PREAMBLE According to the German Federal Soil Protection Act (BBodSchG 1998), the habitat function of soils must be protected. Despite the fact that in the Federal Soil Protection Ordinance (BBodSchV 1999) it has not been established how this goal can be reached reliably, it is clear that such a biological function can only adequately be assessed using biological test methods. This is especially true when a soil is contaminated by a mixture of often unknown chemicals. In such a case the use of chemical analysis aiming at a small range of known substances is not sufficient and must therefore be supplemented by biological methods. For this reason, several standardised test methods are available (e.g. using earthworms, collembolans or plants; Römbke and Knacker 2003; ISO 2003). Since acute tests are usually not sensitive enough for the assessment of potentially contaminated soils (e.g. Hund-Rinke et al. 2002), chronic tests like the earthworm reproduction tests (ISO 1998) are recommended for this purpose.

A chronic plant test for the determination of phytotoxicity was missing until quite recently. The term phytotoxicity is understood here as the capacity of a compound or a contaminated soil to cause temporary or long-lasting damage to plants (EPPO 1997). Therefore, the German Ministry for Education and Research sponsored a project (1997 – 1999) in which – based on existing standardised methods – such a chronic plant laboratory test was developed and partly validated (Kalsch and Römbke 2000). The new test can be used for the evaluation of single chemicals (see Part 1 of this mini-series) as well as for the assessment of contaminated or remediated soils (see Part 2 of this mini-series).

ABSTRACT Background and Scope. A new chronic plant test system which is based on experiences with various acute plant tests (e.g. published by OECD or ISO) and existing North American Plant-Life-Cycle Bioassays was standardised in a project sponsored by the German government. Characteristic properties of the test system, which can be performed either with Brassica rapa (turnip rape) or Avena sativa (oat), are described in Part I of this mini-series.

Methods

This new test was used to assess the effects of natural soil samples contaminated with TNT (2,4,6-Trinitrotoluene) or PAHs (poly-aromatic hydrocarbons). The soils were tested after taken from the field as well as after being remediated. Different control and reference soils were used to evaluate the test results. In addition, they were compared with the results of tests in which either TNT or Pyrene was spiked to field and standard soils (see Part I of this mini-series).

Results

All contaminated soils showed clear effects in the Chronic Plant Test (usually B. rapa was more sensitive than A. sativa). LUFA 2.2 standard soil and OECD artificial soil are well-suited as control and mixture substrates, while reference soils collected at uncontaminated sites were several times phytotoxic. In most of the latter cases, soil properties could be identified as the main cause of these effects (e.g. the pH value). While in general the sensitivity of the reproduction and biomass endpoints did not differ much, it is recommended to measure different endpoints (i.e. biomass and reproduction) due to the different mode-of-action of contaminants. In the case of TNT a good agreement between the results of single chemical tests and tests with TNT-contaminated soils was found (due to the minimal effects of Pyrene the same statement is not possible for PAHs).

Outlook

The results of this work have been used to prepare a draft test guideline, which has recently been standardised by the International Organisation for Standardisation (ISO). Based on these experiences, recommendations are given how this chronic test system could be used as part of a test battery for the assessment of contaminated soils.  相似文献

In cats, primary or secondary immune-mediated thrombocytopenia have rarely been described or characterised. The objective of this study was to determine platelet-bound antibodies (PBA) by a flow cytometric assay in both healthy and thrombocytopenic cats. Direct PBA testing was performed in 42 thrombocytopenic cats (platelet counts 6-179 x 10(9)/l, median 56 x 10(9)/l). Of these 42 cats, 19 had positive PBA test results, 17 of which were considered to have secondary immune-mediated thrombocytopenia (sITP). Underlying diseases included fat necroses (four cases), feline infectious peritonitis (three), feline leukaemia virus (two) or feline immunodeficiency virus (two) infections, lymphoma (two), leukaemia (one), hepatitis (one), pyelonephritis (one), or hyperthyroidism (one). In two cats, no underlying disease was found suggesting a primary immune-mediated thrombocytopenia (pITP). The PBA test was negative in 23 cats diagnosed with varying underlying diseases and in 47 healthy control cats with platelet values within the reference range. Only seven of the 42 cats with thrombocytopenia (platelet count 10-57 x 10(9)/l, median 34 x 10(9)/l) had spontaneous bleeding. This study suggests that immune-mediated destruction of platelets might be an important pathological mechanism for feline thrombocytopenia caused by various underlying diseases. In cats, pITP appears to be rarely diagnosed.  相似文献

