The dynamics of a recent outbreak of the spruce bark beetle (Ips typographus) in Switzerland was ruled by a devastating winter storm in 1999 and the drought and heat of the summer 2003. Starting from a similar level of population sizes, estimated as the rate of infested growing stock, beetle populations increased differently in magnitude and time among different regions in Switzerland. Accordingly, we expected local or regional genetic differentiation as a result of such repeated population expansion/breakdown dynamics. We analyzed 5 nuclear microsatellites of spruce bark beetles sampled from pheromone traps at 30 locations distributed over Switzerland. Our genetic results did not indicate any sign of population differentiation, structure, isolation by distance, or recent bottlenecks. This complete lack of genetic structure suggests that spruce bark beetles are highly mobile, precluding the formation of a spatial structure at neutral molecular markers. Thus, this molecular–genetic approach does not allow us to discriminate among regional gene pools and to identify the origin of expanding beetle populations.  相似文献

A potential limitation of tree-ring based climate reconstructions is related to climate signal age effects (CSAE). CSAE may arise if the climatic response of young tree-rings differs from that of old tree-rings. This could mean that climatic signals become stronger (or weaker) with tree aging, or that the seasonality of signals or the sensitivity to a specific element (e.g., temperature, precipitation) changes over time. Such changes would affect the interpretation of dendroclimatic reconstructions, as the tree-rings included in these records are generally oldest at the end of a record (e.g., 21st century)—which is the time period generally used for calibration with instrumental data.We here addressed this concern by analyzing young and old Pinus cembra trees from three high elevation sites in the central European Alps. Core and disc samples were collected in pre-defined plots to allow for a representative analysis of tree ages with tree-ring width (TRW) measurement series categorized into age classes (i) >1880, (ii) 1880–1939, and (iii) 1940–2002. Notably we report on the signal of the very young category (iii) not yet described in literature, and thus allow estimation of climate response and signal strength characteristics during the first years of the trees’ lifespans.Comparison of age classes (i)–(iii) revealed differences in TRW coherence and size, but little change in climatic signal. CSAE are in the order of the differences recorded among high elevation sites—a conclusion that holds for inter-annual to decadal scale TRW variations at near-treeline Swiss stone pine. Such data are typically included in regional and larger-scale temperature reconstructions; thus, our results add confidence to long-term climate estimates integrating a range of tree-ring age classes. Other findings, such as the reaction wood in juvenile tree-rings, and sensitivity of the climate signal to sample replication, suggest that comparisons of young and old age classes, and separate calibration of these categories against instrumental climate data might further the estimation of long-term uncertainty changes in tree-ring based climate reconstructions.  相似文献

Flours from nonsprouted (ns) kernels and dried sprouted (s) kernels of transgenic rye expressing HMW glutenin subunits (HMW‐GS) 1Dy10 (L10) or 1Dx5+1Dy10 (L5+10) from wheat were compared with flours from the corresponding wildtype rye (Lwt). The crude protein content of nonsprouted flours ranged from 9.2% (Lwt) to 10.4% (L5+10) and was lowered by ≈1% due to sprouting. Flour proteins were separated into albumins/globulins, prolamins, and glutelin subunits by a modified Osborne fractionation and into SDS‐soluble and insoluble fractions. Portions of the prolamin fractions were reduced in the same manner as glutelins. The different fractions were then characterized and quantified by RP‐HPLC on C8 silica gel. The proportion of albumins/globulins did not significantly differ between transgenic lines and wildtype. The proportions of alcohol‐insoluble glutelins and SDS‐insoluble proteins drastically increased in transgenic rye due to a shift of HMW and γ‐75k secalins into the polymeric fractions. Significant differences in the proportion of highly polymeric proteins between nonsprouted and sprouted flours could not be detected. The quantitative data demonstrated that the expression of HMW‐GS led to a higher degree of polymerization of storage proteins in rye flour. The HMW‐GS combination 1Dx5+1Dy10 showed stronger effects than 1Dy10 alone. The analyzed flours contained two HMW secalins (R1, R2), whose amino acid compositions were closely related to those of 1Dy10 and 1Dx5, respectively. The amounts of R1 in Lwt flours determined by RP‐HPLC were 221 mg (ns) and 186 mg (s) per 100 g and those of R2 were 344 mg (ns) and 298 mg (s), respectively. These amounts increased to 240 mg (ns)/201 mg (s) (R1) and 479 mg (ns)/432 mg (s) (R2) in L10 flours. In L5+10 flours, the amount of R1 decreased to 150 mg (ns)/132 mg (s) while R2 increased to 432 mg (ns)/338 mg (s). The amount of HMW‐GS 1Dy10 was almost the same as that of R2 in L10 flours but was strongly increased in L5+10 flour (633 mg [ns]/538 mg [s]). HMW‐GS 1Dx5 was, by far, the major subunit in L5+10 flours (987 mg 7[ns]/896 mg [s]). The summarized amounts of all HMW subunits increased from ≈0.5 g (Lwt) to ≈1.1 g (L10) and ≈2.0 g (L5+10). Thus only L10 flours were similar to wheat flours in HMW subunit content. The baking performance of L10 flour determined by a microbaking test was improved compared with Lwt flour, whereas L5x10 flour showed very poor properties obviously due to the strongly increased proportion of highly cross‐linked glutelins. The breadmaking quality of flours from 1Dy10 seeds and wildtype seeds was reduced by the same degree when flours from sprouted seeds were analyzed.  相似文献

