Gene pyramiding (assembling multiple desirable genes into a single genotype) via conventional methods to combat Asian rust resistance in soybean (Glycine max (L.) Merrill) and developing soybean varieties with durable resistance to this disease continues to be a challenge. Therefore, our objectives were to pyramid four Asian rust (caused by Phakopsora pachyrhizi) resistance (Rpp) genes, viz., Rpp1, Rpp2, Rpp3, and Rpp4 from their respective donors, PI200492 (Komata), PI230971, PI462312 (Ankur), and PI459025 (Bing Nan). Two single crosses (PI200492 × PI230971; PI462312 × PI459025) and a double-cross [(PI200492 × PI230971) × (PI462312 × PI459025)] were made for present study. The scoring of parents and crosses was done according to a 0–9 grading scale, where 0 = 0% disease intensity (absolutely resistant), 1 = 1% disease intensity (highly resistant), 3 = 1.1–10% disease intensity (moderately resistant), 5 = 10.1–25% disease intensity (moderately susceptible), 7 = 25.1–50% disease intensity (susceptible), and 9 = more than 50% disease intensity (highly susceptible). Out of four parents used, Rpp1 gene-donor plant introduction (PI)200492 (Komata) was immune to rust, with a disease grade of 0.92. Rpp2 and Rpp4 gene donors, PI230971 and [PI459025 (Bing Nan)], had reddish-brown (RB) lesions. Rpp3 gene-donor PI462312 (Ankur) showed a few, highly localized patches of tan lesions; however, it was not as susceptible as the susceptible check JS335. In cross-A (PI200492 × PI230971), all the F1s were rust resistant, whereas in the F2, 93.9% (845 plants) were either immune (11.0%, 99 plants) showing no infection or had RB lesions (82.9%, 746 plants) with resistant reaction and the remainder 6.1% (55 plants) had susceptible reaction. In cross-B (PI462312 × PI459025), 80.8% (63 plants) of the F1 had RB lesions and resistant reaction, whereas 19.2% (15 plants) had susceptible reaction. In its F2 population, 24.2% (217 plants) were susceptible. In cross-C [(PI200492 × PI230971) × (PI462312 × PI459025)], i.e., double-cross hybrid (DCH), 85.5% (77 plants) were either immune or had RB lesions, whereas the rest of the plants 14.5% (13 plants) showed susceptible reaction. Based on parental survey, four simple sequence repeat primers were short-listed for amplification of individual rust-resistance gene-specific markers in ten plants of each cross, viz., Satt191-210bp (Rpp1 gene), Sat_361-245bp (Rpp2 gene), Satt263-195bp (Rpp3 gene), and Rpp4TM-128bp (Rpp4 gene). In the single-cross hybrid (SCH)-A (PI200492 × PI230971), six plants had both Rpp1 and Rpp2 genes, whereas in another SCH-B (PI462312 × PI459025), seven plants had both Rpp3 and Rpp4 genes. In DCH [(PI200492 × PI230971) × (PI462312 × PI459025)], a single plant had all four Rpp genes, five plants had two Rpp genes, whereas a single susceptible plant had only Rpp3 gene. The homozygous rust-resistant segregants from these crosses can be used for improving rust resistance of otherwise adapted, high-yielding soybean genotypes.  相似文献

The Allyl-heptaisobutyl-polyhedral oligomeric silsesquioxane (AHO-POSS) grafted polypropylene (PP) was prepared by reactive extrusion and by physical blending routes. The structure and properties of physically blended and reactively blended POSS/PP nanocomposites were investigated by FTIR, wide-angle X-ray diffraction (WAXD), differential scanning calorimetry (DSC), thermogravimetric analysis, SEM, spherutlic growth and mechanical properties studies. Chemical bonding of POSS with PP in reactive extrusion was confirmed by FT-IR spectroscopy. DSC and TGA studies showed that the thermal stability of AHO-POSS/PP nanocomposite prepared by reactive extrusion improved significantly as compared to only physically blended nanocomposites. WAXD studies showed decrease in crystallinity of the AHO-POSS/PP nanocomposites prepared by reactive extrusion. SEM studies showed aggregation tendency in case of physically blended AHO-POSS/PP nanocomposites. Spherulite growth studies show reactive blending retards spherulite growth in PP polymer.  相似文献

The structure and properties of physically blended and γ-ray irradiated AHO-POSS/PP nanocomposite filaments were investigated by FTIR, wide-angle X-ray diffraction (WAXD), Thermo-gravimetric Analysis and mechanical property studies. Chemical bonding of AHO-POSS with PP after γ-ray irradiation was confirmed by FT-IR spectroscopy. Grafting resulted in change in mechanical and thermal properties and the extent of change was critically dependent on loading of AHO-POSS in PP and radiation dose level. In general, tensile strength decreased almost continuously with increase in radiation dose whereas thermal stability increased upto a radiation dose of 5 kGy and then decreased. The loss in tensile strength was caused due to chain scission, cross linking and loss in orientation.  相似文献

