The root-knot nematodes (Meloidogyne spp.) are important nematode pests and cause serious diseases in pepper in the world. No molecular markers linked to the nematodes resistance N gene have been reported. In this paper, ‘Carolina Wonder’ (Capsicum annuum L.), a sweet pepper line resistant to root-knot nematode with N gene, ‘20080-5-29’ (C. annuum L.), an inbred line susceptible to root-knot nematode with good horticultural characteristics, and their F2 progeny with 320 individuals were used as materials. Evaluation of resistance and susceptibility of parental lines, F1 and F2 progeny inoculated with root-knot nematodes (Meloidogyne incognita) were carried out. ‘Bulked segregant analysis’ method was used to search for polymorphic markers from 512 pairs of AFLP primers. Based on the assessment of resistance and susceptibility and polymorphism of the AFLP marker in F2 population, the genetic linkage distance between the AFLP marker and the N gene was estimated. One AFLP marker E39/M41-339 was obtained and transferred to a SCAR marker amplifying a 315 bp DNA fragment linked to the N resistant allele and a 331 bp fragment linked to the N+ susceptible allele. The distance between the molecular marker and the nematodes resistance N gene is 6.3 cM. This research delivered a valuable tool for the marker assisted selection of nematodes resistance in pepper.  相似文献

Estimation of the apparent digestibility coefficient ADC(%) of C3 and C4 plants in experimental diets for fingerlings of tambaqui (Colossoma macropomum, Cuvier, 1818) was calculated by applying chromic oxide (Cr2O3) external marker methodology and by a proposed mathematical expression based on the isotopic composition (δ13C). A total of 240 tambaqui fingerlings each weighing ±48.2 g and measuring ±9.8 cm were maintained in eight 500‐L aquariums specially designed for faeces collection. The ADC(%) of the C3 and C4 diets did not differ significantly between the two methods, producing results of 75.6%; 76.2% and 74.4%; 72.8%, respectively. The ADC(%) results obtained by isotopic method presented less variation than by chromic oxide. The proposed mathematical expression for calculating the ADC(%) based on δ13C values offers an alternative methodology, which can reduce errors and diminish the effort required to collect biological material. However, it is important to note that this method is limited to analysis of diets or food items with distinct isotopic signals.  相似文献

Effects of dietary l ‐carnitine were studied in juvenile black sea bream (Sparus macrocephalus). The semipurified basal diet [crude protein 450 g kg?1 dry matter (DM); crude lipid 126 g kg?1 DM] was formulated to choose white fishmeal as the protein source and fish oil plus corn oil (1 : 1) as the lipid source. Six diets (control + diets 1–5) containing 0.1, 0.12, 0.16, 0.24, 0.39 and 1.1 g of l ‐carnitine kg?1 diet were fed to triplicate groups of black sea bream (initial weight 13.10 ± 0.05 g) for 8 weeks. At the end of the feeding trial, growth performance, body composition and antioxidant status were determined. The results showed that relative growth rate (RGR) was significantly improved by the elevation of dietary l ‐carnitine level from 0.1 to 0.24 g kg?1, but decreased with further increment (P < 0.05). Lipid content decreased significantly (P < 0.05) in the dorsal muscle whereas increased (P < 0.05) in the liver with the addition of dietary l ‐carnitine. Dietary l ‐carnitine supplements elevated enzymatic antioxidants (superoxide dismutase, SOD; catalase, CAT; glutathione‐S‐transferase, GST) activities (P < 0.05) yet decreased the content of non‐enzymatic factor, total sulphydryl groups (TSH) (P < 0.05). In summary, the optimum dietary l ‐carnitine level was 0.284 g kg?1 diet by second‐polynomial regression analysis based on RGR (y = ?647.4x2 +367.97x + 234.55; R2 = 0.977, x = dietary l ‐carnitine levels, y = RGR), and dietary l ‐carnitine addition within the levels adopted in our study could depress lipid peroxidation in tissues of juvenile black sea bream.  相似文献

A 30‐day feeding experiment was conducted in blue tanks (70 × 50 × 60 cm, water volume 180 L) to determine the effects of dietary lipid levels on the survival, growth and body composition of large yellow croaker (Pseudosciaena crocea) larvae (12 days after hatchery, with initial average weight 1.93 ± 0.11 mg). Five practical microdiets, containing 83 g kg?1 (Diet 1), 126 g kg?1 (Diet 2), 164 g kg?1 (Diet 3), 204 g kg?1 (Diet 4) and 248 g kg?1 lipid (Diet 5), were formulated. Live feeds (Artemia sinicia nauplii and live copepods) were used as the control diet (Diet 6). Each diet was randomly assigned to triplicate groups of tanks, and each tank was stocked with 3500 larvae. During the experiment, water temperature was maintained at 23(±1) °C, pH 8.0 (±0.2) and salinity 25 (±2) g L?1. The results showed that dietary lipid significantly influenced the survival and growth of large yellow croaker larvae. Survival increased with the increase of dietary lipid from 83 to 164 g kg?1, and then decreased. The survival of larvae fed the diet with 83 g kg?1 lipid (16.1%) was significantly lower than that of larvae fed other diets. However, the survival in larvae fed the diet with 16.4 g kg?1 lipid was the highest compared with other artificial microdiets. Specific growth rate (SGR) significantly increased with increasing dietary lipid level from 83 to 164 g kg?1 (P < 0.05), and then decreased. The SGR in larvae fed the diet with 164 g kg?1 lipid (10.0% per day) was comparable with 204 g kg?1 lipid (9.6% per day), but were significantly higher than other microdiets (P < 0.05). On the basis of survival and SGR, the optimum dietary lipid level was estimated to be 172 and 177 g kg?1 of diet using second‐order polynomial regression analysis respectively.  相似文献

