During the last 15 years a number of studies have shown increasing forest growth in central Europe, rather than a decline as was expected due to negative effects of air pollution. We have here used data from intensive monitoring plots spread over Europe for a five year period in order to examine the influence of environmental factors on forest growth. Evaluations focussed on the influence of nitrogen, sulphur and acid deposition, temperatures, precipitation and on a drought index calculated as deviation from the long-term mean. The study included the main tree species Norway spruce, Scots pine, common beech as well as European and sessile oak and was based on data from 363 plots. As many other factors besides nitrogen and temperature influence tree growth, expected stem volume increments were modelled using site productivity, stand age and a stand density index. Relative volume increment was then calculated as actual increment in % of expected increment. The site productivity, assumed to be given by site conditions and past environmental conditions, was either taken from expert estimates or computed from site index curves from northern, central and southern Europe. The model explained between 18% and 39% of the variance with site productivity being positively related and age negatively related to actual increment. The various models and statistical approaches were fairly consistent, and indicated a fertilizing effect of nitrogen deposition, with slightly above one percent increase in volume increment per kg of nitrogen deposition per ha and year. This was most clear for spruce and pine, and most pronounced for plots having soil C/N ratios above 25. Also, we found a positive relationship between relative increment and summer temperature, i.e. May–August mean temperature deviation from the 1961–1990 means. The cause–effect relationship here is, however, less certain. Other influences were uncertain. Possibly, sulphur and acid deposition have effects on growth, but these effects are obscured by, and outweighed by the positive effect of nitrogen deposition, because of collinearity between these variables. Drought effects were uncertain also, and one reason for this might be large uncertainties in the precipitation data: precipitation measured on some 50% of the plots correlated poorly with the precipitation data obtained from Europe-wide databases. Helleborus species are members of the family of the Ranunculaceae. These popular perennials are all diploids (2n = 2x = 32). This study investigates polyploidy induction by different antimitotic agents. Colchicine, oryzalin and trifluralin
were tested in vitro on shoots of Helleborus niger, H. orientalis and H. × nigercors. Furthermore the effect of the antimitotic agents on the viability and the multiplication rate of cultured plantlets were
analyzed. Flow cytometry demonstrated that polyploidisation was genotype dependent: using H. niger, tetraploids were obtained using either oryzalin (3 μM) or trifluralin (3 or 10 μM), whereas for H. × nigercors only trifluralin (3 or 10 μM) induced polyploidisation. For H. orientalis neither treatment was effective to produce tetraploids or mixoploids. For these three species, colchicine (100 μM) was ineffective.
The polyploidisation events in H. niger and H. × nigercors were confirmed by chromosome counts of mounted nuclei derived from root tips (2n = 4x = 64). 相似文献

The high molecular weight subunits of wheat glutenin (HMW-GS) are important for bread-making quality. Their composition is routinely identified by Tris-glycine SDS-PAGE after reduction of glutenin disulfide bonds. However, the relation between their molecular weight and, hence, their primary structure, and their mobility in Tris-glycine SDS-PAGE has proven to be ambiguous. We demonstrate a Bis-Tris SDS-PAGE procedure with a neutral, instead of alkaline, pH in the gel and running buffers. In this method commonly occurring HMW-GS from wheat migrated in the order 5 > 2 ≈ 3 > 1 > 6 ≈ 2* > 7 > 8 > 9 > 12 > 10, which is different from the order obtained in the Tris-glycine system. HMW-GS were identified by N-terminal sequencing after isolation with RP-HPLC. Protein sequences of HMW-GS were further confirmed by LC-MS/MS analyses of chymotryptic peptides after comparing the MS data to amino acid sequences in protein databases. The numbers of amino acids of HMW-GS reflected well the mobility order in Bis-Tris SDS-PAGE. The results indicate that Bis-Tris SDS-PAGE may not only be used to identify HMW-GS, but also to estimate the length of their polypeptide chain, as such avoiding previously observed anomalies in migration order. 相似文献

This study investigated the effect of balanced, nonpersuasive information related to safety, healthiness, and sustainability of aquaculture on the image of farmed fish among European consumers. It was demonstrated that there is neither positive nor negative influence of this type of information on the predominantly positive image of fish from aquaculture. Consumers who were exposed to information related to the EU origin and aquaculture related guarantee reported a more positive image of farmed fish. These results provide valuable input for transparent communication about fish farming practices which might increase consumer trust and will not harm the image of fish from aquaculture. Increasing consumers' knowledge about aquaculture and its positive effects on the image of farmed fish is also discussed as a pathway for maintaining and improving aquaculture's positive image in the future. 相似文献

