In previous studies, it has not been reported that protein isolated from chia interferes favorably with antibacterial activity, and reduces cholesterol synthesis. The objective of this study was to determine whether commonly used commercial microbial proteases can be utilized to generate chia protein-based antibacterial and hypocholesterolemic hydrolysates/peptides, considering the effects of protein extraction method. Alcalase, Flavourzyme and sequential Alcalase-Flavourzyme were used to produce hydrolysates from chia protein (CF), protein-rich fraction (PRF) and chia protein concentrates (CPC1 and CPC2). These hydrolysates were evaluated for their antimicrobial activity against Gram-positive (G+) and Gram-negative (G?) microorganisms. The protein hydrolysates were purified by ultrafiltration through a membrane with 3 kDa nominal molecular weight, for evaluation of hypocholesterolemic activity. An inhibition zone was observed when the hydrolysate was tested against S. aureus, and minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) values were obtained. Peptides from chia protein with molecular mass lower than 3 kDa reduced up to 80.7% of 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMG-CoA reductase) enzymatic reaction velocity. It was also observed that, independent of the method used to obtain chia proteins, the fractions showed relevant bioactivity. Moreover, the intensity of the bioactivity varied with the method for obtaining the protein and with the enzyme used in the hydrolysis process. This is the first report to demonstrate that chia peptides are able to inhibit cholesterol homeostasis.  相似文献

Pratylenchus brachyurus is one of the main limiting factors of soybean yield in Brazil, particularly because of inefficiency of the control methods when used individually. The present study aimed to assess the effect of associated methods, using seed treatment with nematicides (ST) and resistance inducers (RI), on nematode control in soybean plant in both field and greenhouse conditions. A field assay was conducted in an infested field and nematode population was assessed at sowing, 45, 75, and 100 days after sowing and the yield measured at end of crop cycle. The experiment was repeated in greenhouse. In another experiment, that was conducted in two different periods in a greenhouse, seed treatments and resistance inducers, alone or combined, were assessed under two initial populations of P. brachyurus (low IP = 500 specimens and high IP = 2000 specimens). The treatments did not reduce the number of nematodes g?1 of root in field assay, but all seed treatments effectively controlled nematode population in the greenhouse assay. Most treatments reduced the number of nematodes g?1 of root when nematode initial population was low (IP = 500) but when initial nematode population was high (IP = 2000) combinations of treatments which includes abamectin inhibited P. brachyurus reproduction. Chemical products did not affect yield but acibenzolar-S-methyl, alone or associated with other products, generally inhibit plant growth.  相似文献

Most technologies used for decontamination presents good results for high concentrations, but limitations for lower ones. The desirable Hg concentration in the water is extremely low because of its toxicity. The aims of this study were to evaluate inorganic mercury (Hg2+) and methylmercury (CH3Hg+) toxicity in Nostoc paludosum, to assess the potential of this cyanobacteria strain to remove these Hg species from aqueous medium and also to investigate Hg methylation by the cyanobacteria. CH3Hg+ determination was performed by gas chromatography-pyrolysis-atomic fluorescence spectrometry in cultures exposed to a concentration of 20 μg L?1 for 30 days. Both Hg species were removed from the supernatant, ranging from 73 to 96% of Hg2+ and from 73 to 95% of CH3Hg+. Ultrastructural Hg2+ effects in the cyanobacteria cells investigated by transmission electron microscopy revealed higher production of glycogen, cyanophycin, and intrathylacoidal spaces than the control group. When Hg was added to the culture in the form of CH3Hg+, a decrease corresponding to approximately 60% of the initial concentration due to Hg volatilization was observed. The production of CH3Hg+ by the cyanobacteria was detected in concentrations near the limit of detection (0.0025%) of the bioaccumulated THg. This is an advantage for biotechnological decontamination applications, as CH3Hg+ is a very toxic specie and can be bioaccumulated and biomagnified. The results demonstrated that cyanobacteria cells are an efficient alternative to retain and/or remove Hg at low concentrations and they constitute a potential tool for a “final cleaning” of contaminated waste water.  相似文献

