Here, we presented new insights of the development of poly(lactic-co-glycolic acid) nanoparticles containing turmeric compounds (turmeric-PLGA-NPs) using emulsion-solvent evaporation method. The nanoparticulate system was characterized by size, zeta potential, morphology, release profile, partition parameter, stability and encapsulation efficiency (%EE). Antioxidant activity studies were also evaluated. The Korsmeyer-Peppas model (Mt/M∞ vs. t) was used to determine the release mechanisms of the studied system. Our results demonstrated the emulsion-solvent evaporation method was shown advantageous for producing turmeric-PLGA-NPs in the range of 145 nm with high homogeneity in size distribution, zeta potential of ?21.8 mV and %EE about 72%. Nanoparticles were stable over a period of one month. In vitro study showed a release of curcumin governed by diffusion and relaxation of the polymeric matrix. The partition parameter of the extract in relation to blank-PLGA-NPs was 0.111?±?0.008 M?1, indicating a low affinity of curcumin for the polymer matrix. Antioxidant ability of the turmeric-PLGA-NPs in scavenging the radical 2,2-azinobis (3-ethylbenzothiazoline- 6-sulfonic acid) (ABTS) was inferior to free turmeric extract and showed a concentration and time-dependent profile. The study concluded that PLGA nanoparticles are potential carriers for turmeric extract delivery.

Isatis tinctoria L. (woad) is one of the earliest known sources of indigo in Europe where it was cultivated since the Middle Ages. Isatisindigotica Fort. (Chinese woad), widely distributed in China, had been used from ancient times as indigo-producing plant and medicinal plant. Both species produce indigo precursors indican (indoxyl β-d glucoside) and isatan B (indoxyl ketogluconate) in their leaves. In order to identify new suitable crops for indigo production in Italy, 17 woad lines were studied under field conditions in Central Italy (Pisa, 43°40′N, 10°19′E) from 2001 to 2003. We analyzed the effects of year, genotype, and harvest times together with their reciprocal interactions on leaf yield and indigo precursors production. Woad lines were then compared with seven I. indigotica lines in a field crop experiment set up in 2003. Extraction and quantification of indigo precursors were accomplished by HPLC-ELSD. In order to be able to fertilize oocytes, human sperm must undergo a series of morphological and structural alterations, known as capacitation. It has been shown that the production of endogenous sperm reactive oxygen species (ROS) plays a key role in causing cells to undergo a massive acrosome reaction (AR). Astaxanthin (Asta), a photo-protective red pigment belonging to the carotenoid family, is recognized as having anti-oxidant, anti-cancer, anti-diabetic and anti-inflammatory properties and is present in many dietary supplements. This study evaluates the effect of Asta in a capacitating buffer which induces low ROS production and low percentages of acrosome-reacted cells (ARC). Sperm cells were incubated in the presence or absence of increasing concentrations of Asta or diamide (Diam) and analyzed for their ROS production, Tyr-phosphorylation (Tyr-P) pattern and percentages of ARC and non-viable cells (NVC). Results show that Asta ameliorated both sperm head Tyr-P and ARC values without affecting the ROS generation curve, whereas Diam succeeded in enhancing the Tyr-P level but only of the flagellum without increasing ARC values. It is suggested that Asta can be inserted in the membrane and therefore create capacitation-like membrane alteration which allow Tyr-P of the head. Once this has occurred, AR can take place and involves a higher numbers of cells. 相似文献
Canine cognitive dysfunction syndrome (CDS) is a neurodegenerative disorder of aged dogs characterized by a progressive decline in cognitive function. In humans and laboratory animals, a variety of neurotransmitter abnormalities have been described in patients affected by age-related dementia. Specifically, the regulatory role of the catecholaminergic, serotonergic, and cholinergic systems has been outlined. The aim of the present study was to measure blood monoamine levels, platelet α2-adrenergic receptors, and lymphocyte muscarinic receptors in healthy adult and aged dogs and in dogs affected by canine cognitive dysfunction. Based on clinical and behavioral examination, 40 dogs were divided into 3 groups: healthy adults (n = 14), aged dogs (n = 17), and aged dogs affected by canine cognitive dysfunction (n = 9). A significant reduction in plasma levels of norepinephrine