A total of 205 cornflake samples collected in Belgian retail stores during 2003-2004 were surveyed for the natural occurrence of fumonisin B1 (FB1), B2 (FB2), and B3 (FB3). These cornflake samples, originating from conventional as well as from organic production, were analyzed using an intralaboratory-validated LC-MS/MS method. Additionally, 90 cornflake samples were subjected to rapid screening using a flow-through enzyme immunoassay method to demonstrate the practicability of a screening test coupled to a validated confirmatory LC-MS/MS method for the management of food safety risks. FB(1) concentrations ranged from not detected (nd) [LOD (FB1) = 20 microg/kg] to 464 microg/kg with mean and median concentrations of respectively 104 +/- 113 and 54 microg/kg. For FB2 and FB3, the concentration ranges varied respectively from nd [LOD (FB2) = 7.5 microg/kg] to 43 microg/kg and from nd [LOD (FB3) = 12.5 microg/kg] to 90 microg/kg. Mean concentrations for FB2 and FB3 were respectively 12 +/- 8 and 21 +/- 15 microg/kg, while the median concentration was 11 microg/kg for FB2 and 19 microg/kg for FB3. From the statistical tests (chi2 and ANOVA model III), it could be concluded that the agricultural practice did not have any significant effect on the fumonisin concentrations but that the variation between different batches was significant (p < 0.0001).  相似文献

The behaviour of earthworms belonging to two different species and ecological types (Aporrectodea nocturna and Allolobophora chlorotica) was studied using two-dimensional (2D) terraria. Two experiments were set up to gain insight into the nature of interactions between these earthworms. Firstly, the evolution of the burrow systems was analysed with the density of the earthworms varying from one to five individuals. Secondly, a burrow system was first established by using one earthworm which was then removed before the introduction of a second earthworm. This second earthworm therefore encountered a burrow system created either by a conspecific earthworm or by an earthworm of the other species. These experiments showed that: (1) intra- and interspecific interactions occur between earthworms, (2) these interactions are dependent on the physical presence of the earthworms, and (3) spatial avoidance can occur (A. chlorotica avoiding burrows created by A. nocturna). The results suggest that earthworm burrow systems are "individual structures", rarely used by other earthworms when inhabited. When abandoned, the burrows may be recolonised depending on the ecological type of the earthworm under consideration.  相似文献

Limb‐sparing for distal radial osteosarcoma has a high rate of complications. Using personalized three‐dimensional (3D)‐printed implants might improve outcome. The goals of this study were to optimize use of patient‐specific, 3D‐printed endoprostheses for limb‐sparing in dogs in the clinical environment and to report the outcome. This was a pilot study where five client‐owned dogs were enrolled. Computed tomography (CT) of the thoracic limbs was performed, which was used to create patient‐specific endoprostheses and cutting guides, and repeated on the day of surgery. Intra‐arterial (IA) carboplatin was introduced in the clinical management. Limb‐sparing was performed. Outcome measures were time required to produce the endoprosthesis and cutting guide, fit between cutting guide and endoprosthesis with host bones, gait analysis, size of the tumour, percent tumour necrosis, complications, disease‐free interval (DFI) and survival time (ST). Four dogs received IA carboplatin. Excessive tumour growth between planning CT and surgery did not occur in any dog. The interval between the CT and surgery ranged from 14 to 70 days. Fit between the cutting‐guide and endoprosthesis with the host bones was good to excellent. At least one complication occurred in all dogs. Two dogs were euthanized with STs of 192 and 531 days. The other dogs were alive with a follow up of 534 to 575 days. IA chemotherapy is a promising strategy to minimize the risk of excessive tumour growth while waiting for the endoprosthesis and cutting‐guide to be made. The design of the cutting‐guide was critical for best fit of the endoprosthesis with host bones.  相似文献

Multiple novel and prevalent astroviruses in pigs   总被引:3,自引:0,他引:3  
Knowledge of porcine astrovirus diversity and epidemiology remains limited. We used a broad range PCR approach to investigate the presence and diversity of astroviruses in healthy pigs of different ages on 20 farms and in 3 slaughterhouses situated in the province of Quebec, Canada between 2005 and 2007. Our study unexpectedly revealed remarkable levels of genetic diversity and high prevalence of astroviruses in pigs of this province. Astroviruses were detected on every farm investigated and in all age groups of pigs, from suckling piglets to adults. In addition, we found that nearly 80% of healthy finisher pigs harbour astroviruses in their intestine at slaughter. Phylogenetic evidence based on partial polymerase and complete capsid sequences, suggests that porcine astroviruses do not form a monophyletic group but are rather found on separate branches across the mamastrovirus tree. In addition to type species strains, we found highly divergent strains that form two additional lineages, one of which falls outside existing taxonomic groups. The presence of diverse astroviruses in a majority of healthy pigs likely represents a continuous source of infection to piglets and possibly to other animal species including humans. Porcine astrovirus strains appeared phylogenetically related not only to prototypical human astroviruses, as was already known, but also to novel human strains recently discovered suggesting multiple cross species transmission events between these hosts and other animal species. Overall, the findings reported in this study suggest an active role of pigs in the evolution and ecology of the Astroviridae.  相似文献