This study evaluated the effect of the protected fatty acid inclusion during estrus synchronization on reproductive parameters. Goats (n = 32) received progestagen sponges for 6 days and 200 IU equine chorionic gonadotropin and 30 µg d‐cloprostenol were given on Day 5. No difference was found among control (C), 1% protected fatty acid inclusion (C + 1%) or 4% protected fatty acid inclusion (C + 4%) groups, respectively, in estrus (100.0, 100.0 or 90.9%), estrus duration (31.6 ± 12.3; 43.2 ± 12.9 or 40.8 ± 14.1 h), animals ovulating (100.0, 90.0 or 100.0%) or ovulation rate (1.3 ± 0.5; 1.1 ± 0.3 or 1.2 ± 0.4). The interval from sponge removal to ovulation and from estrus to ovulation, respectively, were shorter for C + 4% (45.2 ± 8.0 h; 18.3 ± 11.0 h) compared with C (56.3 ± 12.6 h; 30.6 ± 10.5 h) or C + 1% (57.7 ± 8.7 h; 30.3 ± 11.1 h). The average ovulatory follicle diameter was smaller for C + 4% (6.2 ± 0.7 mm) than C (7.5 ± 0.8 mm), but similar to C + 1% (7.0 ± 1.5 mm). Insulin, insulin‐like growth factor 1, glucose and progesterone concentrations were similar among groups. The inclusion of protected fatty acid during synchronization treatment promoted no benefits on ovulation rate, but 4% anticipated the ovulation time.  相似文献

Sixty clinically normal German shepherd dogs, 31 males and 29 females, ranging in age from 1 and 5 years and with a body weight ranging from 22 to 37.2 kg, were examined by the two-dimensional mode, M-mode, and Doppler echocardiography. In Doppler mode, the mitral valve flows were obtained, where the aim was to determine the velocity peaks and ratios of the E and A waves and the mitral E wave deceleration time. The velocity peaks were obtained for the tricuspid, pulmonary and aortic valves. On the left ventricular outflow tract flow, the time velocity integral and aortic cross-sectional area was used to calculate the cardiac output. A statistically significant correlation with the body weight was found for the systolic left atrial and diastolic aortic diameter on two-dimensional mode. On M-mode, there was a significant correlation between the body weight and the systolic left atrium and diastolic aortic dimension, systolic and diastolic left ventricular, septal and posterior wall dimensions. Doppler echocardiography showed that there was no significant correlation between the body weight and the mitral, tricuspid, pulmonary and aortic valves flows. These results demonstrate that it is important to know the normal echocardiographic values for German shepherd dogs because there are some characteristics peculiar to this particular breed. The data obtained is expected to be helpful for studies on small animal cardiology.  相似文献

This study was carried out to evaluate the effects of feed restriction on the performance and feeding behaviour of Brycon orbignyanus. Ten‐day larvae (72.84 ± 8.61 mg) were distributed in 36 aquariums (density of 15 larvae/L) in a completely randomized design consisting of six treatments and six replicates. The treatments were determined according to the number of feed restriction days: 0, 2, 4, 6, 8 and 10 days (F0, F2, F4, F6, F8 and F10, respectively), totalling 10 days. The results for final weight, weight gain and specific growth rate were significant (p < .05), and F2 and F0 presented the highest values among all experimental groups. The adopted feeding strategies did not influence final survival (p > .05), and no cannibalism behaviour was observed in any of the treatments. F2 and F0 treatments also displayed higher swimming speed (p < .05) among the assessed groups. The time spent searching for and capturing prey was significantly lower in the F0 treatment. Muscular atrophy for the F10 group was observed, and F2 animals presented no significant difference (p > .05) in muscle fibre diameter when compared to F0. Thus, dietary restrictions can be applied as a strategy for B. orbignyanus reared in intensive laboratory systems for 2 days without compromising performance, muscle growth and feeding behaviour.  相似文献
