A combination of culture-independent and culturing methods was used to assess the diversity of soil bacterial communities from four locations along 77 °S in Victoria Land, Antarctica. Soil samples were from the coast at Marble Point, in the Wright Valley from Bull Pass and near Lake Vanda, and from Mt. Fleming near the polar plateau. Total carbon and nitrogen, and water content of the soils were low, whereas total P was very high. The pH of the soils varied from extremely alkaline to slightly acid and electrical conductivity was medium to high on the coast and very high in inland soils from Bull Pass and Mt. Fleming. The average monthly air temperature was similar (−18 °C to −24 °C) at all the sites; however, in summer surface soil temperatures were >0 °C at Marble Point and in the Wright Valley for a total of 1100 and 1700 h, respectively. Marble Point soil had the most potential to support bacterial growth and activity with a mean total of 310 h per year when surface soils had a liquid volumetric soil moisture content >5%. Highest counts of culturable heterotrophs occurred in soil from Marble Point, whereas Mt. Fleming soil contained few organisms and had no liquid soil moisture recorded. Seven hundred and twenty-eight clones and 71 bacterial isolates were screened by restriction fragment length polymorphisms, and representatives of those dominant ribotypes that occurred more than 3 times were sequenced. The dominant ribotypes grouped within the bacterial divisions Bacteroidetes, Actinobacteria, Proteobacteria, Thermus-Deinococcus, Acidobacteria, Firmicutes and Cyanobacteria. The closest relatives of the amplicon library clones or cultured bacteria include the genera Hymenobacter, Gillisia, Arthrobacter, Rubrobacter, Friedmanniella, Deinococcus and Leptolyngbya. Many of the clones and bacteria were most similar to others from Antarctic sources, in particular a cyanobacterium-dominated cryptoendolithic community in Beacon sandstone. Some ribotypes were more prevalent in drier soils of the Wright Valley, including relatives of Deinococcus, Rubrobacter and clone FBP460 from Beacon sandstone. Bacterial communities from Marble Point soils were more diverse than those of the Wright Valley. Very few bacteria were isolated from Mt. Fleming soil.  相似文献

Synthetic wood composite films containing cellulose, hemicelluloses, and lignin, the three major components of natural wood, were prepared in a room temperature ionic liquid solvent, 1-ethyl-3-methylimidazolium acetate, [EMIM][Ac]. Various synthetic wood composites were obtained by dissolution of individual wood components together with additives, including polyethylene glycol (PEG), chitosan, and multi-wall carbon nanotubes (MWNTs) in [EMIM][Ac]. The addition of water affords a gel that was dried in either a low humidity environment or under vacuum. Synthetic wood films showed smoother surface textures, higher water resistance, and higher tensile strengths than cellulose films formed by the same methods. Tailor-made synthetic wood composites were also prepared having a variety of desirable properties, including antimicrobial activities, controlled hydro-phobicity/philicity, high relative dielectric constant, and a high degree of cohesiveness.  相似文献

Stellettin B was isolated from marine sponge Jaspis stellifera. In vitro antitumor activities were investigated on 39 human cancer cell lines. Stellettin B exhibited highly potent inhibition against the growth of a human glioblastoma cell line SF295, with a GI50 of 0.01 μM. In contrast, stellettin B showed very weak inhibitory activity on normal cell lines including HMEC, RPTEC, NHBE and PrEC, with GI50s higher than 10 μM, suggesting its relatively selective cytotoxicity against human cancer cells compared to normal human cell lines. We then focused on the antitumor activity of this compound on SF295 cells. Flow cytometric analysis indicated that stellettin B induced apoptosis in SF295 cells in a concentration-dependent manner. Further study indicated that stellettin B increased the production of ROS, the activity of caspase 3/7, as well as the cleavage of PARP, each of which is known to be involved in apoptosis. To investigate the molecular mechanism for cell proliferation inhibition and apoptosis induction, effect on the phosphorylation of several signal proteins of PI3K/Akt and RAS/MAPK pathways was examined. Stellettin B inhibited the phosphorylation of Akt potently, with no activity on p-ERK and p-p38, suggesting that inhibition of PI3K/Akt pathway might be involved in the antiproliferative and apoptosis-inducing effect. However, homogenous time-resolved fluorescence (HTRF) assay indicated that stellettin B did not inhibit PI3K activity, suggesting that the direct target might be signal protein upstream of Akt pathway other than PI3K.  相似文献