The objective of this study was to verify whether the volatile organic sulfur compounds recently discovered in bell pepper (Capsicum annuum, L. cultivars), such as the mercapto-ketones: 4-sulfanyl-2-heptanone and 2-sulfanyl-4-heptanone, the mercapto-alcohols: 4-sulfanyl-2-heptanol and 2-sulfanyl-4-heptanol, and heptane-2,4-dithiol, originate from their corresponding cysteine-S-conjugates. Analysis of aqueous extracts of red and green bell pepper by ultraperformance liquid chromatography-mass spectrometry with electrospray ionization in the positive mode (UPLC-MS ESI(+)) displayed masses corresponding to the expected cysteine-S-conjugates. To confirm this observation, four cysteine-S-conjugates were prepared as authentic samples: S-(3-hydroxy-1-methylhexyl)-L-cysteine, S-(3-hydroxy-1-propylbutyl)-L-cysteine, S-(3-oxo-1-propylbutyl)-L-cysteine, and (2R,2'R)-3,3'-(4-hydroxyheptane-2,6-diyl)bis(sulfanediyl) bis(2-aminopropanoic acid). By comparison with the fragmentation patterns and retention times of synthetic mixtures of cysteine-S-conjugate diastereoisomers, the natural occurrence of cysteine conjugates was confirmed in bell peppers. In addition, the cysteine-S-conjugates from red and green bell pepper extracts were concentrated by ion exchange chromatography and the fractions incubated with a β-lyase (apotryptophanase). The liberated thiols were concentrated by affinity chromatography, and their occurrence, detected by gas chromatography-mass spectrometry, confirmed our predictions. Moreover, 3-sulfanyl-1-hexanol was also detected and the occurrence of S-(1(2-hydroxyethyl)butyl)-L-cysteine confirmed. A quantitative estimation based on external calibration curves, established by UPLC-MS ESI(+) in selected reaction monitoring mode, showed that cysteine-S-conjugates were present at concentrations in the range of 1 to 100 μg/kg (±20%).  相似文献

A reduction in the numbers of macroinvertebrates present in soil may have a negative effect on soil structure, infiltration rates, and gas exchanges. Soil pollution by metal is known to have a detrimental effect on soil macrofauna. The aim of the present study was to evaluate (1) direct and indirect effects of soil pollution on soil macroinvertebrate bioturbation and (2) effects of the two macroinvertebrate communities found in a polluted and a nonpolluted area (one supposed sensitive, the other tolerant to metals) on burrow systems parameters. Macroinvertebrate porosity was studied using X-ray tomography. Three-dimensional reconstructions and characterisation of the burrow system were obtained using image analysis. Results showed that metal pollution principally affected the spatial distribution of macropores (more macropores were found near the soil surface) and the shape of the burrow system (branching rate was higher in the polluted soil), whereas soil macroinvertebrate composition principally affects burrow density parameters (the number of burrows was higher for the sensitive macroinvertebrate community).  相似文献

To test the assumption that changes to earthworm communities subsequently affect macroporosity and then soil water infiltration, we carried out a 3 year study of the earthworm communities in a experimental site having six experimental treatments: 2 tillage management systems and 3 cropping systems. The tillage management was either conventional (CT; annual mouldboard ploughing up to −30 cm depth) or reduced (RT; rotary harrow up to −7 cm depth). The 3 cropping systems were established to obtain a wide range of soil compaction intensities depending on the crop rotations and the rules of decision making. In the spring of 2005, the impact of these different treatments on earthworm induced macroporosity and water infiltration was studied. During the 3 years of observation, tillage management had a significant effect on bulk density (1.27 in CT and 1.49 mg m−3 in RT) whereas cropping system had a significant effect on bulk density in RT plots only. Tillage management did not significantly affect earthworm abundance but significantly influenced the ecological type of earthworms found in each plot (anecic were more abundant in RT). On the contrary cropping system did have a significant negative effect on earthworm abundance (104 and 129 ind. m−2 in the less and most compacted plots, respectively). Significantly higher numbers of Aporrectodea giardi and lower numbers of Aporrectodea caliginosa were found in the most compacted plots. CT affected all classes of porosity leading to a significant decrease in the number of pores and their continuity. Only larger pores, with a diameter superior to 6 mm, however, were adversely affected by soil compaction. Tillage management did not change water infiltration, probably because the increase in macroporosity in RT plots was offset by a significant increase in soil bulk density. However, cropping system had a significant effect on water infiltration (119 vs 79 mm h−1 in the less and most compacted plots, respectively). In RT plots, a significant correlation was observed between larger macropores (diameter > 6 mm) and water infiltration illustrating the potential positive effect of earthworms in these plots.  相似文献

In laboratory experiments, earthworms are often observed to burrow through compacted soil layers, leading to the general assumption that these animals play a significant role in regenerating compacted soils in agricultural plots. To demonstrate this role under field conditions, the abundance of earthworm macropores inside compacted zones was estimated on plots under reduced (RT) or conventional tillage (CT). Then, different types of compacted zones typically found in CT (plough pan and compacted clods) and RT plots (compacted volume under wheel tracks) were experimentally simulated in wooden boxes, buried in the field and inoculated with different earthworm species. After 6 weeks of incubation, the number of macropores inside the compacted zones was examined. Field observations showed that approximately 10% and 30% of the compacted zones were colonised by at least one macropore in CT and RT plots, respectively. A significantly greater number of anecics was found in RT plots, but we could not conclude that this ecological type of earthworm plays a more major role in the regeneration process in these plots since there were fewer compacted zones and these covered a smaller area in CT. The semi-field experiment provided evidence that earthworm-mediated regeneration of compacted zones is possible and its nature varies between ecological types of earthworm. Lumbricus terrestris, which makes individual burrows that are vertical and deep, was the main species to cross through the plough pan. The other three earthworm species (Aporrectodea giardi, A. caliginosa and A. rosea) did burrow inside the other types of compacted zones (“wheel tracks” and “compacted clods”). In every case, however, macropore density was far greater in non-compacted zones, illustrating that avoidance of compacted soil by earthworms is important and should be taken into account when extrapolating results from laboratory studies.  相似文献
