A quantitative and confirmatory method for the analysis of trinexapac (free acid metabolite of trinexapac-ethyl) in wheat is described. Residues were extracted from wheat with acetonitrile in aqueous phosphate buffer (pH 7) overnight. The extract was directly injected into the HPLC system. Chromatographic separation was achieved on an octadecylsilica column, and detection was performed by negative ion electrospray ionization tandem mass spectrometry. The precursor ion of trinexapac [M - H](-) at m/z 223 was subjected to collisional fragmentation with argon to yield two intense diagnostic product ions at m/z 135 and 179, respectively. Accuracy and specificity for routine analysis of trinexapac were demonstrated. The validated concentration range was 10-200 microg/kg based on a 0.10 g/mL wheat sample extract. Recoveries were within the range of 71-94%, with associated relative standard deviations better than 10%. The limit of detection for trinexapac in wheat was estimated at 5 microg/kg. The method has been applied to a survey of 100 samples of wheat. In 46% of the samples analyzed, a quantifiable amount of trinexapac was detected, ranging from 10 to 110 microg/kg. It has been demonstrated that analyses of trinexapac accurately reflect the total amount of residues of the plant growth regulator, trinexapac-ethyl, in the wheat samples following field application. The aim of this research was to assess the antimicrobial activity of nine different industrial essences used in a soft drink factory in relation to their composition, as well as to verify the role of vapor pressure on their bioactivity. The essences were tested against a Saccharomyces cerevisiae strain isolated from spoiled soft drinks. The tests were carried out by adding the essences directly to a liquid medium or into the headspace of closed systems inoculated with the yeast. The headspace composition was evaluated through a solid phase microextraction-gas chromatography technique. The use of a mass spectrometer allowed the identification of the peaks detected. The microbial growth was indirectly monitored by measuring the metabolic CO2 released by the yeast. The results obtained indicated that the most effective essences were characterized by the highest concentration of some terpenes, such as citral, beta-pinene, and p-cymene. A powder of quality elk velvet antler (QEVA) was evaluated on client-owned dogs with osteoarthrosis (OA) in a clinical, double-blind, and placebo-controlled study. Thirteen dogs received a placebo for 30 days and then QEVA for 60 days. Twenty-five other dogs received QEVA for 60 days. Gait analysis measured with a force plate, clinical signs assessed by an orthopedic surgeon, performances in daily life activities and vitality assessed by the owners, and complete blood analyses were obtained at days 0, after 30 days of placebo and/or 60 days of QEVA. On placebo, the 13 dogs did not show significant improvement (P < 0.05); however, their gait, their performances in daily life activities, and their vitality were significantly improved on QEVA, based on changes in values exceeding those observed when placebo was administered. The 25 dogs on QEVA for 60 days showed similar improvements. No clinical changes were revealed on blood analyses. Administration of QEVA was effective in alleviating the condition in arthritic dogs. 相似文献

BACKGROUND: An important part of the production area of common bean (Phaseolus vulgaris L.) in Belgium is located on the sandy soils of the provinces of Antwerp and Limburg where Meloidogyne chitwoodi (Golden), M. fallax (Karssen) and M. hapla (Chitwood) are present. The host plant status of ten bean cultivars for root‐knot nematodes was determined by evaluating penetration, development and egg mass formation after inoculation with second‐stage juveniles. RESULTS: The tested cultivars were poor to good hosts for M. chitwoodi, non‐hosts or bad hosts for M. fallax and excellent hosts for M. hapla. Significantly fewer M. fallax were found in the roots, and their development was delayed. Penetration of M. hapla took place over a longer period than that of M. chitwoodi and M. fallax. The number of mature females of M. chitwoodi in cv. Polder 6 weeks after inoculation was no different from that in other cultivars, although fewer egg masses were found on this cultivar in the screening test. There was no influence of M. chitwoodi on vegetative growth of cv. Polder. CONCLUSION: The differences found in host plant status of bean cultivars stress the importance of a correct diagnosis of the Meloidogyne species in agricultural fields. 结合形态与分子生物学特征对从河北省分离的40个异小杆类昆虫病原线虫种群进行鉴定,其主要形态特征(4个典型种群平均值):侵染性幼虫体长=550(520—610)μm,体宽=22.2(20—24)μm,尾长=85,7(80-88)μm,E%=112.5%(105%-119%);雄虫交合刺长度=36,8(33,75—39,0)μm,引带长度=17,9(16.3—20.0)μm,GS%=0,49(0.41—0.59),SW=1,25(1.15—1,43),与嗜菌异小杆线虫Heterorhabditis bacteriophora形态特征相近。经对其中二个典型种群的ITS-rDNA测序,其ITS1-rDNA序列与已知种类H.bacteriophora的ITS1-rDNA序列相差小于1,2%,和H.argentinensis的差异小于1,5%,而与其他种类的差异则高达18.7%以上。综合其形态与分子特征,确定河北省的异小杆类线虫为嗜菌异小杆线虫。 相似文献

Streptobacillus moniliformis is a zoonotic bacterium. We obtained positive S. moniliformis PCR results in oral swab samples from guinea pigs from an experimental colony and the breeding colony of origin. Comparison of the DNA sequence of an amplicon with deposited 16S rDNA sequences revealed that Leptotrichia sp. can be the source of a false positive S. moniliformis PCR outcome. 相似文献
