A novel bioaugmented organic amendment (SFOA) [consisted of vermicompost (pre‐enriched with plant growth promoting fungi) mixed with pressmud and Azadirachta indica (A. Juss.) seed cake] was developed to reclaim sodic soil and support wheat production. A field trial of the SFOA application with/without chemical fertilizers conducted in completely randomized design with four replications to compare growth, yield and seed protein contents of wheat (Triticum aestivum L.) on sodic soil. The favourable changes occurred in different properties of amended soils were studied. A combined application of chemical fertilizer and SFOA significantly (p < 0·05) increased the number of spikelet per plant (63%) and weight of grains per ear (65%) in the amended soil compared with the control. Likewise, the grain yield, weight of 1000 grains and seed protein contents of wheat were significantly (p < 0·05) increased in the combined application compared with other treatments. The expression of protein bands with molecular weights of 36, 52 and 66 kDa were higher in seeds of wheat under the combined treatment. The improvement in wheat production was attributed to significant favourable changes in different soil characteristics such as bulk density, total organic C, alkaline phosphatase, β‐glucosidase, dehydrogenase and cellulase activities that were increased by 234%, 181%, 234%, 176%, 189% and 150%, respectively, in case of amended soil under the combined treatment compared with the control. The tested SFOA may be recommended as soil amendment for reclaiming sodic soil and supporting wheat cultivation with better crop growth and yield in combination with chemical fertilizers on sodic soil. Structured lipids (SLs) containing palmitic and oleic acids were synthesized by transesterification of tripalmitin with either oleic acid or methyl oleate as acyl donor. This SL with palmitic acid at the sn-2 position and oleic acid at sn-1,3 positions is similar in structure to human milk fat triacylglycerol. LIP1, an isoform of Candida rugosa lipase (CRL), was used as biocatalyst. The effects of reaction temperature, substrate molar ratio, and time on incorporation of oleic acid were investigated. Reaction time and temperature were set at 6, 12, and 24 h, and 35, 45, and 55 degrees C, respectively. Substrate molar ratio was varied from 1:1 to 1:4. The highest incorporation of oleic acid (37.7%) was at 45 degrees C with methyl oleate as acyl donor. Oleic acid resulted in slightly lesser (26.3%) incorporation. Generally, higher percentage incorporation of oleic acid was observed with methyl oleate (transesterification) than with oleic acid (acidolysis). In both cases percentage incorporation increased with reaction time. Incorporation decreased with increase in temperature above 45 degrees C. Initially, oleic acid incorporation increased with increase in substrate molar ratio up to 1:3. LIP1 was also compared with Lipozyme RM IM as biocatalysts. The tested reaction parameters were selected on the basis of maximum incorporation of C18:1 obtained during optimization of LIP1 reaction conditions. Reaction temperature was maintained at 45, 55, and 65 degrees C. Lipozyme RM IM gave highest oleic acid incorporation (49.4%) at 65 degrees C with methyl oleate as acyl donor. Statistically significant (P < 0.05) differences were observed for both enzymes. SL prepared using Lipozyme RM IM may be more suitable for possible use in human milk fat substitutes.  相似文献

Interacting sets of actin assembly factors work together in cells, but the underlying mechanisms have remained obscure. We used triple-color single-molecule fluorescence microscopy to image the tumor suppressor adenomatous polyposis coli (APC) and the formin mDia1 during filament assembly. Complexes consisting of APC, mDia1, and actin monomers initiated actin filament formation, overcoming inhibition by capping protein and profilin. Upon filament polymerization, the complexes separated, with mDia1 moving processively on growing barbed ends while APC remained at the site of nucleation. Thus, the two assembly factors directly interact to initiate filament assembly and then separate but retain independent associations with either end of the growing filament.  相似文献

Fusarium wilt is an important disease of pigeonpea [Cajanus cajan (L.) Millsp.] and it can cause severe yield losses. Chemical control of this disease is difficult and expensive; therefore, cultivation of resistant varieties/hybrids is the most efficient strategy for enhancing the production. In the present study, by using a wilt susceptible cytoplasmic-nuclear male-sterile line and four wilt resistant fertility restorers, one dominant and one recessive gene with dominant suppressive epistatic effects were found responsible for controlling resistance to Fusarium wilt. Considering the annual losses and wide spread nature of wilt diseases in pigeonpea, it is imperative that all the inbred and hybrid cultivars have high level of resistance to this disease. The presence of dominant gene for resistance will increase the efficiency of breeding wilt resistant cultivars because it will yield greater proportion of resistant genotypes in segregating generations. In hybrid breeding also, the presence of dominant gene for wilt resistance will be an advantage. The transfer of this gene in female hybrid parents will ease the breeding of wilt resistant hybrids because this will allow the use of both wilt resistant as well as susceptible restorers in generating wilt resistant hybrid combinations.  相似文献

