Abstract: A 1‐year‐old intact female miniature Dachshund was presented with hematochezia, vomiting, and diarrhea of more than 1‐week duration. An abdominal mass was palpated, which at exploratory surgery was found to be a 7‐cm‐long thickened section of ileum. The thickened ileum was resected. Impression smears revealed numerous small‐ to medium‐sized lymphocytes, with a smaller number of cells resembling Mott cells. The Mott‐like cells contained multiple pale vacuoles that were positive for periodic acid‐Schiff (PAS) in wet‐fixed smears, consistent with Russell bodies. Histologic evaluation of the surgically excised ileum revealed 2 populations of neoplastic lymphoid cells. The majority were uniform medium‐sized lymphocytes with hyperchromatic oval or round nuclei and inconspicuous nucleoli. The remaining cells resembled Mott cells, which contained several PAS‐positive eosinophilic globules in the cytoplasm, occasionally compressing the nucleus. The majority of neoplastic cells stained positively for vimentin, CD20, CD79a, and Pax‐5, but were negative for CD3 and lysozyme; 43.5% of cells stained positively for Ki‐67. The Mott cells were strongly positive for immunoglobulin but were negative for Pax‐5. Using electron microscopy, a homogenous substance of intermediate electron density was observed frequently in the cisternae of rough endoplasmic reticulum in the cytoplasm of the Mott cells, and rarely in the perinuclear cisternae of the lymphoid cells, corresponding to the site of immunoglobulin staining. Monoclonal rearrangement of immunoglobulin heavy‐chain (IgH) gene was observed by PCR testing for lymphocyte–antigen receptor rearrangement. The morphologic features, immunophenotype, and IgH gene rearrangement verified the lymphoid cells were neoplastic (mature cell type) and had a B‐cell phenotype, with evidence of immunoglobulin production and differentiation into Mott cells. This case was unusual because of the age of the dog and because most intestinal lymphomas are T‐cell phenotype. The Mott cell morphology also differed from typical mature B‐cell lymphoma types and may be a unique B‐cell lymphoma variant.  相似文献

Play behavior in young animals has been used to evaluate the condition (health) of livestock. We investigated age, sex, and genetic differences of Japanese Black calves in relation to frequency of play behaviors (galloping, leaping, turning, bucking, head butting objects, and head shaking) and examined how these relationships might affect growth during the suckling stage. Locomotor play behaviors (galloping, leaping, turning, and bucking) and head butting objects gradually declined with the age for both sexes. The frequency of head butting was significantly higher in males than females. We found that significant interaction effects (age × MAOA polymorphism) in play behaviors (except head shaking) and the frequencies of locomotor play in calves without the wild‐type allele were significantly higher than those in younger calves (2 and 6 weeks of age). Weight gain was significantly correlated with the frequency of locomotor play in females, but not in males. This study suggests that play in Japanese Black calves gradually declines as they mature and that play may be controlled by variations in the MAOA gene. In addition, the frequency of locomotor play may be an indicator of health in female calves.  相似文献

We used conventional cross-breeding techniques to develop a carnation with low ethylene sensitivity. A time-lapse video recording system for evaluating ethylene sensitivity of flowers, developed in our earlier study, proved useful for this breeding. We selected 10 new ethylene-resistant lines, in addition to the previously selected line 64–54. Lines 902–48a, 234–43S, and 234–36S showed extremely low ethylene sensitivity. The response time to 10 μL L−1 ethylene of these three lines exceeded 50 h in two independent experiments (2004 and 2005), whereas that of three sensitive control cultivars (‘White Sim’, ‘Excerea’, and ‘Scania’) was 6.2–8.0 h. The mean vase life of the ethylene-resistant lines ranged from 7.6 to 12.7 days (1.4–2.3 times that of ‘White Sim’). The application of ethylene to carnation flowers induced autocatalytic ethylene production in both petals and gynoecia of 9 of the 11 ethylene-resistant lines without petal wilting, indicating that the autocatalytic ethylene production was regulated independently of the petal wilting reaction. This result further suggests that these lines have normal ethylene receptors and produce ethylene autocatalytically, but their petal wilting reaction stops at a point beyond the receptor. Two lines (118–64S and 120–69S) showed extremely low ethylene production at flower senescence and very low autocatalytic ethylene production from petals and gynoecia.  相似文献

We investigated whether high‐quality in vitro matured (IVM) oocytes can be distinguished from poor ones based on the morphological changes after treatment with hyperosmotic medium containing 0.2 mol/L sucrose in pigs. We hypothesize that IVM oocytes maintaining round shape have higher quality than mis‐shapened oocytes following dehydration. Oocyte quality was verified by determining embryonic developmental competence using in vitro fertilization, nuclear transfer and parthenogenetic activation. In all cases, the round oocytes had greater (p < .05) developmental competence than that of mis‐shapened oocytes in terms of blastocyst rate and total cell number in blastocysts obtained after 6 days of in vitro culture. We also confirm that round aged oocytes are higher in quality than mis‐shapened aged oocytes. In an attempt to find out why high‐quality oocytes maintain a round shape whereas poorer oocytes become mis‐shapened following sucrose treatment, we examined the arrangement of actin microfilaments and microtubules. Abnormal organization of these cytoskeletal components was higher (< .05) in mis‐shapened oocytes compared to round oocytes after 52 hr of IVM. In conclusion, sucrose treatment helps selection of high‐quality oocytes, including aged oocytes, in pigs. Abnormal cytoskeleton arrangements partly explain for low developmental competence of mis‐shapened oocytes.  相似文献

