High variability among individuals is often encountered when hemocyte characteristics are measured in bivalves. Such variability is suspected to result partly from genetic factors. In this study, hemocyte characteristics of six families of Crassostrea gigas were compared by flow cytometry at one sampling date in October 2001. These families were obtained from a nested, half-sibling cross design, and reared from July to October 2001 at three sites distributed along the French Atlantic coast from north to south: Baie des Veys (Normandy), Rivière d'Auray (Brittany) and Ronce (Marennes-oléron Basin, Poitou Charentes).
Among the 15 measured hemocyte characteristics, production of reactive oxygen species (ROS) of untreated hemocytes (maintained in filtered sterile seawater) and treated hemocytes (zymosan at 20 particles per hemocyte, and with Vibrio sp. S322 at 50 bacteria per hemocyte) was the most notable differences between families. This supports the existence of a genetic basis, at least partly, for the hemocyte characteristics of oysters, and especially for ROS production.
Among the six families analyzed, three have shown high survival during summer (named as “resistant”, mean mortality 5.2%) and three experienced high mortality during summer (named as “susceptible”, 30.6% mean mortality). Families showing high or low survival to summer mortality had similar hemocyte characteristics, regardless of the environmental conditions or reproductive state. Resistant families were observed to have higher total hemocyte counts and lower production of ROS than susceptible families. Moreover, ROS production of hemocytes from susceptible families was diminished significantly more by pathogenic Vibrio than that of resistant families. However, this study demonstrates also that rearing site strongly affected the hemocyte characteristics of all families of oysters, most notably hemocyte concentration and morphology (size and granularity), production of reactive oxygen species (ROS), and susceptibility to the cytotoxic activity of the pathogenic Vibrio sp. S322 (50 bacteria/hemocyte). Food availability and reproductive state are the most probable explanations for the site differences observed. Finally, it appeared difficult to link oyster survival during summer mortality to hemocyte profiles evaluated at one sampling date; other relevant indicators would probably help explaining oyster survival during summer mortality events. 相似文献
A trial in an 11-year-old stand of radiata pine (Pinus radiata D. Don) was used to analyse the effects of accelerated loss of nutrients from the site on forest productivity and nutrient status. Raking of litter was undertaken over 14 years prior to thinning, then for 2 years after thinning at which time the trial was destroyed in a wind storm. The experimental design was a factorial of three main treatments: (i) removal (raking) versus nil removal of the forest floor, (ii) replacement or non replacement of nutrients to adjust for imbalances between nutrients in litter and those in the tree stem, and (iii) complete replacement (or not) of all nutrients removed in the litter. Additionally, a small trial was incorporated to address components of physical aspects of litter removal by comparing raking with ‘raking and a cover of woven plastic mesh’. Raking and nutrient additions were carried out approximately every 6 months.Over the study period, the raking treatment removed about 75 Mg ha−1 of organic material with contained nutrients (559 kg ha−1 of N, 68 kg ha−1 of P, 323 kg ha−1 of Ca, 91 kg ha−1 of Mg, 243 kg ha−1 of K, 0.9 kg ha−1 of B) and this related to about four normal sawlog harvests or one total tree harvest. Up to the time of thinning, raking reduced basal area increment by 25% while raking together with replacement of nutrients reduced this by about 12%. Nutrient additions to unraked plots led to increases of up to 14% in basal area increment. The raking treatment reduced foliage nitrogen and this was correlated with reduced growth while other nutrients such as boron and sulphur were reduced but not to a degree to affect growth or health. Understory plant biomass, species richness and canopy openness were measured in six-year old hybrid poplar riparian buffer
strips, in the understory of two unrelated clones (MxB-915311 and DxN-3570), planted along headwater streams at three pasture
sites of southern Quebec. Canopy openness was an important factor affecting understory biomass in hybrid poplar buffers, with
lower understory biomass observed on sites and under the clone with lower canopy openness. Although tree size was an important
factor affecting canopy openness, relationships between total stem volume and canopy openness, for each clone, also support
the hypothesis of a clonal effect on canopy openness. Understory biomass and canopy openness as low as 3.6 g m−2 and 7.6% in 1 m2 microplots were measured under clone MxB-915311 at the most productive site. This reduction of understory plant growth could
compromise important buffer functions for water quality protection (runoff control, sediment trapping and surface soil stabilisation),
particularly were concentrated runoff flow paths enter the buffer. On the other hand, tree buffers that maintain relatively
low canopy openness could be interesting to promote native and wetland plant diversity. Significant positive relationships
between canopy openness and introduced species richness (R2 = 0.46, p < 0.001) and cover (R2 = 0.51, p < 0.001) were obtained, while no significant relationship was observed between canopy openness and native (wetland) species
richness and cover. These results suggest that planting riparian buffer strips of fast-growing trees can rapidly lead to the
exclusion of shade-intolerant introduced species, typical colonisers of disturbed habitats such as riparian areas of pastures,
