measure uterine pH (UpH). While EMORT-1 considered a nonadjusted relationship between the number of embryos and CLN, EMORT-2
considered an analysis of covariance using CLN as the covariate. The HP-supplemented ewes had the lowest FERT (p=0.06; 100% vs 50%) and the highest EMORT (EMORT-1, 16.6% vs 53.8%, p = 0.08; EMORT-2, 52.0% vs 14.5%, p = 0.07) when compared to the LP-supplemented ewes. Neither BC nor PL affected CLN, CL weight or P4 release (p > 0.10). While the lowest UpH (p = 0.04) was observed in the HP-supplemented ewes, this group also showed the lowest fertility and the highest embryonic mortality. 相似文献

The aim of this research was to evaluate the efficacy of zinc gluconate associated with dimethyl sulphoxide (DMSO) for chemical neutering in canine males. Fifteen sexually mature male dogs were divided in two groups, named control and treated. An injection was administered to both testicles, at a concentration of 26.2 mg zinc gluconate per ml and 0.5% DMSO in the treated group (11 dogs). The control group was given injections of saline solution (four dogs). Clinical examination and blood collection for a haemogram were done both before and after drug injection. There were 12 spermograms performed to analyse sperm motility, sperm vigour, ejaculate volume, testicle size, pathology and sperm concentrations. Libido was also measured. An ultrasound examination and histopathology were performed at the end of the experiment. Dogs’ libido after chemical injection was reduced by over 50%. The spermogram analysis showed final mean results of 14.54% for sperm motility, 0.72 of sperm vigour and 37 150 per million spermatozoa per millilitre, values considered below the necessary levels at which fertilization can occur. Ultrasound and histopathology analyses of testicles for the treated group revealed more intense injuries when compared with the control group, with compromised testicular parenchyma and a decrease of germ cell number leading to total atrophy, indicating that the treatment reduced the fertilizing potential of male dogs, promoting a possible subfertile status. 相似文献

This work presents data on a suite of diffuse pollutants, monitored in a stream draining an intensively managed grassland on a 30 min time step during a period of intense rainfall to better understand their sources and pathways. Nitrite (92 μg l?1), particulate phosphorus (107 μg l?1) and soluble phosphorus (74 μg l?1) exceeded environmental limits during base flow. Concentrations of nitrate and nitrite were decreased during the storm event, whereas all other pollutants generally increased and exceeded environmental limits where specified, especially when associated with a small subsidiary hydrograph on the rising limb of the main hydrograph. Total pollutants loads, when using a 60 min sampling frequency, would have led to significant over and under-estimations depending on which 60 min sample set was used. In the worst case, loads of ammonium could have been under-estimated by 35% or over estimated by 25% with errors being associated with loads on the rising limb of the hydrograph and more specifically a small subsidiary hydrograph. This subsidiary hydrograph may have occurred as a result of runoff from the farm hard standings within the catchment. Incidental transfer of pollutants associate with this runoff have masked the overall grassland pollutant response. To better understand these different source areas and pollutant dynamics, there is a need for novel tracing techniques to elucidate their relative contribution and pathways. 相似文献

OBJECTIVE: To determine causes of tracheal rupture in cats and the mechanism of injury. DESIGN: A retrospective study was conducted to identify cats with tracheal rupture. A second study was conducted to establish mechanism of injury, and a third study was conducted to determine volume of air needed to obtain an airtight seal when inflating the cuff of an endotracheal tube in a cat. ANIMALS: 16 cats with clinical signs of tracheal rupture, 10 cat cadavers, and 20 clinically normal cats that were undergoing anesthesia. PROCEDURES: Details were extracted from medical records of 16 cats with tracheal rupture (9 treated surgically and 7 treated conservatively). For the cadaver study, the trachea of each cat cadaver was intubated and observed during overinflation of the endotracheal tube cuff. For clinically normal cats, volume of air needed to obtain an airtight seal for the endotracheal tube was recorded. RESULTS: Most ruptures were associated with cats anesthetized for dental procedures. Clinical signs associated with tracheal rupture included subcutaneous emphysema, coughing, gagging, dyspnea, anorexia, and fever. Tracheoscopy was the method of choice for documenting tracheal rupture. Surgical and conservative management were successfully used, unless the injury extended to the carina. In the cadaver study, overinflation of the endotracheal tube cuff with > 6 ml of air resulted in tracheal rupture in 7 of 10 cadavers. For clinically normal cats, the volume of air (mean +/- SD) needed to obtain an airtight seal was 1.6 +/- 0.7 ml. The infectivity in tissues from cattle exposed orally to the agent of BSE was assayed by the intracerebral inoculation of cattle. In addition to the infectivity in the central nervous system and distal ileum at stages of pathogenesis previously indicated by mouse bioassay, traces of infectivity were found in the palatine tonsil of cattle killed 10 months after exposure. Because the infectivity may therefore be present throughout the tonsils in cattle infected with BSE, observations were made of the anatomical and histological distribution of lingual tonsil in the root of the tongue of cattle. Examinations of tongues derived from abattoirs in Britain and intended for human consumption showed that macroscopically identifiable tonsillar tissue was present in more than 75 per cent of them, and even in the tongues in which no visible tonsillar tissue remained, histological examination revealed lymphoid tissue in more than 90 per cent. Variations in the distribution of the lingual tonsil suggested that even after the most rigorous trimming of the root of the tongue, traces of tonsillar tissue may remain. 相似文献
