Synthetic agonists of TLR9 containing novel DNA structures and R'pG (wherein R=1-(2'-deoxy-beta-d-ribofuranosyl)-2-oxo-7-deaza-8-methyl-purine) motifs, referred to as immune modulatory oligonucleotides (IMOs), have been shown to stimulate T(H)-1-type-immune responses and potently reverse allergen-induced T(H)-2 responses to T(H)-1 responses in vitro and in vivo in mice. In order to investigate the immunomodulatory potential of IMOs in dogs, canine peripheral blood mononuclear cells (PBMC) from healthy dogs were stimulated with three different IMOs and a control IMO, alone or in combination with concanavalin A (ConA). Lipopolysaccharide (LPS) was used as a positive control for B lymphocyte activation. Carboxyfluorescein diacetate succinimidyl ester and phenotype staining was used to tag proliferating T and B lymphocytes (CD5(+) and CD21(+)) by flow cytometry. Real-time PCR and ELISA were processed to assay cytokine production of IFN-gamma, IL-10, TGF-beta, IL-6 and IL-10. Like LPS, IMOs alone induced neither proliferation of CD5(+) T cells nor CD21(+) B cells, but both LPS and IMO had the capacity to co-stimulate ConA and induced proliferation of B cells. In combination with ConA, one of the IMOs (IMO1) also induced proliferation of T cells. IMO1 also significantly enhanced the expression of IFN-gamma on the mRNA and protein level in canine PBMC, whereas expression of IL-10, TGF-beta and IL-4 mRNAs was not induced by any of the IMOs. These results indicate that in canine PBMC from healthy dogs, IMO1 was able to induce a T(H)-1 immune response including T- and B-cell proliferation. 相似文献

Endometrial expression of oestrogen receptor‐α (ERα), progesterone receptor (PR) and cyclooxigenase‐2 (COX‐2) was evaluated in non‐pregnant and pregnant llamas during the period when luteolysis/maternal recognition of pregnancy is expected to occur. Females (n = 28) were divided into two groups: non‐pregnant llamas were induced to ovulate with a Buserelin injection, and endometrial biopsies were obtained on day 8 (n = 5) or 12 (n = 5) post‐induction of ovulation. Animals of the pregnant group (n = 18) were mated with a fertile male. Pregnancy was confirmed by the visualization of the embryo collected by transcervical flushing in 5 of 9 animals on day 8 post‐mating and by progesterone profile on day 12 post‐mating in 4 of 9 animals, when endometrial biopsies were obtained. An immunohistochemical technique was used to evaluate receptors population and COX‐2 expression. Pregnant llamas showed a higher percentage of positive cells and stronger intensity for ERα than for non‐pregnant llamas in stroma on day 8 and in the luminal epithelium on day 12 post‐induction of ovulation, while a deep decrease in endometrial PR population was reported in pregnant llamas on that day in luminal and glandular epithelia and stroma. In the luminal epithelium, COX‐2 expression was lower in pregnant than in non‐pregnant animals. Briefly, the increase of ERα in pregnant llamas gives further support to the hypothesis that oestrogens are involved in the mechanism of maternal recognition of pregnancy. Endometrial PR decrease in pregnant llamas might be a necessary event to allow the expression of proteins involved in conceptus attachment, a mechanism widely accepted in other species. Moreover, embryo seems to attenuate maternal PGF2α secretion during early pregnancy by decreasing the endometrial expression of COX‐2 in the luminal epithelium of pregnant llamas. 相似文献

Renal disease includes conditions affecting the glomeruli, tubules, interstitium, pelvis, and vasculature. Diseases of the kidney include glomerular diseases, diseases of the tubules and interstitium, diseases of renal pelvis, and developmental abnormalities. Renal tissue samples (n = 70) submitted to the Department of Veterinary Pathology of Konkuk University from 2003 to 2008 were included in this study. Tissue histopathology was performed using light microscopy with hematoxylin and eosin stains. Masson''s trichrome, Congo Red, and Warthin starry silver staining were applied in several individual cases. Glomerular diseases (22.9%), tubulointerstitial diseases (8.6%), neoplastic diseases (8.6%), conditions secondary to urinary obstruction (24.3%), and other diseases (35.7%) were identified. Glomerulonephritis (GN) cases were classified as acute proliferative GN (5.7%), membranous GN (4.3%), membranoproliferative GN (4.3%), focal segmental GN (2.9%), and other GN (4.2%). The proportion of canine GN cases presently identified was not as high as the proportions identified in human studies. Conversely, urinary obstruction and end-stage renal disease cases were relatively higher in dogs than in human populations. 相似文献