and dopamine was observed both in aged dogs (0.16 ± 0.02 ng/mL, P < 0.01; 0.11 ± 02 ng/mL, P < 0.01, respectively) and in CDS dogs (0.14 ± 0.03 ng/mL, P < 0.05; 0.10 ± 00.005 ng/mL, P < 0.01, respectively) compared with adults (0.29 ± 0.04 ng/mL and 0.15 ± 0.02 ng/mL, respectively). No significant differences were observed among groups for α2-adrenergic receptor concentrations. Canine lymphocytes express 2 distinct classes of muscarinic receptors, characterized by high (HA) and low affinity (LA) for [3H]-N-methyl-scopolamine. A significant age-dependent decrease in HA muscarinic receptors was observed. However, no differences were found between aged dogs (87.65 ± 11.08 sites/cell × 102) and in CDS dogs (90.17 ± 6.75 sites/cell × 102 ) for HA muscarinic receptor concentrations. As far as LA muscarinic receptors are concerned, CDS dogs showed a significant increase (393.48 ± 63 sites/cell × 102; P < 0.05) with respect to healthy adult dogs (188.84 ± 16.50 sites/cell × 102). Our results suggest that the reduction in HA muscarinic receptor-binding sites could be representative of the physiological aging process, whereas the increase in lymphocyte LA muscarinic receptor levels could be related to the cognitive decline. 相似文献
The sorption of four endocrine disruptors, bisphenol A (BPA), estrone (E1), 17β-estradiol (E2), and 17α-ethinylestradiol (EE2)
in tropical sediment samples was studied in batch mode under different conditions of pH, time, and sediment amount. Data obtained
from sorption experiments using the endocrine disruptors (EDs) and sediments containing different amounts of organic matter
showed that there was a greater interaction between the EDs and organic matter (OM) present in the sediment, particularly
at lower pH values. The pseudosecond order kinetics model successfully explained the interaction between the EDs and the sediment
samples. The theoretical and experimentally obtained qe values were similar, and k values were smaller for higher SOM contents. The kF values, obtained from the Freundlich isotherms, varied in the ranges 4.2–7.4 × 10−2 (higher OM sediment sample, S2) and 1.7 × 10−3–3.1 × 10−2 (lower OM sediment sample, S1), the latter case indicating an interaction with the sediment that increased in the order: EE2 > > E2 > E1 > BPA. These results
demonstrate that the availability of endocrine disruptors may be directly related to the presence of organic material in sediment
samples. Studies of this kind provide an important means of understanding the mobility, transport, and/or reactivity of this

Habitat loss and fragmentation promote relatively predicable shifts in the functional signature of tropical forest tree assemblages, but the full extent of cascading effects to biodiversity persistence remains poorly understood. Here we test the hypotheses that habitat fragmentation (a) alters the relative contribution of tree species exhibiting different reproductive traits; (b) reduces the diversity of pollination systems; and (c) facilitates the functional convergence of reproductive traits between edge-affected and early-secondary forest habitats (5-32 years old). This study was carried out in a severely fragmented 670-km2 forest landscape of the Atlantic forest of northeastern Brazil. We assigned 35 categories of reproductive traits to 3552 trees (DBH ? 10 cm) belonging to 179 species, which described their pollination system, floral biology, and sexual system. Trait abundance was calculated for 55 plots of 0.1 ha across four habitats: forest edges, small forest fragments (3.4-83.6 ha), second-growth patches, and core tracts of forest interior within the largest available primary forest fragment (3500 ha) in the region. Edge-affected and secondary habitats showed a species-poor assemblage of trees exhibiting particular pollination systems, a reduced diversity of pollination systems, a higher abundance of reproductive traits associated with pollination by generalist diurnal vectors, and an elevated abundance of hermaphroditic trees. As expected, the reproductive signature of tree assemblages in forest edges and small fragments (edge-affected habitats), which was very similar to that of early second-growth patches, was greatly affected by both habitat type and plot distance to the nearest forest edge. In hyper-fragmented Atlantic forest landscapes, we predict that narrow forest corridors and small fragments will become increasingly dominated by edge-affected habitats that can no longer retain the full complement of tree life-history diversity and its attendant mutualists. 相似文献
