In ovo injection (IOI) of Naringin (N), flavanone was examined on post‐hatch blood biochemical parameters, antioxidant status and bone characteristics. Fertile eggs (n = 700) were distributed in seven groups with 100 eggs. On 14th and 17.5th days of incubation, four groups were injected using 15 or 30 mg active ingredient levels of naringin/0.5 ml saline/egg, low and high level, into amnion sac. Controls include sham (injected normal saline, 0.5 ml/egg on day 14 and 17.5th) and un‐injected group. IOI of high naringin and saline on 14th day of incubation resulted in lower hatchability and then higher mortality in last week of embryonic life. On day hatch, high levels of injected groups more body weight compared to the control. Chick length was increased at high levels of naringin on day 17.5th compared to control and saline injected. Quality traits of bones were improved in naringin‐injected groups compared to control. IOI of naringin influenced thyroid hormones on 14th day of incubation. Naringin groups influenced the Alkaline phosphatase (ALP), Calcium (Ca), superoxide dismutase (SOD), blood biochemical and lipids. Totally, amniotic IOI of naringin in last days of developing embryo may be useful for hatched chick, development of leg long bone or effect on biochemical metabolites by levels of flavanone that it needs more research. 相似文献
Ovulation rate and prolificacy are the most important reproductive traits that have major impact on the efficiency of lamb meat production. Here, we compared the whole genomes of the Romanov sheep, known as one of the high prolific breeds, and four other sheep breeds namely Assaf, Awassi, Cambridge and British du cher, to identify genetic mechanisms underlying prolificacy in sheep. Selection signature analysis revealed 637 and 477 protein‐coding genes under positive selection from FST and nucleotide diversity (Pi) statistics, respectively. Further analysis showed that several candidate genes including LEPR, PDGFRL and KLF5 genes are involved in sheep prolificacy. The identified candidate genes in the selected regions are novel and provide new insights into the genetic mechanisms underlying prolificacy in sheep and can be useful in sheep breeding programmes to develop improved breeds for high reproductive efficiency. 相似文献
Bluetongue virus (BTV), a member of Orbivirus genus (family Reoviridae), is a non-contagious infection of domestic and wild ruminants. The current study was designed to detect various serotypes of BTV in small ruminants of Khyber Pakhtunkhwa (KPK) province of Pakistan, along with their effects on hemato-biochemical parameters. A total of 408 serum samples in four districts (Mansehra, Abbottabad, Swabi, and Kohat) of KPK from small ruminants were screened based on competitive ELISA (cELISA). A total of 204 (50%) samples were found positive for BTV group–specific antibodies. The seropositive samples were processed for the detection of BTV serotypes through real-time polymerase chain reaction (qPCR). Out of 204 cELISA-positive samples, 60 (29.41%) were found positive through qPCR. Three serotypes [6, 8, 9] were detected from Mansehra District and two from Kohat [2, 8] and Abbottabad [6, 8], while only one from Swabi [8]. The serotype “8” was found consistently in all the four study districts. A significant (p?<?0.05) increase in the level of blood urea nitrogen (BUN) and alkaline phosphatase (ALP) was recorded in goats, whereas aspartate aminotransferase (AST) in sheep infected with BTV, compared to healthy animals. The hematological parameters showed significantly (p?<?0.05) raised total leucocyte count (TLC) in both sheep and goats, whereas only hematocrit (HCT) value was increased significantly (p?<?0.05) in infected sheep. This is the first report on serotyping of BTV among small ruminants in Pakistan.

