Improved nutrient‐use efficiency is important to sustain agricultural production. The goal of our study was to investigate the effects of Azovit® (Azotobacter chroococcum) inoculation of seed with N fertilization on crop yield, nutrient uptake, and N‐use efficiency (NUE) of irrigated cotton (Gossypium hirsutum L. cv. C‐6524) in secondary saline soil under continental climatic conditions of Uzbekistan. A randomized complete block design in a 4 × 2 split‐plot experiment was established in the fall of 2013. The main plot was N fertilization (0, 140, 210, and 280 kg ha?1) and the subplot was Azovit inoculation. Azovit inoculation consistently increased the seed and lint yields of cotton by 25 and 27.9%, respectively, at 210 kg N ha?1 compared to the respective control. Azovit with 210 kg N ha?1 significantly increased the cotton harvest index by 21%, when compared to the control. Likewise, nutrient uptake and NUE of cotton were higher when N (210 kg ha?1) was applied with Azovit, as compared to other treatment combinations. An extrapolation of the relationship of relative yield vs. N fertilization showed that Azovit at 210 kg N ha?1 was sufficient to obtain near‐maximum cotton production (90%) with highest NUE, as compared to the respective control. The results suggest that Azovit with 210 kg N ha?1 produces cotton yield higher and/or comparable with the currently used rates of 280 kg N ha?1 or higher, suggesting savings of 70 kg N ha?1 for cotton production in saline soils under continental climatic conditions.  相似文献

DNA sequence analysis of the psbA gene encoding the D1 protein of photosystem II (PS II), the target site of PS II-inhibiting herbicides, identified a point mutation (Asn266 to Thr) in a bromoxynil-resistant Senecio vulgaris L. population collected from peppermint fields in Oregon. Although this mutation has been previously reported in Synechocystis, this is the first report of this particular point mutation in a higher plant exhibiting resistance to PS II-inhibiting herbicides. The resistant population displayed high-level resistance to bromoxynil and terbacil (R/S ratio 10.1 and 9.3, respectively) and low-level resistance to metribuzin and hexazinone (R/S ratio 4.2 and 2.6, respectively) when compared with the susceptible population. However, the population was not resistant to the triazine herbicides atrazine and simazine or to the urea herbicide diuron. A chlorophyll fluorescence assay confirmed the resistance levels and patterns of cross-resistance of the whole-plant studies. The resistant S. vulgaris plants produced fewer seeds. Differences in cross-resistance patterns to PS II-inhibiting herbicides and the difference in fitness cost could be exploited in a weed management program.  相似文献

We examined the effects of baking time and temperature for the preparation of par‐baked French bread, and of thawing and second baking conditions on the characteristics of bread prepared from par‐baked bread. Par‐baked French bread with loaf volume and crumb structure comparable to fully baked bread (control) was obtained with ≥6 min baking at 218°C, which increased the crumb temperature to 97°C. Freezing, thawing, and second baking of par‐baked bread decreased loaf volume by ≥100 mL. The second baking time of par‐baked bread, which was adjusted to have the bread crumb subjected to 97°C for 14 min based on the crumb temperature profile, produced a darker crust of bread compared with the control. The par‐baked bread with 6 min of initial baking at 218°C and frozen at ‐30°C required 12 min of second baking after thawing for 180 min to ≈20°C to produce crust color, crumb moisture, and firmness comparable to that of the control. When thawing time of par‐baked bread was shortened from 180 to 0 min, the second baking time required to yield crust color similar to the control increased from 12 to 16 min. The crumb moisture content was higher in bread baked for 16 min without thawing par‐baked bread than those baked after thawing for 45 or 180 min. Lowering the initial baking temperature of par‐baked bread from 246 to 163°C with the adjustment of baking time from 4 to 12 min decreased crumb firmness of the re‐baked (218°C, 16 min) bread from 2.5 to 1.5 N at 2 hr after baking and from 9.8–10.3 to 6.2–6.3 N at 48 hr.  相似文献

We investigated the relationship between the protein content and quality of wheat flours and characteristics of noodle dough and instant noodles using 14 hard and soft wheat flours with various protein contents and three commercial flours for making noodles. Protein content of wheat flours exhibited negative relationships with the optimum water absorption of noodle dough and lightness (L*) of the instant noodle dough sheet. Protein quality, as determined by SDS sedimentation volume and proportion of alcohol‐ and salt‐soluble protein of flour, also influenced optimum water absorption and yellow‐blueness (b*) of the noodle dough sheet. Wheat flours with high protein content (>13.6%) produced instant noodles with lower fat absorption, higher L*, lower b*, and firmer and more elastic texture than wheat flours with low protein content (<12.2%). L* and free lipid content of instant noodles were >76.8 and <20.8% in hard wheat flours of high SDS sedimentation volume (>36 mL) and low proportion of salt‐soluble protein (<12.5%), and <75.7 and >21.5% in soft wheat flours with low SDS sedimentation volume (<35 mL) and a high proportion of salt‐soluble protein (>15.0%). L* of instant noodles positively correlated with SDS sedimentation volume and negatively correlated with proportion of alcohol‐ and salt‐soluble protein of flour. These protein quality parameters also exhibited a significant relationship with b* of instant noodles. SDS sedimentation volume and proportion of salt‐soluble protein of flours also exhibited a significant relationship with free lipid content of instant noodles (P < 0.01 and P < 0.001, respectively). Protein quality parameters of wheat flour, as well as protein content, showed significant relationship with texture properties of cooked instant noodles.  相似文献

