A 4-base pair deletion in the neuronal serine protease neurotrypsin gene was associated with autosomal recessive nonsyndromic mental retardation (MR). In situ hybridization experiments on human fetal brains showed that neurotrypsin was highly expressed in brain structures involved in learning and memory. Immuno-electron microscopy on adult human brain sections revealed that neurotrypsin is located in presynaptic nerve endings, particularly over the presynaptic membrane lining the synaptic cleft. These findings suggest that neurotrypsin-mediated proteolysis is required for normal synaptic function and suggest potential insights into the pathophysiological bases of mental retardation.  相似文献

Context

Insectivorous birds are sensitive to forest disturbances that may limit the availability of food consisting mainly of invertebrates. However, birds and invertebrates may be differently affected by forest disturbances while invertebrates may interact with disturbances.

Objectives

We aim to determine: (i) the effects of forest degradation on invertebrates and insectivorous birds; (ii) the effect of the availability of invertebrates as a food source on birds; (iii) interactions between food availability and forest degradation.

Methods

We selected 34 1-km radius landscape units, where the abundance of birds and invertebrates was sampled in the canopy and understory. Bird density as well as the abundance and richness of invertebrates were considered as dependent variables and analysed using Generalized Linear Mixed Model and Structural Equation Models. Remote-sensing indices of forest degradation were included as predictors.

Results

Eight indices of forest degradation affected canopy and understory invertebrates differently. Unlike invertebrates, bird abundance was affected by a smaller number of degradation indices, forest amounts as well as the cover of understory and canopy. Only two forest degradation indices had a comparable effect on bird abundance and invertebrates. We found causal relationships between understory invertebrates and the abundance of understory birds (all species and the small-sized ones), but also invertebrate abundance × forest cover interactions affected the abundance of a bird species.

Conclusions

Our results indicate that birds and invertebrates respond differently to forest degradation, but also provide evidence for bottom-up control by forest degradation and suggest food limitation varies with forest amounts.

The atmosphere over Brazilian cities is influenced by a variety of emissions sources. In this study, aerosol collection and back-trajectory analysis were used to determine the influence of local and remote sources. Aerosols were collected at three locations within the state of São Paulo: one megacity and two cities in which sugar cane burning in the surroundings is observed. We quantified the major water-soluble inorganic ions and trace metals. As expected, vehicle emissions influenced the atmosphere of the megacity heavily, and sugar cane burning influenced that of the other locations. During the period of this experiment the back-trajectory analysis revealed that air masses are transported into the state from the northeast of Brazil, where biomass burning occurs. Multivariate statistical analysis revealed that the two principal components account for 48.5% of the total data variance. We conclude that local sources have a strong impact on the concentrations of particulate matter and pollutants. Remote sources also contribute to the concentrations of aerosol pollutants.  相似文献

A total of 249 client-owned dogs with osteoarthritis were treated with firocoxib (5 mg/kg/day) or a positive control, etodolac (10-15 mg/kg/day), for 30 days. Veterinary examinations were performed on approximately days 0 (visit 1), 14 (visit 2), and 29 (visit 3). Based on defined noninferiority criteria, firocoxib and etodolac were comparable. Based on the magnitude of the change from baseline, improvement with firocoxib was significantly greater than with etodolac for lameness at a trot (visits 2 and 3) and for lameness at a walk, pain on manipulation, and range of motion (visit 3) (P < .05). In weekly owner evaluations, firocoxib provided significantly greater improvement than etodolac (P < .05) at each scoring.  相似文献

This study investigated epigenetic mechanisms by which DNA methylation affects the function of bovine adaptive immune system cells, particularly during the peripartum period, when shifts in type 1 and type 2 immune response (IR) biases are thought to occur. Stimulation of CD4+ T-lymphocytes isolated from 5 Holstein dairy cows before and after parturition with concanavalin A (ConA) and stimulation of CD4+ T-lymphocytes isolated from 3 Holstein dairy cows in mid-lactation with ConA alone or ConA plus dexamethasone (Dex) had significant effects on production of the cytokines interferon gamma (IFN-γ, type 1) and interleukin 4 (IL-4, type 2) that were consistent with DNA methylation profiles of the IFN-γ gene promoter region but not consistent for the IL-4 promoter region. ConA stimulation increased the production of both cytokines before and after parturition. It decreased DNA methylation in the IFN-γ promoter region but increased for IL-4 promoter region. Parturition was associated with an increase in IFN-γ production in ConA-stimulated cells that approached significance. Overall, DNA methylation in both promoter regions increased between the prepartum and postpartum periods, although this did not correlate with secreted cytokine concentrations. Dexamethasone treated cells acted in a manner consistent with the glucocorticoid’s immunosuppressive activity, which mimicked the change at the IFN-γ promoter region observed during parturition. These results support pregnancy as type 2 IR biased, with increases of IFN-γ occurring after parturition and an increase in IL-4 production before calving. It is likely that these changes may be epigenetically controlled.  相似文献