ORF69 in the cyanophage infecting Microcystis aeruginosa, Ma-LMM01, shows homology to the family 19 chitinases where the catalytic domain has structural similarity to lysozyme. Chitinases hydrolyze chitin, a β-1, 4-linked monopolymer of N-acetylglucosamine (GlcNAc); whereas lysozymes hydrolyzes peptidoglycan, alternating β-1, 4-linked copolymers of N-acetylmuramic acid (MurNAc) and GlcNAc. Using amino acid sequence comparison to ORF69, the putative sugar binding residues, Gln162 and Lys165, from the barley chitinase (the model enzyme for the family 19 chitinases) corresponding to subsites −4 and −3 were found to be replaced with Gly209 and Ile213, respectively, in ORF69. To analyze their contribution to substrate binding affinity, ORF69 was cloned into Escherichia coli; and two mutant proteins G209Q and I213K were prepared using site-directed mutagenesis. The wild-type gene product (gp69) showed both lysozyme and chitinase activities. In contrast, the I213K mutant showed a decrease (70%) in lysozyme activity and a significant increase (50%) in chitinase activity; whereas, the G209Q mutant almost completely abolished both enzyme activities. The data suggest the Ile213 residue is involved in recognizing the substrate MurNAc; and Gly209 has significant contribution in chitinase and lysozyme activities for the wild-type gp69.  相似文献

The yield-per-recruit (YPR) analysis has been used to assess growth overfishing of exploited fish stocks. Since the ex-vessel price per weight widely depends on the size of the fish, the result of the YPR does not imply the revenue. The value-per-recruit (VPR) analysis, which is an economic expansion of the YPR analysis, was conducted for bighand thornyhead Sebastolobus macrochir caught off the Pacific coast of northern Honshu, Japan. The results showed that, for the current age at first capture (t c = 3), the current fishing mortality (F = 0.12) was close to the optimum level. However, for the current F, optimum t c in the VPR model was 8 years and would lead to a 47% increase compared to the current revenue. The maximum YPR was obtained at t c = 6 years, but the revenue at t c = 6 only gave a 20% increase in the maximum revenue. The difference was caused by the size dependency of the ex-vessel price per weight. The VPR model would generate fisheries management plans with both economical and biological consideration and would help in making general consensus of fisheries management strategies.  相似文献

Saddleback-like syndrome is one of the most frequently observed malformations in hatchery-reared red spotted grouper Epinephelus akaara. Saddleback-like syndrome is characterized by a V-shaped dorsal-profile depression behind the head region accompanied by deformities or misalignments of the first to fourth neural spines on the vertebral column and the first to fourth pterygiophores of the dorsal fin. Abnormal swim bladder inflation in several finfish species leads to malformations, particularly of the vertebral column. The present study examined the influence of swim bladder inflation on saddleback-like syndrome in hatchery-reared red spotted grouper. The incidence of saddleback-like syndrome increased as swim bladder inflation rate increased. Additionally, the incidence of saddleback-like syndrome among juveniles with inflated swim bladders was significantly higher than that among juveniles with uninflated swim bladders. The larval swim bladder is located just below the first to fifth vertebrae and the first to fifth dorsal spines, where saddleback-like syndrome occurs. Our results suggest that an inflated swim bladder increases the risk of saddleback-like syndrome in hatchery-reared red spotted grouper.  相似文献

Occurrence of pale potato cyst nematode, Globodera pallida (Stone) Behrens, was first recorded in Japan in 2015. Among several control measures, cultivation of resistant potato (Solanum tuberosum L.) varieties is the most effective in cost and environmental impact. As no G. pallida-resistant varieties have yet been developed in Japan, great emphasis is being placed on screening of germplasm possessing the resistance and development of the resistant varieties. In this study, we first improved previously reported DNA markers linked to the G. pallida resistance loci (GpaIVsadg and Gpa5) and then used these to screen more than 1,000 germplasms to select several candidate germplasms with resistance. We performed inoculation testing on the selected candidates and identified several resistant germplasms to the Japanese G. pallida population. Furthermore, we developed a simultaneous detection method combining three DNA markers linked to G. pallida and Globodera rostochiensis (Wollenweber) Behrens resistance loci. We validated the ability of C237-I marker to select resistant allele of GpaIVsadg and predict the presence of resistance in a Japanese breeding population. Resistant germplasms identified in this study could potentially be used to develop G. pallida-resistant varieties. The marker evaluation methods developed in this study will contribute to the efficient development of resistant varieties.  相似文献
