A clone bank of Mycoplasma gallisepticum (MG) strain A5969 DNA was prepared in the expression vector phage lambda gt11. Approximately 75% of the resulting phages were recombinants, based upon the insertional inactivation of the lacZ gene of the vector. Clones were screened immunologically with serum prepared from specific-pathogen-free white leghorn chickens that had been infected with aerosolized MG. Approximately 250 clones, or less than 1% of the recombinant phage, reacted positively to various degrees with the test serum and failed to react with serum from uninfected specific-pathogen-free control chickens. A single clone was chosen at random for comparison with a vector control by western immunoblot, revealing a polypeptide of 140,000 molecular weight in the clone profile but not the control profile that reacted with immune serum. Clones expressing MG antigens recognized during infection may provide an improved means for antigen preparation for serologic diagnosis of mycoplasmosis. 相似文献

The mechanisms underlying the social structure of free‐ranging fish shoals have received little attention in comparison to functional studies on shoaling. Recently, however, a number of investigations, both in the marine and in the freshwater environment, have begun to address the underlying mechanisms by concentrating on interactions between free‐ranging shoals. The rates of shoal encounters can influence the opportunities for individual assortment by phenotype and selection of shoal size; act as a constraint on the observed patterns of shoal structure by restricting individual choice behaviour and were found to be high in several small freshwater species (where intershoal distances were small), but lower in marine species where shoals were more dispersed. The duration of encounters may play a role in that it affects both the time available for assessment (of the encountered shoal) as well as that for exchange of individuals. Scarce published information on the outcome of shoal encounters suggests that the outcome of shoal encounters is influenced by shoal composition but not by shoal size. Individual behaviour may have evolved to maintain shoal size when shoals encounter, but when fish populations are depleted by fishing, this trait can exacerbate range and stock collapse. Furthermore, an understanding of the dynamics of shoal encounters has important consequences for the evolution of reciprocal altruism and the transmission of information through social learning within populations. Finally, information on encounter rates between shoals and the number of individuals that are exchanged on such occasions could be important for making predictions about the spread of disease through fish populations. 相似文献

Agricultural systems that receive high amounts of inorganic nitrogen (N) fertilizer in the form of either ammonium (NH4+), nitrate (NO3−) or a combination thereof are expected to differ in soil N transformation rates and fates of NH4+ and NO3−. Using 15N tracer techniques this study examines how crop plants and soil microbes vary in their ability to take up and compete for fertilizer N on a short time scale (hours to days). Single plants of barley (Hordeum vulgare L. cv. Morex) were grown on two agricultural soils in microcosms which received either NH4+, NO3− or NH4NO3. Within each fertilizer treatment traces of 15NH4+ and 15NO3− were added separately. During 8 days of fertilization the fate of fertilizer 15N into plants, microbial biomass and inorganic soil N pools as well as changes in gross N transformation rates were investigated. One week after fertilization 45-80% of initially applied 15N was recovered in crop plants compared to only 1-10% in soil microbes, proving that plants were the strongest competitors for fertilizer N. In terms of N uptake soil microbes out-competed plants only during the first 4 h of N application independent of soil and fertilizer N form. Within one day microbial N uptake declined substantially, probably due to carbon limitation. In both soils, plants and soil microbes took up more NO3− than NH4+ independent of initially applied N form. Surprisingly, no inhibitory effect of NH4+ on the uptake and assimilation of nitrate in both, plants and microbes, was observed, probably because fast nitrification rates led to a swift depletion of the ammonium pool. Compared to plant and microbial NH4+ uptake rates, gross nitrification rates were 3-75-fold higher, indicating that nitrifiers were the strongest competitors for NH4+ in both soils. The rapid conversion of NH4+ to NO3− and preferential use of NO3− by soil microbes suggest that in agricultural systems with high inorganic N fertilizer inputs the soil microbial community could adapt to high concentrations of NO3− and shift towards enhanced reliance on NO3− for their N supply. 相似文献

The multiple functional properties of egg yolk are mostly influenced by its complex protein composition. The high lipid content of egg yolk as well as the low solubility of delipidated egg yolk lipoproteins make analysis by conventional chromatographic or electrophoretic techniques a difficult task. This work describes a method to profile egg yolk proteins after delipidation with acetone using sodium dodecyl sulfate polyacrylamide gel electrophoresis on precast 8-18% T polyacrylamide gradient gels. Twenty bands were obtained for the whole egg yolk profile with molecular weights ranging between 5 and 221 kDa. The bands were identified based on their molecular weight and by comparison with isolated egg yolk subfractions. The dissociation behavior under reducing and nonreducing conditions provided additionally helpful information for identification and characterization of the yolk proteins. The method presented is very well suited for assaying the thermal sensitivity of whole yolk and its components and thus for the characterization of heat treatment processes. 相似文献

AOAC official method 29.119-29.125 was revised to determine ethylenethiourea (ETU) directly by a liquid chromatographic-electrochemical (LC-EC) determinative technique and to improve ETU recovery. ETU is extracted from food products with a methanol-aqueous sodium acetate solution. A portion of the concentrated filtrate is added to a column of diatomaceous earth, and ETU is eluted with 2% methanol in methylene chloride to separate it from food coextractives, which are retained on the column. The eluate is collected in a siliconized flask and evaporated, the residue is dissolved in water, and 20 microL of solution is injected onto an LC graphitized carbon column. ETU is eluted from the LC column with a mobile phase of acetonitrile-aqueous 0.1M phosphoric acid-water (5 + 25 + 70), and the eluted ETU is detected by using an amperometric electrochemical detector equipped with a gold/mercury working electrode. Recovery data were obtained by fortifying 13 food products with ETU: baked potatoes; canned applesauce, mushrooms, creamed spinach, green beans, spinach, and tomatoes; cooked fresh cabbage and frozen collards; fresh celery and lettuce; grape jelly; and powdered sugar cake donuts. Raw celery was found to cause low ETU recoveries. Average percent recoveries of ETU from the other 12 food products were 92 with a standard deviation of 12 for the low (0.05 and 0.1 ppm) fortification levels and 90 with a standard deviation of 6 for the higher (0.5 and 1 ppm) fortification levels.