A plastic-like material can be obtained by thermomolding wheat gluten protein which consists of glutenin and gliadin. We studied the effect of molding temperature (130-170 °C), molding time (5-25 min) and initial wheat gluten moisture content (5.6-18.0%) on the gluten network. Almost no glutenins were extractable after thermomolding irrespective of the molding conditions. At the lowest molding temperature, the extractable gliadin content decreased with increasing molding times and moisture contents. This effect was more pronounced for the α- and γ-gliadins than for the ω-gliadins. Protein extractabilities under reducing conditions revealed that, at this molding temperature, the cross-linking was predominantly based on disulfide bonds. At higher molding temperatures, also non-disulfide bonds contributed to the gluten network. Decreasing cystine contents and increasing free sulfhydryl and dehydroalanine (DHA) contents with increasing molding temperatures and times revealed the occurrence of β-elimination reactions during thermomolding. Under the experimental conditions, the DHA derived cross-link lanthionine (LAN) was detected in all gluten samples thermomolded at 150 and 170 °C. LAN was also formed at 130 °C for gluten samples containing 18.0% moisture. Degradation was observed at 150 °C for samples thermomolded from gluten with 18.0% moisture content or thermomolded at 170 °C for all moisture contents. 相似文献

Pangasius hypophthalmus (referred to as Pangasius or tra fish in Vietnam) has become highly appreciated by consumers in the European Union, USA, Japan, etc. and is of worldwide economic importance. Therefore, the microbial quality and safety of Pangasius fish processed for export was assessed by means of a microbial assessment scheme throughout the entire production process. A total of 144 samples were collected from various processing steps and analyzed. It was determined that the microbial safety and quality of the products was not guaranteed as the contamination levels remained high throughout processing. Escherichia coli, Staphylococcus aureus and Vibrio cholerae were present on the hands of food operators, particularly those in the packaging area. Moreover, the presence of Listeria monocytogenes (1 positive out of 9 samples) and V. cholerae (4/9) on the final products was likely a result of inadequate hygiene practices in the processing environment. Also discussed in this paper are the results of a self-assessment questionnaire, which provide insight into the performance of the food safety management system currently implemented at the company. These data are of major importance in order to contribute valuable information to the local and international trade point of view in general and to the intended customers in particular. 相似文献

In Africa, indigenous multimammate mice (Mastomys natalensis) only appear to live commensally in houses when invasive black rats (Rattus rattus) are absent, yet little is known about the underlying mechanism. Avoidance through smell may cause the absence of M. natalensis from areas occupied by R. rattus, but this hypothesis has not yet been tested. We conducted a Y-maze choice experiment where 15 M. natalensis were offered a choice between corridors containing conspecific scent, R. rattus scent and a control scent. Residence time in the R. rattus corridor was greater than that in the control corridor but equal to that in the M. natalensis corridor, suggesting that multimammate mice do not actively avoid the scent of their invasive competitor. 相似文献

A formerly unknown nucleotide pyrophosphatase/phosphodiesterase (NPP) of wheat (Triticum aestivum L.) (TaNPP) was recombinantly produced in Pichia pastoris (TaNPPr) following assembly of the sequence out of wheat ESTs using sequences of known NPPs. Simultaneously, a phosphodiesterase was purified to homogeneity from wheat germs, characterised and identified as TaNPP. TaNPP contains the highly conserved catalytic substrate and metal binding residues and displays properties [high pH optimum, glycosylation, high thermostability and inhibition by EDTA and adenosine 5′-monophosphate (AMP)] similar to those of mammalian enzymes characterised earlier. The sequence of TaNPP includes that of a putative transmembrane domain, a characteristic of most NPPs. Both recombinant and native TaNPP partially occur as oligomeric proteins on SDS PAGE. Upon addition of DTT, both migrate as monomeric proteins. Part of the native wheat protein even occurs in its truncated form, thus lacking the transmembrane region. Out of a range of tested natural substrates, TaNPP had the highest affinity towards adenine containing nucleotides. While Michaelis-Menten kinetics were valid in the low substrate concentration range, at higher concentrations, they were no longer applicable. TaNPP shows no similarity to the recently characterised rice and barley enzymes and, is hence, one of the first characterised plant NPPs resembling mammalian NPPs. 相似文献

The aim of this study was to increase insight into gluten polymerisation. While previous research on this topic focused on disulfide (SS) bonds, the present paper focuses on cross-links based on dehydro-protein formation through β-elimination reactions. Gliadin, the monomeric fraction of gluten containing no free sulfhydryl (SH) groups, was heated for 120 min at pH 8.0 and 130 °C, and cross-link formation was evaluated by determining extractability in sodium dodecyl sulfate containing buffer, reaction products of β-elimination reactions, and cross-links involving the latter. Heating decreased gliadin extractability. Reduction of SS bonds increased extractability of heated gliadin, but did not restore it to that of non-heated gliadin, suggesting contribution of both SS and non-SS bonds to gliadin cross-linking. Decreased SS levels and the presence of dehydroalanine and SH groups in heated gliadin samples indicated cleavage of SS bonds by β-elimination reactions. Some of the formed free SH groups were then involved in oxidation and/or SH–SS interchange reactions leading to intermolecular SS bonds. In addition, amino acid analysis revealed formation of an irreversible non-SS cross-link between dehydroalanine and the free SH group of cysteine, namely lanthionine. In conclusion, non-SS bonds may well contribute to the gluten network under specified reaction conditions. 相似文献