Reduced growth of plants and excessive generalised and spotty yellowing of leaves occurred in ornamental crotons that were infected with a geminivirus, which was detected by spot hybridization tests using a cloned probe derived from DNA-A of acalypha yellow mosaic geminivirus. The croton virus was transmitted by the whitefly,Bemisia tabaci, toNicotiana tabacum andAcalypha indica, in which it caused leaf curl and yellow mosaic, respectively, but it was not transmitted to five other species. Infected crotons may therefore serve as reservoirs of a geminivirus that is able to infect a crop species.  相似文献

The study was conducted to investigate the efficacy and physio‐metabolic responses of single and multiple doses of fenbendazole (FBZ) in Labeo rohita against gill parasite (Dacylogyrus sp.). Two Isoenergetic (395 Kcal DE/100 g) and isonitrogenous (35% CP) purified diets viz. control (without FBZ) and treatment (0.2% FBZ) were prepared. The efficacy of FBZ was measured after administration of single dose (20 mg/kg b. wt.) on 0th day and multiple doses of FBZ, that is, 20 mg/kg b. wt. on 0th, 3rd and 7th day through diet. Although oral administration of FBZ at single dose could significantly reduce the parasite in fish in comparison with the control group, 100% efficacy of the drug was not achieved up to 15th day of treatment. On the other hand, multiple doses of FBZ could show almost 100% efficacy on 15th day in terms of elimination of parasites from gill. The drug induced oxidative stress in examined fish. There was no significant change in activities of hepatic and muscle aspartate aminotransferase and alanine aminotransferase of L. rohita up to 15th day after oral administration of FBZ at single dose. But both single and multiple doses of FBZ could significantly enhance the hepatic superoxide dismutase, catalase and glutathione‐S‐transferase activities in fish up to 7th day of drug administration and significantly decreased thereafter up to 15th and/or 30th day. It is concluded that oral administration of FBZ at multiple doses (20 mg/kg b. wt.) on 1st, 3rd and 7th day could be effective against Dactylogyrus sp. infection in L. rohita.  相似文献

Six intervarietal crosses involving two resistant and three susceptible genotypes of mungbean were attempted with the objectives to determine the mode of inheritance of gene‐specific Mungbean Yellow Mosaic Virus (MYMV) resistance. An infector row technique along with artificial inoculation was used for evaluating parents, F1, F2 and F3 plants for MYMV resistance. Disease scoring for MYMV indicated that F1s were highly susceptible as were the susceptible parents while resistant parent exhibited resistant reaction. The F2 progeny segregated in the ratio of 9 S:3 MS:3 MR:1 R suggesting that the resistance was governed by digenic recessive genes (rm1 and rm2). When one gene (rm1) was present in the homozygous recessive condition in different plants, it conferred moderately susceptible (MS) reaction, whereas when other gene (rm2) was in homozygous condition, moderately resistant (MR) reaction was obvious. When both genes (rm1 and rm2) were present together in the homozygous recessive condition, resistant reaction (R) was observed. The F2 segregation explained on the basis of phenotypic expression was further confirmed by F3 segregation.  相似文献

Dichlorvos was applied as spray at 1 and 2% concentrations daily for a period of 28 and 21 consecutive days, respectively to buffalo calves. Animals sprayed with 1% dichlorvos displayed mild to moderate clinical signs of toxicosis during the 4th week of exposure. The higher concentration (2%) produced clinical signs of poisoning after 12-16 applications, and was lethal to one of three animals. Daily spraying of dichlorvos at both concentrations inactivated erythrocyte cholinesterase (ChE) (15-21%), plasma ChE (17-20%) and serum carboxylesterase (5-10%) within 3 days. The extent of inhibition of esterases was increased with repeated treatment and maximal inhibition of erythrocyte ChE (80-89%), plasma ChE (81-91%) and serum carboxylesterase (33-54%) with 1 and 2% concentrations was observed on the 28th and 21st day after start of application, respectively. In surviving animals, blood esterases remained inactivated to the extent of 14-65% on the 14th day after the termination of treatment. Dichlorvos at both concentrations significantly (P less than 0.01) elevated the serum levels of aspartate aminotransferase, alanine aminotransferase, acid phosphatase and alkaline phosphatase. The activities of these enzymes in surviving animals recovered to control values within 14 days after the final application of dichlorvos.  相似文献
