Introduction

To make clear the chemical behavior of free iron compounds in paddy soils, it is necessary to find an accurate and rapid method for determining ferrous iron in waterlogged paddy soils. Several methods1,2,3) have been proposed for this purpose, most of which use dilute acids, such as sulfuric and hydrochloric acid, as extracting reagents. According to the writers' experience, however, these methods seem to be inadequate. The reason is that the acid solutions extract not only ferrous iron, but also ferric iron and reducing substances from soils, and the latter two react immediately in the extract to produce ferrous iron, thus we obtain larger value for ferrous iron than the one actually existing in soils. The writers have found that acetate buffer of pH 2.8 is a very suitable extractant for ferrous iron, and they have been able to establish a new method for the determination of ferrous iron in soils using this buffer. We evaluated the postsurgical outcomes of cutaneous or subcutaneous mast cell tumors (MCTs, n=25) in 23 dogs, resected with lateral surgical margins proportional to the widest tumor diameter, including at least one facial plane. The tumor diameter range was 0.3–2.6 cm (median: 0.9 cm), and all were histologically diagnosed as Kiupel’s low-grade MCT. Resection was histologically complete in 20, close (deep margin) in three, and incomplete (deep margin) in two. No dogs developed local recurrence at the site of initial surgery during follow-up of 161–2,219 days (median: 976 days). These results suggest that resection of low-grade, relatively small MCTs with surgical margins proportional to the tumor diameter is a practical procedure with high success rates.  相似文献

Rice bran is abundant in bioactive compounds including tocotrienol (T3, unsaturated vitamin E). T3 has been reported about its potential functionalities (i.e., antiangiogenic effect), so much attention has been paid on usability of rice bran T3. Hence, we developed a rapid screening method for T3-rich rice bran by one-step equilibrium direct solvent extraction followed by normal phase high-performance liquid chromatography (HPLC). The method gave high-extraction rate of rice bran T3 and tocopherol (above 90%), and the determination of vitamin E by HPLC was completed within 15 min. Using the method, an average of total T3 content in 109 kinds of rice bran samples was 830 mug/g dry wt. Kouchi-Akamai, Joushuu, and Wataribune were found as the T3-rich rice bran varieties (1350-1430 microg T3/g dry wt). According to T3 ratio against total vitamin E (wt %), the average ratio was 61%. Hirayama, Moritawase, and Kaneko were found as the varieties having the highest T3 ratio (80-86%). Since T3 content in Koshihikari rice bran (the leading variety in Japan) was a little above the average, we cross-fertilized Koshihikari with T3-rich varieties and found that T3 content or ratio in F1 was improved compared with Koshihikari. The varieties found rich in T3 could be used for nutraceutical purpose.  相似文献

With the increased incidence of lifestyle-related diseases, prevalence of nonalcoholic fatty liver disease (NAFLD) is increasing at an unprecedented rate, leading to rising interest in studying the role of functional foods in its prevention. This study demonstrated the efficacy of salmon milt deoxyribonucleic acid (SM DNA) in preventing liver dysfunction in a diet-induced obesity model. Seven-week-old male C57BL/6J mice were fed normal diet (ND), high-fat diet (HFD), and HFD supplemented with 3% SM DNA (HFD?+?SM DNA) for 12 weeks. Increased body weight, fat-to-body weight ratio, and fat accumulation in HFD-fed mice demonstrated successful establishment of an obesity model. The HFD group showed increased alanine aminotransferase (ALT) levels, a marker of liver function, and NAFLD activity score (NAS) based on histological observations showed steatosis and mild inflammation but no fibrosis, indicating NAFLD but no nonalcoholic steatohepatitis. In the HFD?+?SM DNA group, the improved superoxide dismutase activity and NAFLD activity score suggested that SM DNA prevented liver dysfunction while maintaining antioxidant capacity and suppressing intrahepatocyte fat droplet formation. SM DNA attenuated fat accumulation in the liver and improved hepatic function. This study explored the potential of SM DNA as a functional food to prevent NAFLD and other visceral obesity-related diseases.

