The development of a DNA microarray method on a digital versatile disk (DVD) is described for the simultaneous detection of traces of hazelnut ( Corylus avellana L.), peanut ( Arachis hypogaea ), and soybean ( Glycine max ) in foods. After DNA extraction, multiplex PCR was set up using 5'-labeled specific primers for Cor a 1, Ar h 2, and Le genes, respectively. Digoxin-labeled PCR products were detected by hybridization with 5'-biotinylated probes immobilized on a streptavidin-modified DVD surface. The reaction product attenuates the signal intensity of the laser that reached the DVD drive used as detector, correlating well with the amount of amplified sequence. Analytical performances showed a detection limit of 1 μg/g and good assay reproducibility (RSD 8%), suitable for the simultaneous detection of the three targeted allergens. The developed methodology was tested with several commercially available foodstuffs, demonstrating its applicability. The results were in good agreement, in terms of sensitivity and reproducibility, with those obtained with ELISA, PCR-gel agarose electrophoresis, and RT-PCR.  相似文献

  1. Understanding the interactions among demographic parameters, mating system and population dynamics is key to predict the response of populations to global change. The Mediterranean red coral is a precious octocoral suffering from population decline due to overfishing and warming‐driven mass mortality events.
  2. While the demographic consequences of these two pressures are well characterized, little is known regarding their impact on population dynamics and evolution of red coral populations. The main objective of this study was to fill this gap focusing more particularly on mating pattern and genetic drift.
  3. Combining sibship and progeny arrays analyses, a genetic characterization of the red coral mating system was conducted. In addition, a synchronic approach was developed comparing mating patterns in two populations with contrasting demographic patterns: a pristine‐like population and a declining population.
  4. The results show that polyandry is likely to be the norm in red coral. The similar patterns of genetic diversity between adults and larvae combined with the lack of differential reproductive success among putative fathers did not support significant sweepstakes effects during larval production. While instantaneous biparental inbreeding was detected, no long‐term inbreeding was observed even in the declining population. Mating patterns and effective population sizes in the two populations were not statistically different. Nevertheless, a trend towards a slightly higher inbreeding and a lower number of breeders was observed in the declining population.
  5. Accordingly, we hypothesized that an increase in male gamete dispersal may buffer the increase of genetic drift expected in the declining population. This feedback between demographic decline and reproductive pattern may potentially take part in the long‐term persistence of red coral populations. However, the negative trend reported in the declining population unambiguously supports the need to maintain high densities of reproductive colonies to the functioning of red coral populations.

Mediterranean plants have evolved a complex antioxidant defense system to cope with summer drought. Flavonoids, and particularly flavanols and flavonols, are potent in vitro antioxidants, but their in vivo significance within the complex network of antioxidant defenses remains unclear, especially in plant responses to stress. To gain insight into the role of flavonoids in the antioxidant defense system of Cistus clusii Dunal, we evaluated drought-induced changes in flavonoids in leaves and compared the response of these compounds with that of other low molecular weight antioxidants (ascorbic acid, tocopherols and carotenoids). Among the antioxidant flavonoids analyzed, epigallocatechin gallate was present in the greatest concentrations (up to about 5 micromol dm(-2)). Other flavanols, such as epicatechin and epicatechin gallate, were found at concentrations below 0.25 and 0.03 micromol dm(-2), respectively. Neither of the antioxidant flavonols analyzed, quercetin and kaempferol, were detected in C. clusii leaves. Epigallocatechin gallate, ascorbic acid and alpha-tocopherol concentrations increased to a similar extent (up to 2.8-, 2.6- and 3.3-fold, respectively) in response to drought, but the kinetics of the drought-induced increases differed. Epigallocatechin gallate, epicatechin and epicatechin gallate concentrations increased progressively during drought, reaching maximum values after 30 days of stress. Ascorbic acid concentrations increased twofold after 15 days of drought, and maximum values were attained after 50 days of drought. In contrast, alpha-tocopherol concentrations remained constant during the first 30 days of drought, but increased sharply by 3.3-fold after 50 days of drought. The maximum efficiency of photosystem II photochemistry and the extent of lipid peroxidation remained constant throughout the drought period, whereas the redox state of ascorbic acid and alpha-tocopherol shifted toward their reduced forms in response to drought, indicating that the concerted action of low molecular weight antioxidants may help prevent oxidative damage in plants.  相似文献

The abandonment of traditional rural life in mountain areas has favoured the expansion of forest in the headwaters of Pyrenean rivers. In this paper, we (i) analyse hydro‐climatic trends at the annual and monthly scales in three nested sub‐catchments in a central Pyrenean basin and (ii) quantify the relative contribution of climate change and forest cover on the observed changes in runoff. Land use maps indicate an increase in the forest cover in all sub‐basins for the period 1987–2009. Non‐parametric Mann–Kendall statistic on annual and monthly hydro‐climatic data from 1965 to 2009 was used to identify trends in climate and runoff data. To assess the potential influence of the period length, trends were also examined for the 1941–2009 stretch. Upward trends were detected for temperature and potential evapotranspiration, particularly during summer and winter months. Precipitation trends and their significance depended on the length of the period considered, although overall results indicated a decrease, particularly for February and July. The effects of climate change and land cover change on annual runoff were identified by means of a change point analysis. Results indicated that a change in annual runoff took place in the 1980s, suggesting the non‐linearity in the relationship between precipitation and runoff, and thus the influence of land use factors. The influence of forest cover was confirmed by comparing observed runoff trends with those resulting from a hydro‐climatic model that does not take into account land use changes. Divergence between both trends revealed that forest cover can account for ~40% of the observed decrease in annual runoff. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

Water flow is a fundamental characteristic required for the ecological integrity of stream ecosystems. However, populations of many freshwater fishes in the Mediterranean region are threatened by man‐induced drought due to water diversion. Mark‐recapture methodology is an effective tool for estimating fish abundance and survival probability, but it has been seldom used with Mediterranean freshwater fish. We tagged over 2,400 individuals of two threatened cyprinids (Mediterranean barbel Barbus meridionalis and Catalan chub Squalius laietanus) inhabiting an Iberian stream affected by water diversion, and used mark‐recapture methods to evaluate the effects of flow, temperature and depth on their apparent survival and abundance at hydrologically altered and perennial reaches. Based on estimates over the summer drying season, survival of Mediterranean barbel was more than six times higher in an upstream reach with permanent flow than a middle reach impacted by water diversion. Water depth was the most important habitat feature accounting for differences in survival, with both barbel and chub exhibiting higher survival and abundance at permanent sites farther downstream where flow had been restored and in an unaltered tributary. Our results show clear negative impacts of water diversion on fish populations. Both species, but particularly barbel, have life‐cycle characteristics that may allow populations to recover quickly if natural streamflow was restored by conserving water and reducing water diversion.  相似文献