The Korean 2008 self-sufficiency rate for grain was only 26.2%. Because of this, the quantity virtual water (VW) for crop product imports is much greater than that of other countries. International VW trade is especially important to Korea due to its dependency on foreign imports to maintain food security and to establish an agricultural water resource policy. Using international crop products trade statistics during 2003?C2007, this study analyzed the virtual water content (VWC) and international virtual water flow (VWF) of major crops. The national water savings and global water savings were also estimated. Major grain products, including 28 products made from 13 crops, were selected for the analysis, based on the net import and export of products totaling more than 10,000 tons. VWCs were computed for the selected major crop products using the VWC of the primary crop of Korea. International VWFs were estimated using the VWC of each crop products. The amount of imported VW was 16,804 and 226?M?m3 was exported, so that the net imported VW was 16,578?M?m3. VW import is concentrated in wheat, rice, maize (corn), and soybean crops. A small number of countries, including the USA, China, Brazil, etc., account for over 96% of the imported VW, indicating Korea??s heavy dependence on these countries. The average national water savings for Korea and the average global water savings according to crop were estimated using VW flow from international crop products trade during 2003?C2007. The estimate of national water savings was 23,870.3?M?m3. Three major crops, namely wheat, maize and soybean, account for 95.3% of this total VW saving. Global water savings from the VW trade amounted to 7,253.0?M?m3. Korea depends heavily on VW imports concentrated in specific crops and which are primarily imported from a particular set of countries. This indicates that Korea is vulnerable to disruptions in the international grain harvest such as those caused by natural disasters such as floods and drought. Any such disruption could easily become a critical issue for governmental planners who establish food and water supply policies for Korea. 相似文献

The effect of functional rice giant embryo and Aranghyangchal on the glucose metabolism and antioxidative defense status in high fat-fed C57BL/6N mice were investigated. The animals were randomly divided into four dietary groups: normal control (NC), high fat (HF), and high fat supplemented with giant embryo rice (HF-GE) or Aranghyangchal rice (HF-AR). After 8 weeks of feeding them with experimental diets, the HF mice exhibited a marked increase in the blood glucose concentration, plasma insulin level, and lipid peroxidation and a significant decrease in the hepatic glycogen level relative to the NC group. On the other hand, diet supplementation of the rice samples suppressed this high fat diet-induced hyperglycemia and oxidative stress through inhibition of the glucose-regulating enzymes and activation of the antioxidant enzymes. These findings demonstrate that the giant embryo and Aranghyangchal rice may be beneficial as biomaterials in the development of functional food with hypoglycemic and antioxidative properties. 相似文献

This retrospective study was done to characterize the levels of vascular endothelial growth factor (VEGF) and hypoxia inducible factor 1 (HIF-1α) in dog brains with neo-vascularization in the cerebral cortex of frontal, temporal, and parietal lobe by using immunohistochemistry (IHC) and Western blot. In neo-vascularized (NV) brains, we analyzed the number and area of blood vessels and the expression of VEGF and HIF-1α. The IHC results showed that the number and area of blood vessels, as assessed by immunolabeling for von Willebrand factor, was higher in the NV brain than in the control brain. The Western blot results showed that the level of VEGF was increased, predominantly in NV brain of the cerebral cortex relative to the clinically normal cerebral cortex, whereas the expression of HIF-1α in NV brains was not different from the control brains. Our study showed that dilatation of vessels and development of new vessels in the cerebral cortex were observed in cases of canine CNS disease and found increased expression of VEGF in canine brains with neo-vascularization. 相似文献