The resistance status of Rhipicephalus (Boophilus) microplus to "Diazinon" was evaluated in 20 locations situated at various agro-climatic regions of India. Adult immersion test (AIT) was optimized using laboratory reared acaricide susceptible IVRI-I strain of R. (B.) microplus and minimum effective concentration of Diazinon was determined as 635.2 ppm. The discriminating dose (DD) was worked out as 1270.4 ppm and was tested on female ticks collected from organized and unorganized farms located at different agro-climatic regions of India. On the basis of the data generated on three variables viz., mortality, egg masses and reproductive index, the resistance level was categorized as I, II, III and IV. The average resistance factor (RF) of 6.1 (level II) was recorded in the ticks collected from the northern sub-temperate trans-gangetic plains while high average RF values of 26.65 (level III) was recorded in the ticks collected from tropical middle-gangetic plains. The tropical middle gangetic plain has a very high density of animal populations where farmers use Diazinon for tick control, for agricultural practices and for mosquito control. Due to the continuous use of OP compounds the environmental load of Diazinon has become high in the area. This is the first experimental data generated on Diazinon resistant status in ticks of India.  相似文献

The 5'-leader of the HIV-1 genome regulates multiple functions during viral replication via mechanisms that have yet to be established. We developed a nuclear magnetic resonance approach that enabled direct detection of structural elements within the intact leader (712-nucleotide dimer) that are critical for genome packaging. Residues spanning the gag start codon (AUG) form a hairpin in the monomeric leader and base pair with residues of the unique-5' region (U5) in the dimer. U5:AUG formation promotes dimerization by displacing and exposing a dimer-promoting hairpin and enhances binding by the nucleocapsid (NC) protein, which is the cognate domain of the viral Gag polyprotein that directs packaging. Our findings support a packaging mechanism in which translation, dimerization, NC binding, and packaging are regulated by a common RNA structural switch.  相似文献

In December 2020, Euphorbia leucocephala plants exhibiting symptoms of powdery mildew were observed in the Botanical Garden of Tuljaram Chaturchand College, Baramati, Maharashtra, India. Based on morphological identification, DNA sequence analysis, and pathogenicity, the fungal pathogen of the disease was identified as Leveillula clavata. This is the first report of L. clavata on Euphorbia leucocephala from India and in general.  相似文献

Objectives of the present study were to evaluate the effects of supplementation of bypass fat on milk production and reproductive performance of crossbred cows. Nineteen multiparous crossbred cows (2–4 lactation) were divided in two groups on the basis of most probable production ability (MPPA). The animals in group 1 (nine cows, MPPA 3,441.32 kg, control group G1) were fed chaffed wheat straw, chopped green maize, and concentrate mixture as per requirements while the animals in group 2 (10 cows, MPPA 3,457.2 kg, treatment group G2) were fed the same ration supplemented with 2.5% bypass fat (on DMI basis). The cows of G2 were supplemented bypass fat 40 days prepartum to 90 days postpartum and carry over effect of supplementation on milk production and reproductive parameters was monitored up to 210 days of lactation. Average birth weights of the calves were 24.94 and 27.95 kg in G1 and G2, respectively. The calving per cent in G1 (88.88%) was lower than that of G2 (100%). The time taken for expulsion of fetal membranes was decreased (P < 0.05) by 5.4 h in G2 compared to G1. Days required for involution of uterus was less (P < 0.05) in G2 (35.40 days) than that of G1 (49.44 days). Less number of cases of retention of fetal membranes (RFM) and metritis were observed in G2 as compared to that of G1. The average milk yield (210 days) in G2 was higher (P < 0.05) than that of G1 (18.65 vs 17.57 kg/day). Similarly, the milk yield during the carry over period (90 days) was higher (P < 0.05) in G2 than that of G1 (14.81 vs 13.79 kg/day). The time required for onset of cyclicity was reduced (P < 0.05) by 6.5 days in G2 as compared to that of G1. The service period and AI per conception were also reduced (P < 0.05) in G2 while bypass fat feeding showed no effect on conception rate (P < 0.80). There were no differences between the two groups in plasma cholesterol, progesterone and insulin concentrations. Hence, it was concluded that bypass fat supplementation at 2.5% of DMI increased the milk production and reduced the time required for involution of uterus and commencement of cyclicity. AI per conception, incidences of metritis and RFM were also reduced on supplementing bypass fat.  相似文献

Salvadora persica trees with symptoms of witches’-broom, little leaf and severe leaf curling were observed in the Biodiversity Park, North Delhi (India) during February 2010. Using a nested-PCR assay targeting two phytoplasma specific loci (16S ribosomal RNA and the ribosomal protein gene) and sequence analysis of the amplified products, we identified the associated phytopathogen as a phytoplasma of group 16SrI. RFLP-mediated characterization revealed that the phytoplasma belongs to subgroup 16SrI-B. Phylogenetic reconstruction also grouped the phytoplasma within group 16SrI. The study reports a new host of phytoplasma in India and worldwide.  相似文献