Concentration of airborne endotoxins, airborne aerobic bacteria and airborne aerobic gram-negative bacteria were measured in 3 rabbit houses. Further, the species composition of the airborne gram-negative bacterial flora was investigated. The total amount of airborne endotoxin ranged from 22 to 774 EU/m3 (Endotoxin Units/m3). The number of total airborne aerobic bacteria varied between 780 and 20100 CFU/m3, the number of airborne aerobic gram-negative bacteria between 39 and 1030 CFU/m3. Most gram-negative bacterial isolates belonged to the family Enterobacteriaceae with E. coli as primary species. In two rabbit houses also airborne Pasteurella multocida spp. multocida, the most common respiratory pathogen of rabbits, was isolated.  相似文献

The use of urea as nitrogen (N) fertilizer in agriculture needs to consider environmental, economic and resource conservation aspects because of low N-use efficiency (NUE). Polymer-coated urea (PCU) offers an effective way to improve the NUE of urea and to reduce its environmental trade-offs. However, we lack information on the impact of climate and soil properties on N release from PCU. Therefore, this study was performed to quantify the effects of soil texture, moisture and temperature on the release kinetics of N from PCU. We designed a test system for soil incubation experiments and investigated three fertilizers with different release patterns, five topsoils, three moisture levels and two temperatures over 48 days. We analysed the concentrations of inorganic N ( NH 4 + N and NO 3 N ) in the soil and estimated N release rates using the unified Richards model. Soil texture did not change the N release patterns, but release rates varied significantly among the investigated soils. Changes in soil moisture for a given soil had no effect on N release from PCU and urea when fertilizers were incorporated into the soil at conditions supportive of crop growth. Lowering soil temperatures, however, decreased N release rates from PCU by 16%–49% but only in silt loam and not in sandy loam. We conclude that PCU improves the N residence time in soil, but predictions on N release from PCU must be adapted to the prevailing environmental conditions and cannot be generalized across differently textured soils.  相似文献

The use of biochar is expected to improve soil fertility and close nutrient cycles in degraded strongly weathered tropical soils. We, therefore, hypothesized that biochar amendment to tree plantations (a) increases nutrient fluxes with litterfall alone and with mineral fertilizer plus lime and (b) reduces N losses reflected by lower δ15N values of litterfall and soils than in unamended controls. We grew the native leguminous Schizolobium parahyba var. amazonicum (Ducke) Barneby and the exotic Gmelina arborea Roxb at two sites. We used a replicated full factorial split–split plot design of amendment of mineral fertilizer plus lime, 3 and 6 t ha−1 biochar, and a control. We collected litterfall biweekly (2012–2013) and topsoil samples (0–0.25 m) in 2009 before tree planting, in 2011 and 2013. Fertilizer plus lime increased the mean annual concentrations of P, Ca and Zn in litterfall but decreased that of Mn. At the same time, fertilizer plus lime increased the annual fluxes of nutrients, Na and Al with litterfall. During the dry season, biochar decreased the N concentration in litterfall and the K flux with litterfall. During the rainy season, biochar increased the concentrations of Ca and Zn in litterfall and their fluxes with litterfall. Biochar did not influence the δ15N values of soil and litterfall after 51 months of tree growth. Fertilizer plus lime decreased the δ15N values of soil, because of the lower δ15N value of the used urea (−0.30‰) than the soil (4.5‰–7.8‰). Moreover, fertilizer plus lime increased the δ15N values of litterfall, possibly because of enhanced 14N leaching from the N-rich canopies. The amendment of up to 6 t ha−1 biochar did not contribute to close nutrient cycles.  相似文献
