The pathogenicity-related traits of biotrophic plant pathogens are usually measured on the individual host plant, at the scale of a single pathogen life cycle, whereas epidemic development in the field encompasses a succession of cycles. It remains unclear which traits make the greatest contribution to pathogen fitness in the field and to epidemic severity. The objective of this study was to determine the contributions of elementary pathogenicity traits to epidemic development in field conditions. We challenged a set of wheat cultivars with three different leaf rust isolates, under both controlled and field conditions, in 3 consecutive years. Infection efficiency, latent period, lesion size, spore production per lesion and spore production capacity were measured in the greenhouse, whereas disease severity was measured in the field. Most, but not all, of the pathogenicity traits were related to each other. All traits contributed to epidemic development in the field, but to different extents. Surprisingly, lesion size and spore production per lesion were inversely correlated with epidemic severity. Conversely, there was a strong positive correlation between spore production capacity and pathogen fitness in the field, in accordance with the concept of propagule pressure as a strong determinant of invasion success. Severe epidemics were mostly associated with small lesions with a high spore production capacity, high infection efficiency and a short latent period.  相似文献

This study reports a comparative analysis of soil enzyme activities (β-glucosidase, protease, urease, arylsulphatase, phosphatase and fluorescein diacetate hydrolase), ATP, total N and organic matter contents in three vegetal successional stages (meadow, low shrubland and high maquis) of a Mediterranean ecosystem in the Natural Reserve of Castel Volturno (Campania, Italy). Because water availability is a major limiting factor of soil microbial activity in Mediterranean ecosystems, the analysis was performed in late spring (May), after the rainy period, and in early autumn (October), after the long dry summer.A significant decrease in protease, arylsulphatase, urease and β-glucosidase activities was observed in meadow soil in the autumn sampling, probably due to the prolonged summer drought. Combining the values measured in the two sampling dates, the high maquis tended to have higher levels of enzymes activities than shrubland and meadow. Notably, high maquis had significantly higher phosphatase and arylsulphatase activities than shrubland and meadow and, in addition, a higher ATP content compared to meadow. Drastic changes were observed in EA/ATP ratios between the sampling periods in the meadow and shrubland, suggesting changes in the efficiency of microbial community more likely linked to climatic fluctuations than to the successional stage. The more stable EA/ATP ratio in the maquis probably reflects a constant contribution of microbial biomass to enzyme secretion.In conclusion, our results point to an increase in soil microbial activity accompanying the succession from meadow to high maquis that probably reflects a parallel increase in soil functions. Nevertheless, spatial heterogeneity and, more important, temporal variations in soil activities often may obscure differences related to the plant cover type.  相似文献

Neurodegenerative diseases are characterised by selective damage to specific neurons in the nervous system. Interest in such diseases in humans has resulted in considerable progress in the molecular understanding of these disorders in recent decades. Numerous neurodegenerative diseases have also been described in domestic animals but relatively little molecular work has been reported. In the present review, we have classified neurodegenerative disease according to neuroanatomical criteria. We have established two large groups, based on whether the neuronal cell body or its axon was primarily affected. Conditions such as motor neuron diseases, cerebellar degenerations and neuroaxonal dystrophies are discussed in terms of their clinical and neuropathological features. In the most studied disorders, we also present what is known about underlying pathomechanisms, and compare them with their human counterparts. The purpose of this review is to re-kindle interest in this group of diseases and to encourage veterinary researchers to investigate molecular mechanisms by taking advantage of current diagnostic tools.  相似文献

In this paper, based on the analysis of a long-term energy balance monitoring programme, a Bowen ratio-based method (BR) was proposed to resolve the lack of closure of the eddy covariance technique to obtain reliable sensible (H) and latent heat fluxes (λE). Evapotranspiration (ET) values determined from the BR method (ETc,corr) were compared with the upscaled transpiration data determined by the sap flow heat pulse (HP) technique, evidencing the degree of correspondence between instantaneous transpirational flux at tree level and the micrometeorological measurement of ET at orchard level. Using the BR-corrected λE fluxes, a crop ET model implementing the Penman–Monteith approach, where the canopy surface resistance was determined from standard microclimatic variables, was applied to determine the crop coefficient values. The performance of the model was evaluated by comparing it with the sap flow HP data. The results of the comparison were satisfactory, and therefore, the proposed methodology may be considered valid for characterizing the ET process for orange orchards grown in a Mediterranean climate. By contrast to reports in the FAO 56 paper, the crop growth coefficient of the orange orchard being studied was not constant throughout the growing season.  相似文献

Cooling of equine semen obtained from some stallions results in lower seminal quality and viability when the seminal plasma (SP) is present. The objective of this study was to evaluate the effect of the removal of SP using a Sperm Filter on the viability of cooled stallion semen. For this purpose, 31 stallions were used. Their ejaculates were divided into three groups: CN, semen was diluted with an extender; FLT, SP was removed by filtration; and CT, SP was removed by centrifugation and cooled to 15°C for 24 hours. Sperm kinetics and plasma membrane integrity were evaluated immediately after collection (T0) and after 24 hours of refrigeration (T1). No difference (P > .05) was noted at T1 for total sperm motility (TM), progressive sperm motility, or plasma membrane integrity when semen samples from all the stallions were analyzed. However, when samples from stallions termed “bad coolers” were analyzed (TM = <30% at T1), a difference was observed in TM and progressive sperm motility for CN compared with FLT and CT at T1. Sperm recovery was greater when SP was removed using the filter (FLT) to that when the SP was removed by centrifugation (CN) (89% vs. 81%). Thus, we concluded that filtering with a Sperm Filter is an efficient and practical method for removal of SP from stallion ejaculates, with lower sperm loss than centrifugation. We also found that the presence of SP reduces the quality and viability of cooled semen from stallions whose semen is sensitive to the process of refrigeration.  相似文献