Global warming, altering the physiology and irrigation demand of grapevines, has already been perceived in certain premium viticulture regions across the world. Selection of proper rootstocks for especially new cultivars has vital role for a sustainable viticulture under water-deficit conditions. The grapevine cultivar ‘Michele Palieri’ has been finding a good reception on the global markets. The objective of this study was to determine the response of ‘Michele Palieri’ cultivar to deficit irrigation using different rootstocks with distinct genetic origins. Two irrigation regimes (Full Irrigation [FI] and Deficit Irrigation [DI]) were applied to the vines of different grafting combinations of ‘Michele Palieri’ with Kober 5 BB, Richter 99, Richter 110, 140 Ruggeri, 44–53 Malégue or grown on own roots. Two years old vines were cultivated in 60?L pots containing sterile peat under controlled glasshouse conditions. Irrigations were regulated according to soil water matric potential (Ψm) levels using tensiometers. The volume of the irrigation water that has to be applied to attain 100% field capacity was performed as FI, while 50% of FI was considered as DI. The water was transported directly into the pots by micro-irrigation systems consisting of individual spaghetti tubes. The vines of DI treatment showed visible symptoms of mild water stress (e.?g., loss of turgor in shoot tips), but no defoliation or leaf necrosis occurred. DI treatment reduced the gs of ‘Michele Palieri’ scion cultivar in varying levels depending on the rootstock. DI treatment also affected vegetative growth of the scion cultivar in different levels. For example, the greatest decrease (26.7%) in shoot length resulting from DI was determined in ‘Michele Palieri’/5 BB grapevines, followed by own rooted vines (13.1%), while the lowest change (2.4%) was found in vines grafted on 110?R. Overall findings of this study imply that the rootstocks originating from V. berlandiery?×?V. rupestris hybrids (110?R, 99?R, 140?Ru and 44–53?M) better performed in a similar genetic aptitude under deficit irrigation regime while the rootstock 5 BB (V. berlandiery?×?V. rupestris) showed more susceptible responses. On the other hand, the general response of own rooted vines were better than those grafted on 5 BB. Background: Gonocytes give rise to spermatogonial stem cells, and thereby play an essential role in establishing spermatogenesis. Optimized culture conditions for gonocytes provide an opportunity for their study and in vitro manipulation for potential application in reproductive technologies. Using six experiments in a step-wise design, we examined the effects of several culture conditions on the maintenance, proliferation, and colony formation of porcine gonocytes. Testis cells from neonatal piglets were cultured for 7 d in DMEM supplemented with 10% fetal bovine serum. The examined culture conditions included using different cell seeding densities, gonocyte proportions, incubation temperatures, sampling strategies, and medium changing regimens.Results: Confluency of cel s was optimal(90% by ~6 d) when 3.0 × 104 testis cel s/cm2 containing ~40% gonocytes were used. Incubating the cel s at 35 °C or 37 °C resulted in similar cel number and viability at confluency, but incubation at 35 °C resulted in a delayed confluency. In the first 2 d of culture, gonocytes remained mostly floating in the medium and gradual y settled over the next 5 d. Consequently, not changing the medium for 7 d(as opposed to changing it every 2 d) led to a significant increase in the number of gonocyte colonies by reducing the loss of "floating gonocytes".Conclusion: We found that gonocytes require the presence of a critical minimum number of somatic cel s for settlement, and can proliferate and form growing colonies even in a basic medium. Large numbers of viable gonocytes remain floating in the medium for several days. The optimized culture conditions in the present study included seeding with 3.0 × 104 testis cel s/cm2 containing ~40% gonocytes, incubating at 37 °C, and without changing the medium in the first week, which can result in improved colony formation of porcine gonocytes. 相似文献
