In May 2012, an outbreak of campylobacteriosis occurred in southern Sweden at a wedding reception affecting 44 persons. A total of 17 cases were notified (13 were culture positive for Campylobacter spp.). Epidemiological investigation suspected chicken liver pâté as the source of infection. The liver pâté had been deliberately undercooked, lightly fried to keep the right texture and mixed with spices. Campylobacter isolates from six cases as well as three Campylobacter isolates from chicken flocks previously raised by the producer delivering the liver were subtyped using pulsed‐field gel electrophoresis and whole‐genome sequencing. Indistinguishable PFGE profiles were identified among five human and one chicken C. jejuni isolates as well among the two C. coli isolates, one from a human case and one from a chicken. WGS supported the PFGE findings; the six C. jejuni isolates belonged to one cluster. All these six isolates were of MLST type ST 50 (ST‐CC 21). This study highlights the importance of a combination of strict biosecurity at the flock‐level as well as adequate cooking of chicken liver to prevent transmission of Campylobacter to humans. 相似文献

The life cycle of the endangered freshwater pearl mussel (Margaritifera margaritifera) includes a parasitic larval phase (glochidia) on the gills of a salmonid host. Glochidia encystment has been shown to affect both swimming ability and prey capture success of brown trout (Salmo trutta), which suggests possible fitness consequences for host fish. To further investigate the relationship between glochidia encystment and behavioural parameters in brown trout, pairs (n = 14) of wild‐caught trout (infested vs. uninfested) were allowed to drift feed in large stream aquaria and foraging success, activity, agonistic behaviour and fish coloration were observed. No differences were found between infested and uninfested fish except for in coloration, where infested fish were significantly darker than uninfested fish. Glochidia load per fish varied from one to several hundred glochidia, however, and high loads had significant effects on foraging, activity and behaviour. Trout with high glochidia loads captured less prey, were less active and showed more subordinate behaviour than did fish with lower loads. Heavy glochidia loads therefore may negatively influence host fitness due to reduced competitive ability. These findings have implications not only for management of mussel populations in the streams, but also for captive breeding programmes which perhaps should avoid high infestation rates. Thus, low levels of infestation on host fish which do not affect trout behaviour but maintains mussel populations may be optimal in these cases. 相似文献

Cultivation systems with mulching have been successfully tested in spring cereals for several years in Sweden. The water conserving effect of the mulch was considered to be important in these field experiments. Three experiments with lysimeters were conducted in this study to investigate the effect on evaporation of a) different amounts of mulch, b) different types of mulch and c) different frequencies of rain in combination with mulching. The lysimeters contained undisturbed soil cores from a heavy clay soil. The protection against evaporation was largest at the beginning of soil drying. It increased with increasing amounts of mulch, but there were no differences in protection between the various kinds of mulch tested. Accumulated evaporation was 12, 14 and 30% lower in the mulched compared to bare soil after 28 days with no rain, infrequent rain and frequent rain respectively. The implications for designing a cultivation system with mulching are discussed as well as its potential in different weathers. 相似文献

Background
In late 2011, a new Orthobunyavirus of the Simbu serogroup named Schmallenberg virus (SBV) emerged in continental Europe. The virus is transmitted by hematophagous arthropods, with the Culicoides species as, so far known, main vectors. Infection with the virus can cause clinical signs in adult ruminants including diarrhea, fever and reduced milk production. Transplacental infection of the developing fetus can lead to malformations of varying severity. To assess seroprevalence of SBV in Sweden an indirect enzyme-linked immunosorbent assay (ELISA) was established in connection with the surveys. Here, we describe the development and evaluation of the indirect ELISA, based on whole virus as the coating antigen and a monoclonal antibody for the detection of antibodies to SBV in ruminant sera. The evaluation includes comparison between the in-house ELISA, virus neutralization test and an indirect commercial ELISA.Results
The optimal working dilutions of antigens and conjugate were estimated with checkerboard titrations. Comparative studies, including ROC analyses, were used for the selection of an optimal cut-off (S/P value = sample value as percentage of positive control value). With an estimated S/P value of 15% the whole virus ELISA showed a specificity of 100% and a sensitivity of 99.19% compared to virus neutralization test (VNT) and with a good consistency as shown in reproducibility and variability experiments. Furthermore, the comparison of our whole virus indirect ELISA to an indirect ELISA with a SBV nucleoprotein antigen, demonstrated a higher sensitivity of our test.Conclusion
The indirect whole virus ELISA described in this paper is a readily available test for serological analysis of SBV antibodies. Since this in-house ELISA demonstrates a specificity and sensitivity comparable to virus neutralization test and also shows a higher sensitivity compared to commercially available indirect ELISA, it is a useful alternative for surveillance and screening purposes of SBV. 相似文献46.

