We investigated the role of the cell wall and plasma membrane (PM) of root-tip cells in Al tolerance in Al-tolerant and Al-sensitive cultivars of five plant species (rice, maize, pea, wheat, and sorghum). No correlation was found between the differences in Al tolerance and the cation exchange capacity of cell walls isolated from root-tips (0–1 em). Preliminary exposure to Al for 1 h was sufficient to inhibit subsequent root re-elongation in an Al-free solution, and the inhibitory effect was more pronounced in the Al-sensitive cultivars than in the Al-tolerant ones. Together with the inhibition of root re-elongation, the PM of the root-tip cells of all the Al-sensitive cultivars was more permeabilized than that of the Al-tolerant cultivars, based on the FDA-PI fluorescence staining technique. Exposure for 30 min to Al treatment at 100 µM significantly increased the PM permeability of protoplasts isolated from the root-tips for the Al-sensitive pea cultivar placed in a moderately hypotonic medium. Protoplasts from root-tip portions of all the Al-sensitive cultivars took up more Al than those of the Al-tolerant ones when treated with 100 pM Al under isotonic conditions for 30 min. The co-existence of DNP or hypotonic conditions led to a larger increase of Al uptake by the protoplasts from Al-sensitive maize cultivars. These results suggest that Al ions rapidly alter the PM of the root-tip portion in the Al-sensitive cultivars, irrespective of plant species, resulting in an increase of the PM permeability.  相似文献

Recently, riparian forests have attracted attention as they are effective for ecological preservation and landscape enhancement. Uses of such forests in flood prevention, sediment control, and erosion control works have been actively promoted. This study aims to clarify the effect of density of riparian trees on drag exerted on trees along river channels under the regime of bed load transport. Hydraulic model experiments were performed using a straight channel, and the methods to calculate the drag coefficient necessary for deriving drag exerted on trees were studied. Previous studies have stated that the drag coefficient of a cylinder is fairly constant when Reynolds number,R e, is between 103 and 105 (e.g., Schlichting, 1979). This study clarified, however, that in cases of relatively dense arrangements of model trees (cylinders), the drag coefficient varies greatly with the density of the trees. As such, correlation between the drag coefficient and Reynolds number was found to be slight. Test results indicated that the drag coefficient of trees,C d, correlates strongly with the coefficient of velocity,U′/U * , friction factor of the channel bed,f′, and roughness concentration of trees × flow depth,aH′ or ratio of the area occupied by trees,λ. Furthermore, the drag coefficient,C d, was also found to be slightly correlated with the gradient of the channel bed,I. Equations using either of the following parameters were obtained for deriving the drag coefficient,C d, based on the experiment results:aH′; λ; U′/U * ; and Froude numberF r. C-reactive protein (CRP) was isolated from the acute phase serum of dogs subjected to surgical stimulation. Its properties were characterized. Canine CRP was isolated by ion-exchange chromatography using DEAE-Sephacel and DEAE-Sephadex A-50 and affinity chromatography using protein A-Sepharose CL 4B in combination with agar-block electrophoresis. In immunoelectrophoresis, canine CRP had the same γ-mobility as human γ-type CRP. The molecular weight of purifined canine CRP was estimated by gel filtration and sodium dodecyl sulfate polyacrylamide gel electrophoresis to be approximately 157 000 and 155 000 respectively. This CRP was a thermolabile protein which completely lost its antigenicity by heating at 70°C for 15 min. The serum concentration of CRP in normal beagle dogs ranged from 0.198 to 0.826 μg ml−1 (0.486±0.170 μg ml−1). The concentration was acutely increased by surgery as it was in man and was rapidly decreased with convalescence. Dogs can be a useful animal model for investigation of the mechanism of CRP production and the function of CRP.  相似文献

Four cattle persistently infected with non-cytopathogenic bovine viral diarrhoea-mucosal disease (BVD-MD) virus and 3 normal controls were challenge exposed to cytopathogenic BVD-MD viruses that are antigenically different from the persistent viruses. Two of the persistently infected cattle developed mucosal disease and became moribund on postinoculation days (PID) 28 and 14, respectively; one developed severe and chronic diarrhoea and became moribund on PID 32; and the other remained healthy until the end of the experiment (PID 150). All control cattle showed transient fever, but no diarrhoea, and recovered from infection. Cytopathogenic viruses were isolated from blood of all cattle early in infection (PID 5-10) and from carcasses at necropsy. The former viruses were antigenically identical with the challenge viruses. On the other hand, the antigenicity of the cytopathogenic viruses isolated from carcasses at necropsy were different from that of the challenge viruses but similar to that of the non-cytopathogenic persistent viruses. Three of 4 persistently infected cattle, but not the calf which became moribund on PID 14, produced serum neutralizing (SN) antibodies to the challenge viruses, but not to the persistent viruses and the cytopathogenic viruses isolated from carcasses at necropsy. Control cattle produced SN antibodies to both the challenge and persistent viruses.  相似文献

The aim of this study was to investigate the ovarian follicular development, developmental competence of oocytes, and plasma anti‐Müllerian hormone (AMH) levels of Japanese wild boar crossbred (wild hybrid) gilts, whose litter size is inferior to that of European breeds. Ovary and plasma samples were collected from two different breeds of gilts (wild hybrid and Large White breeds). The ovaries from the wild hybrid gilts had a lower average numbers of secondary follicles and vesicular follicles in ovarian cross‐sections and of good quality oocytes collected from ovarian follicles as compared with those from Large White gilts (< 0.05). The development rate to the blastocyst stage of good quality oocytes after in vitro maturation, fertilization and culture was also lower (< 0.05) in wild hybrid gilts than in Large White gilts. Plasma AMH levels with >0.16 ng/ml were detected in 8.3% of the examined wild hybrid gilts and 33% of the Large White gilts. These results indicate that the low reproductive performance of wild hybrid breed may result in part from low numbers of vesicular follicles and good quality oocytes, and low developmental competence of oocytes. Moreover, plasma AMH levels may support low number of vesicular follicles in ovaries of wild hybrid gilts.  相似文献