Clostridium butyrium CB2 isolated from chickens was tested as a potential fish probiotic in the Chinese drum, Miichthys miiuy . Fish were fed live (CB), dead CB2 (D-CB) cells (108 cells g−1) or spent culture supernatant (SCS), for 30 days and challenged with Vibrio anguillarum or Aeromonas hydrophila . Survival was higher in both the CB and the CB-D fed groups, but the SCS group was not significantly different from the control. After feeding live or dead CB2 cells, there was increase in phagocytic activity of the head kidney macrophages, the lysozyme activity of serum and gut mucosa and immunoglobulin (Ig) level. The SCS group showed no obvious change in immune parameters. The results suggest that live or dead Clostridium butyrium CB2 has an immunomodulatory effect on fish.  相似文献

Late blight, caused by the oomycete Phytophthora infestans (Mont.) de Bary, is a devastating disease in potato and tomato and causes yield and quality losses worldwide. The disease first emerged in central America and has since spread in North America including the United States and Canada. Several new genotypes of P. infestans have recently emerged, including US-22, US-23 and US-24. Due to significant economic and environmental impacts, there has been an increasing interest in the rapid identification of P. infestans genotypes. In addition to providing details regarding the various phenotypic characteristics such as fungicide resistance, host preference, and pathogenicity associated with various P. infestans genotypes, information related to pathogen movement and potential recombination may also be determined from the genetic analyses. Restriction fragment length polymorphism (RFLP) analysis with the RG57 loci is one of the most reliable procedures used to genotype P. infestans. However, the RFLP procedure requires propagation and isolation of the pathogen and relatively large amounts of DNA. Isolation of the late blight pathogen is sometimes impossible due to the poor condition of the infected tissues or the presence of fungicide residues. In this study, we describe a procedure to identify P. infestans at the molecular level in planta using terminal restriction fragment length polymorphism (T-RFLP) of the RG57 loci. This T-RFLP assay is sufficiently sensitive to detect and differentiate P. infestans genotypes directly in planta without propagation and isolation of the pathogen, to facilitate the timely implementation of best management practices.  相似文献

Potassium (K) fertilization and antioxidant enzymes both positively influence plant growth and development. However, it is not known whether K treatment improves fruit development via increasing soluble sugar. In this study, K-treated apple (Malus domestica Borkh.) fruit was harvested from 60 to 150 days after full bloom and was analyzed for ROS production and trehalose metabolism. The results show that K fertilization improved fruit firmness, increased growth according to several parameters, increased soluble sugar content, and decreased ROS production. The ascorbate metabolic pathway more effectively reduced ROS production than catalase and peroxidase (POD) did under K treatment. Trehalose-treated fruit also showed higher activity of ascorbate-related enzymes (DHAR, GR, and APX) compared with non-treated fruit. The changes of antioxidant enzyme activity in trehalose-treated fruit corresponded to those in K-treated fruit. Moreover, trehalase (TREH) activity in fruit was notably reduced by K treatment. This demonstrates that K influences ROS production via regulating trehalose content and TREH activity in fruit. This study provides new insight into the K mechanism which improves fruit development, including fruit firmness and size.  相似文献

The objective of this study was to evaluate the efficacy of potassium diformate (KDF) as a potential additive for alfalfa silage. Fresh alfalfa was untreated or treated with formic acid (4 g/kg fresh weight, FW) or three concentrations of KDF (4, 5.5 or 7 g/kg FW). After 60 days of ensiling, the addition of formic acid and greater levels of KDF (5.5 and 7 g/kg) effectively reduced silage pH and inhibited the undesirable bacteria, indicated by lower butyric acid, ethanol, ammonia N concentrations and microbial populations (including enterobacteria, yeasts, moulds and clostridia). Additives decreased the dry‐matter loss, and more water‐soluble carbohydrates were preserved in the silages with formic acid or potassium diformate than in the control. Alfalfa silages treated with formic acid at 4 g/kg FW or potassium diformate at 5.5 or 7 g/kg FW were classified as the highest quality silage based on the higher Flieg's point (above 70) and remained stable for more than 9 days during aerobic exposure. Potassium diformate is recommended as an effective additive for alfalfa silages at a level of 5.5 or 7 g/kg FW under the humid and hot conditions of southern China.  相似文献

Efficient management of whitefly-borne diseases remains a challenge due to the lack of a comprehensive understanding of their epidemiology, particularly of the diseases tomato golden mosaic and tomato yellowing. Here, by monitoring 16 plots in four commercial fields, the temporal and spatial distribution of these two diseases were studied in tomato fields in Brazil. In the experimental plots these diseases were caused by tomato severe rugose virus (ToSRV) and tomato chlorosis virus (ToCV), respectively. The incidence of each virus was similar in the plots within a field but varied greatly among fields. Plants with symptoms for both diseases were randomly distributed in three of four spatial analyses. The curves representing the progress of both diseases were similar and contained small fluctuations, indicating that the spread of both viruses was similar under field conditions. In transmission experiments of ToSRV and ToCV by Bemisia tabaci MEAM1 (former biotype B), these viruses had a similar transmission rate in single or mixed infections. It was then shown that primary and secondary spread of ToCV were not efficiently controlled by insecticide applications. Finally, in a typical monomolecular model of disease progress, simulation of the primary dissemination of ToSRV and ToCV showed that infected plants were predominantly randomly distributed. It is concluded that, although the manner of vector transmission differs between ToSRV (persistent) and ToCV (semipersistent), the main dispersal mechanisms are most probably similar for these two diseases: primary spread is the predominant mechanism, and epidemics of these diseases have been caused by several influxes of viruliferous whiteflies.  相似文献
