Treatment of HeLa S(3) cells with actinomycin D during mitosis suppresses the synthesis of several major classes of nonhistone chromosomal proteins during the subsequent period before DNA replication, but allows the synthesis of other species of these proteins. Such results are consistent with the utilization of preexisting, as well as newly transcribed, messenger RNA's for nonhistone chromosomal protein synthesis during the prereplicative phase of the cell cycle. 相似文献

The reaction of ferricytochrome c with hydrated electrons produced at pH 6.8 by radiolysis with electron pulses lasting 50 to 1000 nanoseconds has a specific rate constant of 5.5 x 10(10) liter mole(-1) sec(-1) for the formation of the primary adduct. By using appropriate wavelengths, another fast, consecutive process was demonstrated, with the pure first order rate constant 1.0 x 10(5) sec(-1). Functional correlation between aromatic and divalent-sulfur amino acids in ribonuclease is shown in the reaction of ribonuclease with hydrogen atoms in aqueous solution. Measurements made once or twice a year from 1977 through 1982 show large correlated changes in gravity, elevation, and strain in several southern California networks. Precise gravity surveys indicate changes of as much as 25 microgals between surveys 6 months apart. Repeated surveys show that annual elevation changes as large as 100 millimeters occur along baselines 40 to 100 kilometers long. Laser-ranging surveys reveal coherent changes in areal strain of 1 to 2 parts per million occurred over much of southern California during 1978 and 1979. A method was developed to analyze rat tissue, fat, and blood for some of the chlorinated compounds found in an extract of soil from an industrial waste site. Extraction with hexane and then with ethyl ether-hexane (1 + 1) was followed by concentration over steam, and gas chromatographic analysis with an electron capture detector. Volatile compounds were analyzed in a glass column coated with 6% SP-2100 plus 4% OV-11 on Chromosorb W. Semivolatile compounds, chlorinated compounds, and pesticides were analyzed in a 70 m glass capillary column coated with 5% OV-101. Phenols were analyzed in a glass column packed with 1% SP-1240 DA on Supelcoport. However, the most efficient means of separation was to use the same glass column for volatile compounds, a DB-5 fused silica capillary column for semivolatile compounds, pesticides, and phenols, and the same 1% SP-1240 DA glass column for separation of beta-BHC and pentachlorophenol. Recoveries ranged from 86.3 +/- 9.1% (mean +/- standard deviation) to 105 +/- 10.4%. Sensitivities for semivolatile chlorinated compounds, pesticides, and phenols were about 4 ng/g for fat, 1 ng/g for tissue, and 0.2 ng/mL for blood. Sensitivities for volatile compounds were about 4-fold higher (16, 4, and 0.8, respectively). Sensitivities for dichlorobenzenes and dichlorotoluenes were 8 ng/g for fat, 2 ng/g for tissue, and 0.4 ng/mL for blood. 相似文献

Soybean cultivars carrying the `long juvenile trait' show a delayed flowering response under short day conditions. The incorporation
of this character into genotypes of agronomic interest may allow a broader range of sowing dates and latitudes for a single
cultivar adaptation. The objective of this work was to identify molecular markers linked to the juvenile locus in soybean.
Experiments were carried out using two pairs of near isogenic lines(NILs) differing in the presence of the long juvenile trait,
and RAPD markers. Four hundred primers were first screened to find polymorphism associated with the trait. Additional differences
between NILs were sought by digesting the genomic DNA with five restriction enzymes. Polymorphic fragments detected between
NILs were tested for linkage to the juvenile locus in the corresponding F2 segregating populations. Marker bc357-HaeIII was linked (χ2L = 46.316) to the juvenile locus with an estimated recombination frequency of 0.13 ± 0.03in one of the genetic backgrounds
studied. The fragment was cloned, sequenced and converted into a SCAR marker. Moreover,bc357-HaeIII was used as RFLP probe. Both, SCAR and RFLP generated markers linked to the juvenile locus in the two genetic backgrounds
analysed. Results presented in this work can be utilised for both, the localisation of the gene associated with the character
and for tagging the juvenile trait in soybean breeding programs.