Background, Aim and Scope

Continuous application of pesticides may pollute soils and affect non-target organisms. Soil is a complex ecosystem; its components can modulate the effects of pesticides. Therefore, it is recommended to evaluate the potential environmental risk of these compounds in local conditions. We performed an integrated field-laboratory study on an Argentine soya field sprayed with glyphosate and chlorpyrifos under controlled conditions. Our aim was to compare the sensitivity of a series of endpoints for the assessment of adverse effects of the extensive use of these agrochemicals.

Materials and Methods

A RR soya field in a traditional farming area of Argentina was sprayed with glyphosate (GLY) or chlorpyrifos (CPF) formulations at the commercially recommended rates, according to a randomized complete block design with 3 replicates. In laboratory assays, Eisenia fetida andrei were exposed to soil samples (0–10 cm depth) collected between the rows of soya. Endpoints linked to behavior and biological activity (reproduction, avoidance behavior and bait-lamina tests) and cellular/subcellular assays (Neutral Red Retention Time — NRRT; DNA damage — Comet assay) were tested. Field assays included litterbag and bait-lamina tests. Physico/chemical analyses were performed on soil samples.

Results

GLY reduced cocoon viability, decreasing the number of juveniles. Moreover, earthworms avoided soils treated with GLY. No effects on either reproduction or on avoidance were observed at the very low CPF concentration measured in the soils sampled 10 days after treatment. Both pesticides caused a reduction in the feeding activity under laboratory and field conditions. NRRT was responsive to formulations of CPF and GLY. Comet assay showed significantly increased DNA damage in earthworms exposed to CPF treated soils. No significant differences in DNA migration were observed with GLY treated soils. Litterbag field assay showed no differences between treated and control plots.

Discussion

The ecotoxicological effects of pesticides can be assessed by monitoring the status of communities in real ecosystems or through the use of laboratory toxicity tests. Litterbag field test showed no influence of the treatments on the organic matter breakdown, suggesting a scarce contribution of soil macrofauna. The bait-lamina test, however, seemed to be useful for detecting the effects of GLY and CPF treatments on the activity of the soil fauna. CPF failed to give significant differences with the controls in the reproduction test and the results were not conclusive in the avoidance test. Although the field population density of earthworms could be affected by multiple factors, the effects observed on the reproduction and avoidance tests caused by GLY could contribute to its decrease, with the subsequent loss of their beneficial functions. Biomarkers measuring effects on suborganism level could be useful to predict adverse effects on soil organisms and populations. Among them, NRRT, a lysosomal destabilization biomarker, resulted in demonstrating more sensitivity than the reproduction and avoidance tests. The Comet assay was responsive only to CPF. Since DNA damage can have severe consequences on populations, it could be regarded as an important indicator to be used in the assessment of soil health.

Conclusions

Reproduction and avoidance tests were sensitive indicators of GLY exposure, with the former being more labor intensive. Bait-lamina test was sensitive to both CPF and GLY. NRRT and Comet assays revealed alterations at a subcellular level, and could be considered complementary to the biological activity tests. Because of their simplicity, some of these bioassays seemed to be appropriate pre-screening tests, prior to more extensive and invasive testing.

Recommendations and Perspectives

This study showed deleterious effects of GLY and CPF formulations when applied at the nominal concentrations recommended for soya crops. Further validation is needed before these endpoints could be used as field monitoring tools in Argentine soya soils (ecotoxicological risk assessment — ERA tools).

Cryopreservation causes damage to spermatozoa, and methods minimizing this damage are therefore needed. Although much discussed, seminal plasma removal has become an alternative to improve sperm quality and viability after freezing and has been applied to different species in attempt to obtain good results. The objective of this study was to evaluate semen quality in buffaloes submitted to two methods for seminal plasma removal (filtration and centrifugation). Semen samples were collected from seven Murrah buffalo bulls (Bubalus bubalis) once a week for 8 weeks. Each ejaculate was divided into three groups: control (presence of seminal plasma), centrifugation and filtration. Sperm kinetics was evaluated with the computer‐assisted sperm analysis (CASA) system. Plasmalemma and acrosomal membrane integrity, mitochondrial membrane potential and reactive oxygen species (ROS) were measured by flow cytometry, and lipid peroxidation was evaluated by the thiobarbituric acid reactive substances (TBARS) assay. Seminal plasma removal did not improve sperm kinetics compared to the control group. Centrifugation increased the number of cells with damaged acrosomal membranes (0.77 ± 0.05) and filtration caused greater plasmalemma and acrosomal membrane damage (22.18 ± 1.07). No difference in the mitochondrial membrane potential was observed between groups. In contrast, ROS production was higher in the centrifugation group compared to the control and filtration groups, although no differences in TBARS formation were detected. In conclusion, seminal plasma removal did not improve the quality of thawed buffalo semen compared to control in terms of sperm kinetics, membrane integrity, mitochondrial membrane potential or lipid peroxidation.  相似文献
