To produce 5-reductase inhibitory compounds, resveratrol was enzymatically oxidized in a horseradish peroxidase (HRP)/H2O2 system. Ethyl acetate extract of the oxidation products showed strong 5-reductase inhibitory activity with 10%–15% organic solvents in the system, whereas without organic solvent little inhibitory activity was exhibited. The optimum pH of enzymatic oxidation for acquisition of the inhibitory activity was 4.5. The inhibitory compounds were isolated and identified as resveratroltrans-dehydrodimer and resveratrolcis-dehydrodimer by comparing with published nuclear magnetic resonance data. The two resveratrol dehydrodimers have stronger inhibitory activity than natural resveratrol dimers and trimers found inShorea species. 相似文献

Conclusions When the hydroxyl values of materials containing phenolic 3 or sterically hindered hydroxyl groups were determined by the esterification method, the values were considerably smaller than the theoretical values. In contrast, the NCO method provided hydroxyl values much closer to the theoretical values for such materials, though the values varied depending on the kinds and amounts of catalysts.The quantity of hydroxyl groups reacting with the NCO group could be directly determined by the NCO method using a catalyst mixture consisting of DABCO and DBTDL in a molar ratio of 21. The morphological features and dietary functional components of two strains (FPF-13 and Oninome-B) of Pholiota adiposa (numerisugitake mushroom) grown on artificial bed blocks were examined. The components examined were chitin, mannitol, trehalose,
guanosine 5′-monophosphate, ergosterol, and β-glucan. There was a significant difference in the external shape of the two strains. However, the differences in the contents
of functional components between the two strains, as well as between the pilei and stipes of the strains, were small. In both
strains the trehalose content was much higher than the mannitol content, in contrast to those of Lentinula edodes. From a commercial point of view, Oninome-B has a clear advantage over other strains of P. adiposa owing to its less removable scale.

The optimum conditions for β-thujaplicin production in a Cupressus lusitanica cell suspension culture were investigated. The conditions required for β-thujaplicin production were clearly different from the conditions for cell growth. The initial phosphate concentration and
pH did not affect β-thujaplicin production. A total nitrogen source concentration higher than 3.2 mM suppressed production due to the presence
of the ammonium ion. β-Thujaplicin production was observed at 95 mg/l without adding the ammonium ion to the medium. Strict control of major inorganic
nutrients was not necessary to produce β-thujaplicin. This finding seems to be favorable for future automated production of β-thujaplicin in commercial cell culture plants.

Heavy thinning may provide a feasible means to convert single-species coniferous plantations to broadleaved or mixed forests. To assess this possibility, we monitored tree seedling dynamics of other tree species, the understory microenvironment, and seed availability following heavy thinning (50 % by volume) in two hinoki (Chamaecyparis obtusa) plantations in Shikoku, southwestern Japan. Conventional (35 % by volume) and row thinning (50 % by volume) treatments were also applied at one of the sites. Diffuse transmittance in the understory dramatically increased after all thinning treatments then rapidly decreased, except after row thinning. Heavy thinning accelerated recruitment of small seedlings (with stems <30 cm high) at one of the sites. However, the densities of these seedlings declined sharply, and increases in the densities of larger seedlings recorded in subsequent surveys were very small. Furthermore, at the other site, recruitment of small seedlings under heavy thinning was equivalent to that under conventional thinning and lower than that under row thinning. Abundant species in the recruited seedlings were pioneer or subcanopy species found in the soil seed bank. Low seed rain density at both sites appeared to be responsible for the poor recruitment of canopy tree species. Our results suggest that heavy thinning had limited effects on the seedling recruitment of other tree species in these hinoki plantations due to insufficient seed rain and rapid canopy closure. 相似文献