The locations of the pole and rotation axis of asteroid 25143 Itokawa were derived from Asteroid Multiband Imaging Camera data on the Hayabusa spacecraft. The retrograde pole orientation had a right ascension of 90.53 degrees and a declination of -66.30 degrees (52000 equinox) or equivalently 128.5 degrees and -89.66 degrees in ecliptic coordinates with a 3.9 degrees margin of error. The surface area is 0.393 square kilometers, the volume is 0.018378 cubic kilometers with a 5% margin of error, and the three axis lengths are 535 meters by 294 meters by 209 meters. The global Itokawa revealed a boomerang-shaped appearance composed of two distinct parts with partly faceted regions and a constricted ring structure.  相似文献

To enhance numerical modeling of the coastal ecosystem complex (CEC), we reviewed the CEC and related concepts along with the current coastal ecosystem model framework in this study. We identified two model implementation paths from the initial objectives to numerical models: specific model building, and the use of existing model frameworks. As the CEC is still at the conceptual stage, both paths are possible. Four important ecological features of CEC modeling (population connectivity, habitat heterogeneity, ontogeny of organisms, and trophic interactions) were also identified. Models for population connectivity, species distributions, life histories, and food webs were categorized using these features. We found that some previously established concepts (between–habitat interactions, coastal ecosystem mosaic, and seascape nursery) overlap with the CEC concept. Several existing integrated model frameworks were reviewed, focusing on their potential to simulate CEC processes. Building specific models for the CEC at the current conceptual stage will be challenging, and modification of existing models will be needed if they are to be used for CEC modeling. Habitat function, ontogenetic development in early life stages, and recruitment variability are important factors when modifying existing models for the development of CEC models. Although model complexity should become high to reproduce observed ecoclogical processes, an intermediate level of model ccomplexity is feasible to decrease parameter uncertainty in models for fisheries management.  相似文献

Canine transitional cell carcinoma (cTCC) is the most common naturally occurring bladder cancer and accounts for 1–2% of canine tumors. The prognosis is poor due to the high rate of invasiveness and metastasis at diagnosis. Sorafenib is a multi-kinase inhibitor that targets rapidly accelerated fibrosarcoma (RAF), vascular endothelial growth factor receptor (VEGFR)-1, VEGFR-2, VEGFR-3, platelet-derived growth factor receptor-β (PDGFR-β), and KIT. In previous studies, a somatic mutation of B-rapidly accelerated fibrosarcoma (BRAF) and expressions of VEGFR-2 and PDGFR-β were observed in over 80% of patients with cTCC. Therefore, in this study, we investigated the anti-tumor effects of sorafenib on cTCC. Five cTCC cell lines were used in the in vitro experiments. All five cTCC cell lines expressed VEGFR-2 and PDGFR-β and sorafenib showed growth inhibitory effect on cTCC cell lines. Cell cycle arrest at the G0/G1 phase and subsequent apoptosis were observed following sorafenib treatment. In the in vivo experiments, cTCC (Sora) cells were subcutaneously injected into nude mice. Mice were orally administered with sorafenib (30 mg/kg daily) for 14 days. Sorafenib inhibited tumor growth compared to vehicle control. The necrotic area in the tumor tissues was increased in the sorafenib-treated group. Sorafenib also inhibited angiogenesis in the tumor microenvironment. Thus, sorafenib may be potential therapeutic agent for cTCC via its direct anti-tumor effect and inhibition of angiogenesis.  相似文献