Microstructures of lapilli were examined for reared larvae and juveniles of black-spot tuskfish Choerodon schoenleinii. Lapilli of larvae at 1 day after hatching have one diffuse and obscure ring with an otolith radius of 4.3 ± 0.50 μm (mean ± SD, N = 8). The slope and intercept of the regression between the number of days after hatching and increment counts did not differ significantly from one and zero, respectively, indicating that lapillus increments were formed on a daily basis after hatching. There was an ontogenetic shift in the relative values of somatic and otolith growth, which corresponded to the transition from pelagic larvae to settlement stage. Simultaneously, the daily increment width reached the maximum value. These findings suggest that age at maximum value of increment width can be used as an indicator of the planktonic larval duration while settlement mark is not found. Since ontogenetic shift in the relationship between otolith radius and body size was observed, back-calculation of somatic growth in black-spot tuskfish using the otolith radius during the early life stages should be analyzed with caution, and the method requires further validation.  相似文献

For the purpose of investigation of working mechanisms in endocrine disruptors, we evaluated the dose-related effects of fetal and/or neonatal exposure to an estrogenic compound on the male reproductive organs in adult mice, particularly with respect to gene expression of steroidogenic acute regulatory protein (StAR). The pregnant ICR mice were given subcutaneous injections of 10 micro g/day/animal of diethylstilbestrol (DES) to subject the fetal mice to in utero exposure (IUE). Subsequently, the newborn male mice were subjected to neonatal exposure (NE) by treatment with vehicle or 0.1-10 micro g/day/animal of DES. Fertility rates of each group were as follows: control, 100%; IUE only, 60%; IUE+NE 0.1 micro g, 25%; IUE+NE 1 micro g, 0%; IUE+NE 10 micro g, 0%. In general histology, germ cell layers in the seminiferous tubules were thinned in the group of IUE+NE 10 micro g. Hypoplasia of the Leydig cells, in which the staining intensity of eosin was diminished, was also observed in the groups of IUE+NE 0.1-10 micro g. The androgen receptor (AR) and estrogen receptor alpha (ERalpha) immunoexpression in the Leydig cells of IUE+NE 1-10 micro g was slightly lower than that in the controls. Long-term dysfunction of the hypothalamo-pituitary-testicular axis, including sustained hypoproduction of gonadotropin and testosterone, and altered expressions of steroid hormone receptors and StAR genes were observed. The hypothalamo-pituitary control of gonadotropin secretion may be affected by the smaller doses of estrogenic agents than the reproductive organs. Furthermore, the fertility rate in the male mice exposed to this estrogenic agent was closely correlated with the testosterone levels, and even more so with the rate-limiting factor of steroidogenesis, StAR. This finding suggests that endocrine disruptors have an important pronounced effect on StAR gene expression.  相似文献

Poly(N-vinylacetamide-co-acrylic acid) coupled with d-octaarginine (VP-R8) promotes the cellular uptake of peptides/proteins in vitro; however, details of the transfection efficacy of VP-R8, such as the cell types possessing high gene transfer, are not known. Herein, we compared the ability of VP-R8 to induce the cellular uptake of plasmid DNA in mouse and human cell lines from different tissues and organs. A green fluorescent protein (GFP)-expression plasmid was used as model genetic material, and fluorescence as an indicator of uptake and plasmid-derived protein expression. Three mouse and three human cell lines were incubated with a mixture of plasmid and VP-R8, and fluorescence analysis were performed two days after transfection. To confirm stable transgene expression, we performed drug selection three days after transfection. A commercially available polymer-based DNA transfection reagent (PTR) was used as the transfection control and standard for comparing transgene expression efficiency. In the case of transient transgene expression, slight-to-moderate GFP expression was observed in all cell lines transfected with plasmid via VP-R8; however, transfection efficiency was lower than using the PTR for gene delivery. In the case of stable transgene expression, VP-R8 promoted drug-resistance acquisition more efficiently than the PTR did. Cells that developed drug resistance after VP-R8-mediated gene transfection expressed GFP more efficiently than cells that developed drug resistance after transfection with the PTR. Thus, VP-R8 shows potential as an in vitro or ex vivo nonviral transfection tool for generating cell lines with stable transgene expression.  相似文献