Sponges are at the forefront of marine natural product research. In the deep sea, extreme conditions have driven secondary metabolite pathway evolution such that we might expect deep-sea sponges to yield a broad range of unique natural products. Here, we investigate the chemodiversity of a deep-sea tetractinellid sponge, Characella pachastrelloides, collected from ~800 m depth in Irish waters. First, we analyzed the MS/MS data obtained from fractions of this sponge on the GNPS public online platform to guide our exploration of its chemodiversity. Novel glycolipopeptides named characellides were previously isolated from the sponge and herein cyanocobalamin, a manufactured form of vitamin B12, not previously found in nature, was isolated in a large amount. We also identified several poecillastrins from the molecular network, a class of polyketide known to exhibit cytotoxicity. Light sensitivity prevented the isolation and characterization of these polyketides, but their presence was confirmed by characteristic NMR and MS signals. Finally, we isolated the new betaine 6-methylhercynine, which contains a unique methylation at C-2 of the imidazole ring. This compound showed potent cytotoxicity towards against HeLa (cervical cancer) cells.  相似文献

This study was conducted to understand the effect of N supply in autumn on its uptake and distribution in tree parts and the utilization of reserve N the following year in persimmon trees (Diospyros kaki cv. Fuyu). The treated trees received 22.5-g N each for two successive years as a 3.5% urea solution from September 18 at 5-day intervals. Trees absorbed about 30% on average of the N applied in autumn. Sixty four to 83% of the N absorbed in autumn was in perennial parts, and 65–72% of that was in roots. Total N in perennial parts of the tree fertilized with N increased by over 4.28 g while leaf N changed little during senescence, indicating that the reserve N was constituted mostly by the N absorbed in autumn. Total N in the new growth was about the same as the amount of N declined in spring from perennial parts, indicating that there was little contribution by soil N to sustain new growth. Total dry weights of new growth the following year in a with-N tree were greater by over 34 g than those in a without-N tree.  相似文献

Aeromonads are ubiquitous in aquatic environments and have been implicated in fish and human infections. In this study, we isolated, studied antimicrobial susceptibility patterns and screened the existence of 15 virulence genes in aeromonads from two famously consumed fish species—seven marine Tiger Grouper (Epinephelus fuscoguttatus) and eight freshwater Marble Goby (Oxyeleotris marmoratus) from the aquaculture hatchery in Sabah, Malaysia. A total of 30 aeromonads (17 A. caviae, 9 A. rivuli, 4 A. dhakensis) were identified using PCR targeting GCAT gene, rpoD‐restriction fragment length polymorphism and multi‐locus phylogenetic analysis. All 30 strains were resistant to amoxicillin and cephalothin and five strains were multidrug‐resistant. Nine virulence genes (lip, ela, eno, fla, aerA, hylA, dam, alt and ser) present in A. dhakensis, suggesting the virulence potential of this species as a fish pathogen. This study offers as a baseline for future studies in monitoring and managing these two fish in aquaculture industry.  相似文献

Malaysia is a non‐endemic country for hepatitis E virus (HEV) infection. However, seroprevalence as high as 50% among samples of aboriginal people were reported over two decades ago. A total of 207 samples collected from seven aboriginal villages in rural settlements across two states in Malaysia were analysed for anti‐HEV IgG and IgM by an enzyme‐linked immunoassay. Following the detection of anti‐HEV seroprevalence, we organized health outreach to inform and educate the community. Qualitative interviews were conducted with individuals tested positive for anti‐HEV antibodies. Data derived from interviews and observations were used to investigate possible lifestyle behaviours associated with HEV infection. Anti‐HEV IgG was detected in six samples (5.9%) from the village of Dusun Kubur. Qualitative inquiry and observation study revealed poor dietary and household hygiene, contaminated food and water, contact with animal faeces, unsanitary and domestic waste disposal, and wildlife reservoirs could be the contributing factors for transmission and acquisition of HEV infection. Investigation during health outreach is important to provide insights for future empirical research and implementation for improvement of lifestyle behaviours among the aborigines. Managing the risk of HEV infection in the aborigines may reduce the risk of HEV transmission to the local communities.  相似文献

Buckwheat (Fagopyrum esculentum), a highly nutritious pseudocereal rich in bioactive compounds, is principally cultivated in central and eastern European countries. Buckwheat groats and husks of 10 cultivars were subjected to nutritional composition analysis and in vitro starch digestibility determination. Significant genetic variation was detected in buckwheat groats for 1,000‐kernel weight (16.5–39.8 g), protein content (10.2–17.9%), soluble dietary fiber (1.4–3.4%), insoluble dietary fiber (2.3–8.6%), total dietary fiber (3.6–10.6%), free phenolics (4.5–17.1 mg of gallic acid equivalent [GA]/g), and total phenolics content (6.8–20.7 mg of GA/g). The buckwheat husks exhibited large differences between cultivars in protein content (3.0–6.5%), bound phenolics (6.7–26.1 mg of GA/g), and total phenolics content (32.4–58.6 mg of GA/g), which was 1.5–8 times higher than in the groat. Cooked and cooled buckwheat groats exhibited lower starch digestibility and greater resistant starch content than raw buckwheat groats. Buckwheat cultivars with unique nutritional composition, such as Co901 and Ta‐1, were identified for future breeding.  相似文献