This study was carried out in Arsin (Trabzon/Turkey) in 2011. The effects of different harvest time and altitudes on the quality of the nuts have been investigated. The study was performed on Fo?a hazelnut and the harvest process has been conducted at three terms, which are on normal harvest time and ten days before and after harvest time. The harvested nuts were dried in the shade on the concrete floor until their moisture content decreased to 5?%. Some properties of nuts including yield, fruit weight, internal weight, shell thickness, and protein, oleic, and linoleic acid amounts have been investigated. As evaluated all of the fruit properties it can be concluded that 11 August is the most suitable harvest date for coast zone. On the other hand, no significant differences were obtained in the point of protein, oleic, and linoleic acid amounts for different harvest time and altitudes. 相似文献
In the present study, selected almond genotypes in Keciborlu/Isparta region by Y?ld?r?m (2007) based on late blooming and superior nut chacteristics were used to evaluate seedling growth performances and the reaction to root knot nematodes. Seeds were planted on perlite medium and stored 60 days at +4 °C in order to break seed dormancy. After germinating, seedlings were transferred to plastic pots to evaluate seedling growth characteristics and remaining seedlings were inoculated with Meloidogyne javanica and Meloidogyne incognita in order to asses resistance status of the selected seedlings. The highest germination percentage was obtained from Genotype 54 (96.1?%), the highest plant was observed from Genotype 9 (62.18 cm), the longest primary root was found from Genotype 54 (30.28 cm), the average secondary root lenght was highest in Genotype 9 (31.04 cm) and the widest root collar was observed in Genotype 33 (8.38 mm). Seedling stem diameter changed between 4.76 mm (Genoype 40) to 7.67 mm (Genotype 33). The lowest variation for stem widht was observed in Genotype 55 (0.58?%) while the lowest variation for seedling hight was found in Genotype 33 (0.50?%). Evaluated almond genotypes showed different reactions to studied nematode species, M. javanica and M. incognita. Resistant reactions of almond genotypes to nematode species were classified as susceptible, tolerant and resistant based on gal index values. Two experiments were designed to evaluate the effect of silymarin on stored spermatozoa using four rams. In experiment 1, silymarin was evaluated as a supplement for Tris–glucose extender. Semen samples (n = 20) were diluted with extender containing 0, 50, 100, 150 and 200 μg/ml silymarin and incubated at 5°C for 72 h. Membrane integrity, acrosome integrity, sperm viability and motility were evaluated at 72 h. Concentration of malondialdehyde (MDA) was determined after 48 h. Membrane integrity was higher in 100 μg/ml silymarin (65.2%) than control group (43.2%, p < 0.05). Acrosome integrity was highest in 100 μg/ml silymarin (71.3%, p < 0.05). Progressive motility was higher in 100 (58.5%), 150 (60.62%) and 200 μg/ml silymarin (54.7%) than control group (30.7%, p < 0.05). The highest MDA concentration was observed in control group (400 mm /10 × 106 sperm; p < 0.05). The goal of experiment 2 was to determine the interaction between silymarin and caproic acid on ram stored sperm. Ejaculates (n = 20) were diluted by Tris–glucose extender, added 0 (S?) or 100 μg/ml (S+) silymarin and 0 (C?) or 0.3125% (C+) caproic acid, and thereafter, aliquots were incubated at 5°C for 72 h. Membrane integrity was lower in C?S? (57.6%) than C?S+ (73.2%), C+S? (80.2%) and C+S+ (72.1%, p > 0.05). The highest sperm viability and acrosome integrity were observed in C+S‐ (82.4 and 80.1%, respectively; p < 0.05). There was no difference between C‐S+ and C+S+ on sperm viability and membrane integrity, progressive motility and MDA concentration (p > 0.05). The aim of this study was to examine the longest survival time of Echinococcus granulosus protoscolices stored at different humidities and constant temperatures from -10 to +40 degrees C. Sheep livers containing hydatid cysts obtained from slaughterhouses were taken to the laboratory within 3h and transferred into incubation cabinet previously set at -10, 0, +10, +20, +30 and +40 degrees C with different relative humidity (RH). Viability of protoscolices was assessed by 0.1% eosin staining. The longest survival times were 3 days at -10 degrees C (50% RH), 36 days at 0 degrees C (60% RH), 28 days at 10 degrees C (65% RH), 12 days at 20 degrees C (70% RH), 4 days at 30 degrees C (75% RH) and 3 days at 40 degrees C (80% RH). 相似文献
