A new foliar disease was observed on sissoo in Jaipur and Udaipur districts of Rajasthan, India. The disease occurred in approximately 30% of the trees surveyed. Leaf spots were mostly irregular and these start from leaf margin to inwards. The fungus was identified as Curvularia affinis Boedijn and Koch’s postulates fulfilled. On the basis of the literature, this is the first report of C. affinis causing leaf spot of Dalbergia sissoo from India as well as worldwide.  相似文献

Pasteurella multocida is a causative agent of many major diseases of which haemorrhagic septiciemia (HS) in cattle & a buffalo is responsible for significant losses to livestock sector in India and south Asia. The disease outcome is affected by various host- and pathogen-specific determinants. Several bacterial species-specific putative virulence factors including the capsular and virulence associated genes have been proposed to play a key role in this interaction. A total of 23 isolates of P. multocida were obtained from 335 cases of various clinically healthy and diseased cattle. These isolates were examined for capsule synthesis genes (capA, B, D, E and F) and eleven virulence associated genes (tbpA, pfhA, toxA, hgbB, hgbA, nanH, nanB, sodA, sodC, oma87 and ptfA) by PCR. A total of 19 P. multocida isolates belonging to capsular type B and 4 of capsular type A were isolated. All isolates of capsular type B harboured the virulence associated genes: tbpA, pfhA, hgbA, sodC and nanH, coding for transferrin binding protein, filamentous hemagglutinin, haemoglobin binding protein, superoxide dismutase and neuraminidases, respectively; while isolates belonging to capsular type A also carried tbpA, pfhA, hgbA and nanH genes. Only 50 % of capsular type A isolates contained sodC gene while 100 % of capsular type B isolates had sodC gene. The gene nanB and toxA were absent in all the 23 isolates. In capsular type A isolates, either sodA or sodC gene was present & these genes did not occur concurrently. The presence of virulence associated gene ptfA revealed a positive association with the disease outcome in cattle and could therefore be an important epidemiological marker gene for characterizing P. multocida isolates.  相似文献

The consistent failure to isolate bona fide pluripotent cell lines from livestock indicates that the underlying mechanisms of early lineage specification are poorly defined. Unlike other species, the contrivances of segregation have been comprehensively studied in the mouse. In mouse, FGF/MAPK signalling pathway dictates the segregation of hypoblast (primitive endoderm). However, it is not evident whether this mechanism is also conserved in livestock. Here, in this study, we examined the roles of FGF/MAP kinase signalling pathways in porcine parthenogenetic embryos during the early development. Porcine parthenogenetic embryos were cultured in the medium addition with FGFR inhibitor BGJ398 (10 μm ) or DEMOS. Pluripotency‐ and lineage‐related gene expressions in the early porcine embryos were determined. Compared to control, total cell numbers on day 7 were significantly higher (55 ± 5.96 vs 47 ± 1.97, p < 0.05) in embryos cultured in the presence of BGJ398, but had no significant effect on the rate of blastocyst development (47% vs 44%, p > 0.05). Nonetheless, BGJ398 treatment significantly augmented the expression of pluripotency and trophoblast marker genes (SOX2, OCT4, KLF4 and CDX2), but did not significantly change the expression of NANOG and hypoblast marker gene (GATA4). Furthermore, the addition of FGF signalling agonist (FGF2) during the embryo development significantly decreased the expression of pluripotency and trophoblast marker genes (SOX2, NANOG, KLF4 and CDX2), but no significant effect on the expression of OCT4 and GATA4 was observed. Here, we exhibit that inhibition of FGF signalling could improve the quality of the porcine embryo and escalate the chance to capture pluripotency. Besides, it also promotes the trophoblast development of porcine parthenogenetic embryo. In addition, the data suggested that FGF signalling pathway is dispensable for the segregation of hypoblast and epiblast lineages in porcine embryo during the early development.  相似文献