AbstractWhen developing total merit indices (TMI's), estimated breeding values (EBV's) may be lacking for several traits. Methodology exists to address these challenges, calculating index weights using economic values and genetic (co)variances between traits included in the TMI and aggregate genotype. TMI's including production and functional traits were developed for beef breeds in the Norwegian breeding scheme, and potential for genetic gain was evaluated. Selection based on the TMI developed in this study was found to improve growth and carcass quality, while genetic deterioration was expected for calving difficulty. Alternative selection indices were applied to avoid these effects, but reduced the genetic gain for production traits. The results of our study show that is possible to achieve genetic gain in most economically important traits for beef cattle by using a TMI. When additional EBV's and genetic correlations become available, these should be included into the TMI for further improvement. 相似文献

Point-source infections are most likely the cause for Neospora caninum-induced abortion outbreaks in cattle, whereas an increased annual abortion rate may be a consequence of vertical transmission. The aims of the present study were to examine the reproductive effects of neosporosis in a beef herd for 3 years, after a point-source outbreak and to use IgG avidity serology to examine the chronicity of infections and patterns of transmission. During the study, 76-78% of animals were seropositive for N. caninum. The pregnancy rate varied from 88% to 94%, without any reduction in the pregnancy rate of seropositive cows compared with seronegative cows. The annual abortion rate was 2.5-5.5%, and all but 1 abortion occurred in seropositive dams. The efficiency of vertical transmission was estimated to be 85%. Several calves, born to seronegative dams, were seropositive at 6-13 months of age, indicating a 22% mean annual rate of horizontal transmission. The mean avidity in seropositive cows increased from 30 during the initial outbreak to 74 after 3 years. The mode of IgG avidity was 21-40 during the initial abortion outbreak, 41-60 after 1 year, and 61-80 after 2 and 3 years. The results reveal high annual rates of both vertical and horizontal transmission of N. caninum in a herd of beef cows and provide further validation on the ability of the N. caninum IgG avidity ELISA to accurately assess the chronicity of infection. 相似文献

Gastric evacuation rate in brown trout (Salmo trutta L.) was studied in water qualities characteristic of non-acidified and acidified waters. The level of plasma chloride was reduced and hematocrit increased in the group exposed to acid water. The gastric evacuation rate was described by an exponential function. Instantaneous rates of gastric evacuation were not significantly different between the two groups. In both of the groups the rates were lower than observed in corresponding studies of brown trout. The chronically acid Hovvatn and the adjoining pond Pollen were limed in March 1981. During the first 4 yr after the liming, a total of 11 437 brown trout were stocked in the two locations. The fish population was monitored by annual testfishing in a five yr period following the liming. The stocking material constituted 6 cohorts and included fish stocked at age 0+, 1+ and 2+. The stocking program assured a 7.9 times higher density of fish ha?1 in Pollen than in Hovvatn. Consequently, the fish from Hovvatn grew significantly better and had a significantly higher condition factor than fish in Pollen. Monitoring of water chemistry showed that the cohorts were exposed to a gradual reacidification process, abrupted by episodic events of severe acidification. Within 2–3 yr after the liming, the locations had reacidified to conditions which were considered critical for fish. However, the acidinduced increase in mortality appeared 1–2 yr later than what was expected from water chemistry data. This discrepancy could most likely be ascribed to the existence of water chemical refugia which enhanced the survival of the fish. Recaptures were significantly correlated to pH, Ca, labile-Al and ANC. Growth were significantly reduced throughout the reacidification period. Estimated yield showed that fish stocked at age 2+, as opposed to fish stocked at a younger age, managed to turn the high food availability into high growthrates before the reacidification retarded growth and survival. 相似文献