The rate at which creosote-treated pilings release polycyclic aromatic hydrocarbons (PAHs) into the environment should diminish with structure age and weathering, and so, it may be assumed that PAH concentrations in the vicinity of old structures (>30 years) may approach background levels. However, this may not be true in cold environments where PAH release and degradation rates are slow and where pilings continue to experience significant physical damage. Moreover, PAHs will remain high in the vicinity of pilings if current and wave action is insufficient to disperse and dilute PAHs over a wider area and/or where PAHs do not become diluted and buried by uncontaminated sediments. This is demonstrated in our investigation of the sediments of the Grey Owl Marina, located in Prince Albert National Park, in central Saskatchewan, Canada. The marina, constructed in the early 1960s, consists of six piers and is protected from strong wave action by a breakwater. PAH concentrations were high in sediments collected under the piers and inside the boat slips, exceeding probable effect levels for several compounds. Various lines of physical and chemical evidence pointed to creosote as being the primary PAH source with a mixture of relatively undegraded and weathered PAHs being present. PAH concentrations decreased rapidly 2 m and further away from the pilings as a result of dilution with increasing dispersal over a broadening area. There was evidence of benthic community impairment, with total abundances negatively correlated with PAH concentrations. According to the Methods for Ranking Contaminated Aquatic Sites on Canadian Federal Properties, areas with the highest hazard scores were under the piers and inside the boat slips, while areas with the lowest hazard scores were >10 m from the pilings. Therefore, remedial actions based on piling and contaminated sediment removal may need to be conducted over only a small area, i.e., within 2 m of the pilings.  相似文献

Sampling at blueberry farms found Botryosphaeriaceae fungi in five of seven farms sampled; overall incidence was 41.4%, with Neofusicoccum australe (79.0%), N. luteum (8.0%), N. ribis (8.0%) and N. parvum (5.0%). Sampling of nursery plants found infections in all four nurseries with 45% incidence in mainly asymptomatic plants, which were infected with N. australe (66.0%), N. parvum (31.5%) and N. ribis (2.5%). Asymptomatic propagation cuttings from one nursery were found to have external contamination of Botryosphaeriaceae DNA (90.0%) and internal infection (65.0%) by the four main species found in the blueberry farms and nurseries. For propagation media nested PCR showed that out of the 98 samples received, 43 samples were positive for the presence of Botryosphaeriaceae DNA (44.0%). Results from the SSCP indicated that N. australe, N. luteum, N. parvum/ N. ribis and Diplodia mutila were present in the propagation media received. Isolates of the four main species recovered from farms and nurseries were pathogenic on blueberry stems but pathogenicity differed significantly between species and isolates within a species, with N. ribis being the most pathogenic, then N. parvum, N. luteum and N. australe. Overall, the high rate of infection in nursery plants indicated that nurseries can be a major source of infection for blueberry farms. Since propagating cuttings are the likely sources of infection for nurseries, these should be targeted in the control strategies.  相似文献

We present observations on an unusual tetratrichomonad species isolated from preputial smegma of virgin bulls. Ultrastructural studies were performed using scanning and electron microscopy techniques. This protozoan presents four anterior flagella of unequal length and a recurrent one forming the undulating membrane. It shows one anterior nucleus, a Golgi complex, an axostyle, and a costa. The hydrogenosomes are rather elongated, seen in groups, and presenting different electron densities. Vacuoles of different sizes containing bacteria and material in process of digestion were frequently found. PCR was also used in order to compare the species herein described with other trichomonad species. The amplification products were seen only with primers TFR1 and TFR2 (specific to trichomonads), but not with TFR3 and TFR4 (specific to Tritrichomonas foetus), suggesting that although collected from the genital tract of the bull, this protist was not T. foetus. We propose that the appearance of these tetratrichomonads were probably due to the sodomy practiced among bulls. Concomitant contamination of preputial cavity with feces could explain the presence of the opportunistic organism. The observations presented here show the importance of the correct diagnostic when investigating samples obtained from the urogenital tract of cattle. We also suggest that this flagellate belongs to the species Tetratrichomonas buttreyi.  相似文献

Twenty-four client-owned dogs with histologically diagnosed appendicular osteosarcoma (OSA) and no evidence of gross metastatic disease were treated with amputation or limb salvage followed by combination chemotherapy consisting of carboplatin (175mg/ m2 IV, day 1) and doxorubicin (15 mg/m2 IV, day 2) given on a 21-day cycle for a maximum of 4 cycles. Hematologic and gastrointestinal adverse effects were graded according to National Cancer Institute guidelines. Thoracic radiographs were obtained before the 3rd chemotherapy cycle and then every 2 months. Median disease-free interval was 195 days (95% confidence interval 111-228 days) and median survival was 235 days (95% confidence interval 150-283 days). Two patients required dose reductions: 1 for grade 3 thrombocytopenia and 1 for grade 3 adverse gastrointestinal effects. Patients with a longer duration of clinical signs before definitive diagnosis and surgery (greater than 30 days) were more likely to develop progressive disease and to die or be euthanized because of progressive disease on any day; hazard ratios were 3.0 (P = .02) and 3.7 (P .02), respectively. In conclusion, although this combination chemotherapy protocol was well tolerated, it did not provide any improvement over historical single-agent protocols.  相似文献
