ABSTRACT:   The genetic diversity of wild and hatchery-released Pacific herring Clupea pallasii collected from three brackish lakes and two bays in Honshu and Hokkaido, Japan was examined with five microsatellite loci. All loci showed high genetic variability with expected heterozygosities ranging from 0.815 to 0.945. Significant differences in genotypic and allelic distributions were detected among all locations except for between the two bays in Honshu Island. Pairwise population analysis based on the F ST values showed close genetic relationships among the locations in Hokkaido Island, and the hierarchical analyses of molecular variance showed significant genetic difference between the two islands. Those results suggest the existence of subpopulations due to natal homing. In addition, stocked fish showed as much genetic diversity as the wild fish. The pairwise population analyses also showed close relationships between the hatchery fish and the wild fish in respective stocking areas, showing that no effects of stocking programs on genetic diversity of wild populations were detected.  相似文献

ABSTRACT:   Parvalbumin is a sarcoplasmic Ca2+-binding protein of 12 kDa and represents the major fish allergen. Several peptide segments are identified as immunoglobulin E (IgE)-binding epitopes of cod parvalbumin. However, carp parvalbumin (Cyp c 1) shows a markedly reduced IgE-binding ability upon depletion of Ca2+, suggesting the importance of conformational epitopes associated with Ca2+-chelating. In this study, the IgE reactivity of Pacific mackerel Scomber japonicus parvalbumin (Sco j 1) was demonstrated to be markedly reduced (60–100% reduction) by Ca2+-depletion, similar to Cyp c 1. Three Sco j 1 mutants (D51A, D90A, D51/90A), with modifications in either one or both of the two Ca2+-binding sites, were then constructed by site-directed mutagenesis, followed by expression in Escherichia coli , and evaluated for their IgE reactivity. Interestingly, the double mutant (D51/90A), probably devoid of Ca2+-binding capacity, exhibited a significantly reduced IgE reactivity (equivalent to 0.0–7.5% of the IgE reactivity of natural Sco j 1). The results suggest that the IgE-binding ability of Sco j 1 largely depends on the solid conformation mediated by Ca2+-chelating, and that the hypoallergenic D51/90A will be a useful tool for the specific immunotherapy of fish allergy.  相似文献

The rotifer Brachionus plicatilis (S-type Hawaiian strain) was cultured with various combinations of baker's yeast and Nannochloropsis oculata. There were no significant differences in the daily rotifer production and amino acid profiles of the resulting rotifers. The significantly lower levels of fatty acids (C14, C16, C20:4n−6, C20:5n−3 and C22:6n−3) observed in the rotifers were found to correspond with the amount of yeast presented in their diet. The low survival and growth of striped mullet larvae recorded at Day 15 posthatching indicates that rotifers fed only yeast were nutritionally deficient in fatty acids. The fatty acid requirements of mullet, however, appeared to be satisfied with rotifers cultured on a combination of yeast and N. oculata. In contrast, no significant differences in larval milkfish survival and growth at Day 10 posthatching were detected when using rotifers fed the various diets in the larval rearing protocol.  相似文献

ABSTRACT:   Quality control of imported fish products is a constant challenge for Customs officials. To help officials access the quality and integrity of imported fish products, and to enhance the effectiveness and accuracy of product screening, a method was developed to identify boiled and dried larval and juvenile sardines (Chirimen) at the genus level by direct sequencing using a partial sequence (606 bp) of mitochondrial 16S rDNA. A phylogenetic tree was constructed, based on nucleotide sequences of seven kinds of Chirimen imported from four countries with 16 standard samples belonging to the orders Clupeiformes and Salmoniformes. To identify the fish genera in imported Chirimen, bootstrap values and branch lengths of the phylogenetic tree as well as genetic distances were used. The data revealed that Chirimen imported from China and Korea contains species of the genus Engraulis . Regarding other imported Chirimen, the least that could be verified was that the sardine ingredients do not belong to regulated genera.  相似文献

We evaluated the annual process of oocyte and ovarian development of the planktivorous Amazonian catfish, the mapará (Hypophthalmus marginatus), aiming to support captive reproduction for domestication. A total of 149 females were captured from the Tocantins River at the Tucuruí Dam (3°49'55"S, 49°39'9"W) in 13 sampling events. For each individual, we evaluated ovary histology and plasma steroid concentrations. Two annual reproductive cycles, with similar characteristics, including a long period of rest, a short vitellogenic stage and a well‐defined spawning season from November to March, were described. A 17β‐estradiol (E2) rise and a 17α,20β‐dihydroxy‐4‐pregnen‐3‐one (DHP) peak were associated with vitellogenesis and spawning season, respectively, in the first cycle but not in the second reproductive cycle. In conclusion, the studied species presents a reproductive cycle similar to that of other migratory total spawners; however, unusually for this group of fish, there were three (but not two) batches of vitellogenic oocytes in “in maturation” and “mature” stage ovaries, pointing to the possibility of more than one spawn during the spawning season.  相似文献

The Lincoln strain of bovine rotavirus was found to replicate with cytopathic effects in cultures of GBK cells, a stable cell line derived from bovine kidney, when the cultures were maintained in the presence of trypsin. The virus was readily passaged and the infected cells were shown to contain specific viral antigen by indirect immunofluorescent staining. The virus formed plaques in GBK cell monolayers, when trypsin was incorporated in the agar overlay medium. The plaque count increased about twofold when diethylaminoethyl dextran was further included in the overlay medium. Plaque assay in GBK cells was more sensitive than that in MA-104 cells previously reported by Matsuno et al. The specificity of plaques was confirmed by specific inhibition with antiserum against the Lincoln strain.  相似文献

To disclose the mechanism of cellular injury following porcine circovirus (PCV) infection, 12 pigs were examined by the terminal deoxynucleotidyl transferase-mediated dUTP-nick end labeling (TUNEL) method and immunohistochemistry. Histologically, the lymphoid tissues were characterized by marked apoptosis of lymphocytes, lymphocyte depletion, and macrophages and giant cells containing numerous inclusion bodies with or without apoptotic bodies. Immunohistochemically, there were many lysozyme-positive macrophages in the lymphoid follicles, while the number of CD79a-positive B lymphocytes was scanty. Apoptotic cells, which were proved to be TUNEL positive, revealed CD79a positivity. Although detectable mainly in the cytoplasm of macrophages, PCV antigens were found also in the nuclei of macrophages and apoptotic lymphocytes. Ultrastructurally, the presence of PCV virions was confirmed in apoptotic bodies phagocytosed by macrophages. These findings suggested that lymphocyte depletion with apoptotic death of B lymphocytes was caused by PCV, and that some of the inclusion bodies were phagolysosomes derived from the apoptosis. Thus, PCV may trigger the development of wasting disease syndrome by producing an immunocompromised state in pigs.  相似文献