Sie kann im Rahmen eines Gesamt-Bekämptungsplanes jedoch nur als Teil einer Gesamtanfgabe gewertet werden.b) Die Ausrottung der Rinderdasselfliegen kann nur durch zusäliche und planmäßige Sommerabdasslung auf Weide und Alpe erreicht werden.c) Die prophylaktische Anwendung von Sprühemulsionen auf kontaktinsektizider Basis gestattet, den zukü nftigen Befall stark herabzuseen oder schlagartig zu verhüten.d) Neben dem individuellen Schaden kann somit der Herden- und Häuteschaden merklich herabgeset oder gänzlich vermieden werden.e) Der methodische Wert der Einzelbeulenbehandlung gewinnt durch Prophylaxis weiter an Wert. Dadurch steigt der Erfolg planmäßiger Abdasslungen mit dieser Methode. Im Verein mit der Prophylaxis erscheint es möglich, die Gesamtbekämpfungszeit der Rinderdasselfliegen innerhalb eines Gebietes bis zur gänzlichen Ausrottung stark abzukürzen.f) Die Weidebeständigkeit der Rinderdasselffiegein bedarf nah neuen bioklimatologischen Beobachtungen einer Nachprüfung.g) Günstig beurteilte Folgen der prophylaktischen Besprühungen geälpter Tiere auf deren Fleisch- und Entwicklungszustand durch gleichzeitige Fernhaltung bzw. Soil depth plays a decisive role in determining soil properties in mountainous regions for ecological site assessment. To evaluate the use of ground‐penetrating radar (GPR) for fast and high‐resolution mapping within mountainous regions, we examined the possibilities and limitations of GPR to determine soil depth over bedrock and to delineate individual substrate layers formed during the Pleistocene in a periglacial environment (Pleistocene periglacial slope deposits, PPSD). Selected catenae in representative subregions of the study area (Dill catchment, SE Rhenish Massif, Germany) have been successfully mapped using GPR. A practicable method was developed using a 400 MHz antenna to reach a mean penetration depth of 1.5 m and to map different substrates and layers of PPSD based on calibrations of the GPR at soil pits along 12 catenae. Colluvium, the three types of PPSD layers, as well as the in situ bedrock could be distinguished in most sections of the GPR surveys. Characteristic GPR facies caused by intrinsic material properties of the different substrates, such as stone content and soil moisture content, could be distinguished in different geomorphologic and lithological settings. A layer‐based velocity distribution was determined for characteristic substrate layers at soil pits enabling us to considerably enhance the accuracy of soil‐depth prediction. Compared to traditionally surveyed soil profiles, our results demonstrate an accuracy of layer thickness surveying within a standard deviation of approx. 0.1 m. It is demonstrated that the combination of GPR with conventional soil‐pit mapping is an efficient and valid method to produce high‐resolution data of substrate distribution.  相似文献

This study characterizes dehydration and white blush processes, structural alterations and synthesis of phenolic compounds (lignin and suberin), in relation to development of white blush on baby carrot surfaces. Carrots were minimally processed as baby carrots and kept on polypropylene trays with or without polyvinyl chloride (PVC) film at 5 ± 2 °C, 90 ± 5% RH. During storage, baby carrots that were not wrapped with PVC film were rehydrated 1, 1.5, 15 and 17 h after minimal processing. Fresh-cut baby carrots were evaluated for white blush index, sensory analysis (visual scores), fresh matter loss, phenylalanine ammonia-lyase (PAL) activity and structural and histochemical changes. Increases in white blush index and subjective visual scores on the carrot surface occurred in the first hours, when the material was kept on trays without PVC film and after 3 and 6 d, when stored on trays covered with PVC film. Visual assessment of white blush resulted in a more accurate assessment than instrumental evaluation because it allowed the perception of minor differences between distinct white blush levels, especially at the tips. Hydrating baby carrot surfaces for 1 and 1.5 h after processing allowed partial absorption of water by tissues and the orange color was reestablished on the surface. Fifteen hours after processing, even after rehydration, the color did not return to the original orange standard. The rapid increase in PAL activity in the secondary phloem of baby carrots, compared to that of whole carrots, suggested a physiological response as a result of abrasion. Dehydration and structural alterations of the more superficial cell layers were the main causes of white blush in baby carrots that was not related to lignin accumulation, but rather to synthesis of non-structural phenolic compounds.  相似文献

Objectives: To determine (1) the short‐ (to hospital discharge) and long‐ (>6 months) term survival, (2) factors associated with short‐term survival, and (3) the perioperative course for horses with resection and anastomosis of the descending colon. Study Design: Multicentered case series. Animals: Horses (n=43) that had descending colon resection and anastomosis. Methods: Medical records (January 1995–June 2009) of 7 equine referral hospitals were reviewed for horses that had descending colon resection and anastomosis and were recovered from anesthesia. Retrieved data included history, results of clinical and clinicopathologic examinations, surgical findings, postsurgical treatment and complications, and short‐term survival (hospital discharge). Long‐term survival was defined as survival ≥6 months after hospital discharge. Results: Of 43 horses, 36 (84%) were discharged from the hospital. Twenty‐eight of 30 horses with follow‐up information survived ≥6 months. No significant associations between perioperative factors and short‐term survival were identified. Lesions included strangulating lipoma (n=27), postfoaling trauma (4), infarction (4), intraluminal obstruction (2), and other (6). Common postoperative complications included fever and diarrhea. During hospitalization 7 horses were euthanatized or died because of septic peritonitis (3), endotoxemia (3), and colic and ileus (1). Conclusions: Descending colon resection and anastomosis has a favorable prognosis for hospital discharge and survival ≥6 months. The most common cause of small colon incarceration was strangulating lipoma. Clinical Relevance: Complications include postoperative fever and diarrhea but the prognosis is good after small colon resection and anastomosis.  相似文献