Three populations of an intervarietal durum wheat cross IWP5308/PDW208, F5, F5BIP1 (population derived after intermating in F2) and F5BIP2 (population derived after intermating in BIPF1), were evaluated under three different agronomic environments for mean performance and stability of genotypes for grain yield, yield components and protein content. Though the biparental progenies indicated a higher mean performance, they did not differ significantly from progenies of the pedigree method for almost all characters. The biparental progenies, however, produced a higher number of stable genotypes for grain yield per plant, grains per ear and protein content. The F5 population had a higher number of stable genotypes for 1000 grain weight and number of tillers per plant. The BIP progenies also had a higher number of genotypes with above average mean performance, and many were significantly higher than the checks WH896 and WH542, compared with F5 progenies. Hence, in spite of high G x E interactions, the use of cycles of biparental mating and selection of top yielding lines on the basis of yield components can enable selection of stable genotypes with high protein content. Number of tillers per plant and 1000 grain weight were the yield component characters which made maximum contribution to phenotypic stability of the genotypes. This paper presents an experimental study on thedevelopment of an efficient energy dissipator ascompared to Garde's energy dissipator (Garde et al.1986) for pipe outlets. The recommended designs werelaboratory tested on scale models with pipe outletdiameter of 10 cm and 7.5 cm for Froude number rangingfrom 1.70 to 5.50. The performance of the newdissipator improved tremendously by using new shapes,sizes and locations of the appurtenances for the samelength of stilling basin. This was possible due tobetter spreading of efflux jet by using a propersplitter block. The formation of a strong vortexinfront of the solid impact wall, in place of a grid,and an additional horizontal shear at the bottomproduced more fine grained eddies and turbulence,which finally reduced the energy of the outgoing flow. The low bottom velocities at the end of the basinresulted in reduction of the scour. The performanceof various models has been compared by estimating thescour index based on maximum depth of scour and itslocation at the end of run time.  相似文献

Sodic soils are characterized by high exchangeable sodium on exchange sites, soil pH greater than 8.5, relatively low electrical conductivity, low infiltration rate and dispersed clay. These characteristics restrict the capacity of soil to absorb water, resulting in poor infiltration. Evidently, these soils require application of irrigation water at shorter intervals for crop production. Thus, irrigation strategy for sodic soils differs from that of normal soils. An experiment to determine the suitable irrigation strategy along with methods of application namely: surface (farmer’s practice), sprinkler (double nozzle impact sprinkler), and low-energy water application device (LEWA) were initiated in the year 2012 for rice crop. Irrigation depths of 6 cm in case of surface method and 4 cm in case of sprinkler and LEWA methods were applied at each irrigation event. The irrigation events for rice were scheduled at 2-DAD (days after the disappearance of the ponded water), 3-DAD, and 4-DAD through surface method, and at daily, 1- and 2-day intervals (after initial ponding disappeared) by sprinkler and LEWA methods. Sprinkler and LEWA methods resulted in highest rice yield of 4.4 t ha?1 in irrigated plots at the 2-day interval which was at par with the highest yielding surface-irrigated plot scheduled at 2-DAD. At the same time, irrigation strategy of 2-day interval through sprinkler and LEWA methods registered water saving to the extent of 30–40% over 2-DAD under surface irrigation method. Results revealed that there could be substantial saving of water and energy (electricity and diesel) through the use of sprinkling devices for irrigating rice under sodic soil environments.  相似文献

A simple, cost-effective and rapid viral nucleic acid release (NAR) buffer suitable for RT-PCR based diagnostic assay was developed for the detection of potato viruses. The NAR buffer and commercially available RNA isolation kit were compared for RT-PCR based assay, where an amplicon of expected size (~380 bp) targeting PVY was observed in both isolations indicating that it can be used in RT-PCR based diagnostic assays. The same was further validated for its repeatability by running across more than hundred suspected potato leaf samples collected from different sources where, it showed consistent results for the presence of PVY indicating its reliability. The NAR buffer assay was examined for its sensitivity in comparison with the kit based isolation where both the assays were able to detect even up to 10?5 dilution without affecting the sensitivity. NAR buffer was found stable up to 28 days at -20 °C and for 14 days at 4 °C without losing PCR sensitivity. The assay was also found effective to release the nucleic acid from potato leaves, thrips and aphids for PCR and RT-PCR based detection of DNA viruses like ToLCNDV-potato and other RNA viruses. The developed protocol is simple, less laborious, time-saving (10-15 min) and economical (1/100th of kit) as compared to kit based protocol. The assay can be adopted in diagnostic laboratories for detection of RNA/DNA viruses from potato plants and in thrips as well.  相似文献

The study was conducted to investigate the aggregate stability and distribution of organic carbon (C) in different-sized aggregates and mineral fractions in a loamy soil under rice-wheat system with continuous application of fertilizer nitrogen, phosphorus and potassium (NPK), farmyard manure (FYM), N+FYM and NPK+FYM, compared with unfertilized control. Macro-aggregates comprised 37.1–49.3% of the total water-stable aggregates (WSAs), compared with 23.3–30.3% as micro-aggregates. Application of inorganic fertilizers and FYM significantly increased the proportion of macro-aggregates, which were linearly related to total organic C (TOC). Organic C concentration in coarse macro-aggregates (CMacA) was higher than the micro-aggregates (CMicA). Application of FYM significantly increased the concentration of organic C in different-sized aggregates and mineral fraction, compared with the unfertilized control. Averaged across treatments, mineral-associated C comprised 26% of TOC. Macro-aggregates, on an average, constituted 66–68% of C preservation capacity of WSA. The amount of TOC sequestered was higher when NPK+FYM was applied together.  相似文献