while having no significant effect on native (wetland) diversity. Forest canopy created by the poplars was probably an important
physical barrier controlling introduced plant richness and abundance in agricultural riparian corridors. A strong linear relationship
(R2 = 0.73) between mean total species richness and mean introduced species richness was also observed, supporting the hypothesis
that the richest communities are the most invaded by introduced species, possibly because of higher canopy openness, as seen
at the least productive site (low poplar growth). Finally, results of this study highlight the need for a better understanding
of relationships between tree growth, canopy openness, understory biomass and plant diversity in narrow strips of planted
trees. This would be useful in designing multifunctional riparian buffer systems in agricultural landscapes. 相似文献
Symptoms resembling off-target plant growth regulator (PGR) herbicide injury are frequently found in soybean fields, but the causal agent is often difficult to identify. The expression of GH3, an auxin-regulated soybean gene, was quantified from soybean leaves injured by PGR herbicides using real-time RT-PCR. Expression of GH3 was analyzed to ascertain its suitability for use in a diagnostic assay to determine whether PGR herbicides are the cause of injury. GH3 was highly induced by dicamba within 3 days after treatment (DAT) and remained high at 7 DAT, but induction was much lower at 17 DAT. GH3 was also highly induced at 7 DAT by dicamba + diflufenzopyr, and to a lesser extent by the other PGR herbicides clopyralid and 2,4-D. The non-PGR herbicides glyphosate, imazethapyr, and fomesafen did not significantly induce GH3 expression above a low constitutive level. These results indicate that a diagnostic assay for PGR herbicide injury based on overexpression of auxin-responsive genes is feasible, and that GH3 is a potential candidate from which a diagnostic assay could be developed. However, time course analysis of GH3 expression indicates the assay would be effective for a limited time after exposure to the herbicide. 相似文献
Daylight observations on NW African and W Turkish populations of the Mediterranean spur-thighed tortoise Testudo graeca L. showed that activity, depending upon maturity, sex and size, is influenced by air temperature and sunshine. Tortoises are active above 18°C while refuge from sunshine during the middle of the day occurs above 28°C. Morning basking increases body temperature, which under glass in S England was shown to be thermoregulated at about 35°C. With movements in and out of sunshine, most heat is dissipated through the plastron. Field sighting frequency (no.man-h?1) as a relative index of abundance in low density populations is influenced by activity and behaviour type. This is discussed with regard to tortoise detection being increased by locomotory rustling and mating sounds in thick vegetation. 相似文献
Variability of phosphorus (P) availability among forage species and plant maturity is largely ignored when formulating ruminant diets. To determine if variability in P availability changes with forage species and/or maturity, ruminal in situ and total (ruminal+post-ruminal) dry matter (DM) and phosphorus disappearance (PD) from alfalfa (ALF; Medicago sativa) and coastal bermudagrass (CB; Cynodon dactylon) harvested at four stages of maturity was measured in cattle. Forages were hand clipped at 14, 21, 28 and 35 days after first cutting. Ruminal in situ DM disappearance (DMD) and PD were measured after 24 h ruminal incubation in Dacron bags. Total tract DMD and PD were measured using the mobile nylon bag technique. Disappearance of DM and P were greater (p≤0.05) in the rumen than post-rumen for both species regardless of maturity; however, 80 g PD/kg DM in 35-day ALF (9% of total PD) and 224 g PD/kg of 35-day CB (38% of total PD) occurred post ruminally. Alfalfa DM disappeared to a greater (p ≤ 0.05) extent than CB and showed 5% greater total tract PD at 14 days and 13% more at 35 days compared to the grass. Alfalfa total tract PD decreased (p ≤ 0.05) 5.4% from 14- to 35-day maturity while the decrease was far greater for CB, 12.4%. Results from this study indicate that ruminant nutritionists should take into account forage species and maturity when calculating PD in diets; these details can be used to aid in formulating more precise rations that reduce fecal-phosphorus excretion into the environment. 相似文献
Withdrawal periods required when doses of 24,000 IU and 66,000 IU of procaine penicillin G/kg body weight were administered to yearling beef steers by intramuscular injection daily for five consecutive days were investigated. These dosages are in excess of product label recommendations, but are in the range of procaine penicillin G dosages that have been administered for the treatment of some feedlot bacterial diseases. The approved dose in Canada is 7,500 IU/kg body weight intramuscularly, once daily, with a withdrawal period of five days. Based on the tissue residue data from this study, the appropriate withdrawal period is ten days for the 24,000 IU/kg body weight dose and 21 days for the 66,000 IU/kg body weight dose when administered intramuscularly to yearling beef steers. In a related study, 18 yearling beef steers received 66,000 IU of procaine penicillin G/kg body weight administered by subcutaneous injection, an extra-label treatment in terms of both dose and route of administration, typical of current practice in some circumstances. Deposits of the drug were visible at subcutaneous injection sites up to ten days after injection, with more inflammation and hemorrhage observed than for intramuscular injections of the same dose. These results suggest that procaine penicillin G should not be administered subcutaneously at high doses; and therefore a withdrawal period was not established for subcutaneous injection. 相似文献
