竹类植物在中国有着非常悠久的栽培历史,并被广泛应用于园林、造林、庭院以及食品等行业。本研究以翠竹和菲白竹为材料提取其叶片总DNA,采用PCR方法获得翠竹SpLEA3基因与菲白竹SfLEA3-1、SfLEA3-2基因;其中SpLEA3全长804 bp,编码195个氨基酸,GC含量为68.4%;SfLEA3-1全长803 bp,编码195个氨基酸,GC含量为68.2%;SfLEA3-2全长557 bp,编码144个氨基酸,GC含量为67.8%。通过ProtParamy等生物软件分析,SpLEA3、SfLEA3-1和SfLEA3-2与贵州悬竹LEA3基因同源性高达61%以上,且3个基因编码蛋白均属于亲水性蛋白。源自2个竹种LEA3基因均包含一个完整的开放阅读框,其编码的氨基酸含有4~6个由11个氨基酸组成的保守基元序列。本研究不仅为深入了解竹类植物抗旱的分子机理研究提供了基础数据,也为竹类植物的抗旱育种后续研究提供了科学依据。 相似文献

爆裂玉米3个膨爆特性的非条件和条件QTL分析 总被引:2,自引:0,他引:2
膨化倍数(PF)、膨化体积(PV)和爆花率(PR)是爆裂玉米的主要品质指标。本研究以普通玉米自交系丹232和爆裂玉米自交系N04杂交构建的259个F2:3家系为定位群体,采用完全随机区组设计在郑州夏播条件下测定了3个膨爆特性指标。利用覆盖玉米10条染色体的183对多态性分子标记构建连锁图,采用复合区间作图(CIM)方法对各性状进行非条件QTL定位分析,采用条件遗传统计软件对PF进行条件QTL分析。3个膨爆特性指标非条件QTL定位共检测出31个QTL,单个QTL的贡献率为3.09% ̄12.46%,累计贡献率为51.90%、77.37%和61.45%。在8个标记区间(占42.11%)同时检测到控制2 ̄3个性状的QTL。加性和部分显性在膨爆特性的遗传中起主要作用,同时存在显性和超显性基因效应。条件QTL定位结果表明,PV和PR以不同方式显著影响PF的QTL表达,PV比PR更重要。 相似文献

为明确菊酯类杀虫剂对昆虫线粒体膜流动性的影响,以1,6-二苯基-1,3,5-己三烯(1,6-diphenyl-1,3,5-hexatriene,DPH)为荧光探剂,采用荧光偏振法研究了4种菊酯类杀虫剂对枸杞蚜虫Aphis sp.线粒体膜流动性的影响。结果表明,4种菊酯类杀虫剂对枸杞蚜虫线粒体膜流动性均有显著影响,其荧光偏振度显著增大,线粒体膜流动性显著减小。当药剂浓度为100 μmol/L时,对照组枸杞蚜虫线粒体膜的荧光偏振度为0.239,高效氯氰菊酯的影响最大,线粒体膜的荧光偏振度为0.315,联苯菊酯的影响最小,线粒体膜的荧光偏振度为0.287。4种菊酯类杀虫剂对线粒体膜流动性的影响都存在一定的浓度效应,当药剂浓度在0~100 μmol/L范围内,浓度越大影响越大。4种菊酯类杀虫剂在不同温度下对枸杞蚜虫线粒体膜流动性的影响不同,高效氯氰菊酯、联苯菊酯和甲氰菊酯对线粒体膜流动性的影响基本不受温度影响,而氯菊酯在高温条件下产生的影响大,在低温条件下产生的影响小,表现为明显的正温度系数。表明DPH可以作为荧光探剂研究枸杞蚜虫线粒体膜的流动性。 相似文献

An 8-week feeding trial was conducted to evaluate the effect of N-acetyl cysteine (NAC) and glycine supplementation on growth performance, glutathione (GSH) synthesis, and antioxidative ability of grass carp, Ctenopharyngodon idella. Four practical diets were formulated: control, control + 0.2% NAC, control + 0.5% glycine, and control + 0.2% NAC + 0.5% glycine. Each diet was randomly assigned to quadruplicate groups of 30 fish (approximately 8.8 g). Weight gain and specific growth rate were significantly increased with the supplementation of NAC. Supplementation of NAC plus glycine significantly increased the feed efficiency. Glutathione peroxidase (GPx) and γ-glutamine cysteine synthase (γ-GCS) in plasma were significantly increased with the supplementation of NAC plus glycine. GSH in plasma increased and malondialdehyde (MDA) decreased in fish fed diets supplemented with NAC. Respiratory burst, superoxide dismutase (SOD), and catalase (CAT) activity were not affected by NAC or glycine. These results clearly indicated that NAC improved the growth performance and restored GSH of grass carp, supplemented NAC together with glycine enhanced GSH synthesis, and improved the antioxidative ability of grass carp. 相似文献

An 8‐week feeding trial was conducted to evaluate the effects of ascorbic acid (AsA), in the form of l ‐ascorbyl‐2‐polyphosphate (LAPP) on growth performance, body composition, antioxidative capacity and salinity stress tolerance of juvenile Pacific white shrimp, Litopenaeus vannamei. Five practical diets (46% crude protein and 7.6% lipid) supplemented with graded levels of AsA (14.64, 48.55, 84.98, 308.36 and 639.27 mg kg?1 diet) were fed to five replicate groups of L. vannamei (mean initial wet weight 0.57 g). No significant differences were found on growth performance among all treatments. However, whole body lipid content significantly decreased with dietary AsA levels increasing. Activities of total antioxidant capacity, glutathione reductase and glutathione peroxidase were significantly affected by dietary AsA levels. Shrimp fed LAPP‐free diet had higher malondialdehyde content than those fed the diets supplemented with LAPP. Dietary AsA levels higher than 308.36 mg kg?1 diet increased the survival of shrimps after 1, 2 and 3 h of acute salinity change. Broken‐line regression analysis on survival after 3 h of salinity stress and second‐degree polynomial regression analysis on glutathione reductase data indicated that the optimal dietary AsA requirement of L. vannamei was estimated to be 306.39, 319.75 mg kg?1 diet respectively. 相似文献

Edwardsiella tarda is a pathogen that causes edwardsiellosis in aquatic animals. The emergence of multiple antibiotic‐resistant strains makes antibiotic treatment difficult. This study aimed to investigate the antibiotic susceptibility patterns and the genotypic characterization of E. tarda isolated from cage‐cultured red tilapia in Thailand. A total of 30 isolates were identified as E. tarda using biochemical and molecular analysis. The disc diffusion method for testing antibiotic susceptibility showed all the isolates were resistant to colistin sulphate and oxolinic acid. High levels of resistance to amoxicillin, ampicillin, ceftazidime, oxytetracycline and sulphamethoxazole/trimethoprim were observed as well. The multiple antibiotic resistance index ranged from 0.25 to 0.92, indicating that these isolates had been exposed to high risk sources of contamination where antibiotics were commonly used. All the isolates carried the blaTEM gene based on polymerase chain reaction (PCR). The tetA and sul3 genes were detected in 90% (27/30) and 26.7% (8/30) of the isolates respectively. Nine different genetic groups of isolates were obtained using enterobacterial repetitive intergenic consensus polymerase chain reaction (ERIC‐PCR). A correlation between genetic types and multiple antibiotic‐resistant patterns was found.