Manganese peroxidase (MnP) production by Phlebia sp. MG-60 is strongly regulated by Mn2+ and NH4+ at various sea salt incubation conditions. Extra-added Mn2+ and NH4+ obviously inhibited MnP production, but sea salts relieved the inhibition partially or completely. Three media were prepared: Kirk medium with addition of 0%–5% sea salts (KS medium), a high level of Mn2+ (300mg/l) in KS medium (HM-KS medium), and a high level of NH4+ (430mg/l) in KS medium (HN-KS medium). Without addition of sea salts, the dye Poly R-478 was significantly decolorized by low MnP activity (about 200U/l) and a low level of laccase activity (about 100U/l) in KS and HM-KS media. In the cultures in which laccase activity was almost completely inhibited by 3% and 5% sea salts, MnP activity higher than 400U/l was necessary for Poly R-478 decolorization in all of the three media. Iron-binding compounds were isolated from a culture ofPhanerochaete sordida YK-624 and were found to bind to Fe(III) preferentially compared with Fe(II). Two iron-binding compounds were purified to near-homogeneity with gel permeation chromatography. Hydrolysis of the iron-binding compounds with 6N hydrochloric acid gave ninhydrin-negative products. The molecular weight of these compounds was 3–5 kDa. These compounds may play an important role in the reduction of extracellular manganese dioxide to Mn(II) by intracellular ferrireductases for lignin degradation by manganese peroxidase. 相似文献

The impregnation of various simple phenolic and natural polycyclic compounds into wood was investigated from the viewpoints of vibrational property and dimensional stabilizing effect. When simple phenolic compounds were impregnated, the loss tangent (tan ) in the longitudinal direction increased linearly with increasing weight gain. Meanwhile, among the natural polycyclic compounds hematoxylin decreased the tan drastically by impregnation. It was suggested that the five hydroxyl groups and the pyran ring oxygen in the hematoxylin molecule contribute to formation of the crosslinkage-type hydrogen bonds between wood components. The rigidity of hematoxylin molecules may also be important. By impregnation of about 10% catechol, resorcinol, and saligenin, a 40% level of antiswelling efficiency (ASE) was attained, although a significant dimensional stabilizing effect was not observed after impregnation of natural polycyclic compounds.Part of this work has been published as a Rapid Communication inMokuzai Gakkaishi 43(12). Antioxidant activity of carotenoids is suggested to be one of the factors for their disease preventing effects. Marine carotenoids fucoxanthin and its two metabolites, fucoxanthinol and halocynthiaxanthin, have been shown to exhibit several biological effects. The antioxidant activities of these three carotenoids were assessed in vitro with respect to radical scavenging and singlet oxygen quenching abilities. The 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity of fucoxanthin and fucoxanthinol was higher than that of halocynthiaxanthin, with the effective concentration for 50% scavenging (EC 50) being 164.60, 153.78, and 826.39 microM, respectively. 2,2'-Azinobis-3-ethylbenzo thizoline-6-sulphonate radical scavenging activity of fucoxanthinol (EC 50, 2.49 microM) was stronger than that of fucoxanthin (EC 50, 8.94 microM). Hydroxyl radical scavenging activity as measured by the chemiluminescence technique showed that the scavenging activity of fucoxanthin was 7.9 times higher than that by fucoxanthinol, 16.3 times higher than that by halocynthiaxanthin, and 13.5 times higher than that by alpha-tocopherol. A similar trend was observed when the hydroxyl radical scavenging was assessed by the electron spin resonance (ESR) technique. ESR analysis of the superoxide radical scavenging activity also showed the superiority of fucoxanthin over the other two carotenoids tested. Singlet oxygen quenching ability of the three carotenoids was lower than that of beta-carotene, with quenching rate constants ( k Q, x10 (10) M (-1) s (-1)) being 1.19, 1.81, 0.80, and 12.78 for fucoxanthin, fucoxanthinol, halocynthiaxanthin, and beta-carotene, respectively. The higher radical scavenging activity of fucoxanthin and fucoxanthinol compared with halocynthiaxanthin is assumed to be due to presence of the allenic bond. 相似文献
