Presented is an African giant rat (Cricetomys gambianus) following zinc ingestion. The sick rat was lethargic, withdrawn, had soft, mucus-impregnated faeces and diahorrea, foot twitching and icterus. Comparative age, sex and body weight (b.wt.)-matched analyses were made with a healthy giant rat. Twelve-hourly Urine volume (UV), Haematocrit (Hct), urinary glucose, plasma zinc and Alkaline Phosphatase (ALP) were performed over an 8-week period. Full blood counts were performed and differential WBC counts and microscopic observations were made on blood smears obtained from both healthy and sick rats. Consecutive blood samples were drawn at the end of each week (Weeks <2-6 treatment; Weeks 7-8 post-treatment). Treatment involved oral vitamin B12 supplement at 4 microg/day and 2 ml diethylenetriaminepentaacetic acid (DTPA) intramuscular injections at 1 ml/450g b.wt./5 wks (Week 2 - 6). Day 1 showed neutropaenia, Heinz bodies on RBCs (reticulocytes and immature forms). Zinc (Day 1 - end Week 7), glucose (Day 1 - end Week 4), ALP (Day 1 -Week 4) and UV were elevated (Day 1 - end Week 6). Indications of moderate zinc toxicosis following ingestion and stress-associated glucosuria were concluded.  相似文献

The modem crop scientist has a large amount of available nucleotide sequence information to identify genes of potential agronomic importance. Using reverse genetic approaches, specific genes can be disrupted, and hypotheses regarding gene function directly tested in vivo. Although a number of reverse genetic methods have been introduced, many are limited in application because they are organism-specific, expensive, transgenic, or only transiently disrupt gene function. However, traditional mutagenesis using chemical mutagens has been widely used as a forward genetics strategy to create many new crop plant varieties at relatively low cost. Mutagens such as ethyl methanesulphonate (EMS) cause stable point mutations and thus produce an allelic series of truncation and missense changes that can provide a range of phenotypes (Greene et al., 2003). TILLING (targeting induced local lesions IN genomes) is a high-throughput reverse genetic strategy that combines traditional mutagenesis and SNP discovery methods (Colbert et al., 2001; McCallum et al., 2000). To identify mutations, target regions of-l.5 kb are amplified with fluorescently labeled gene specific primers. Since its inception, in 1885 the Buckeye Irrigation Canal has delivered irrigation water in the Buckeye Valley, southwest of Phoenix, Arizona. Throughout its tenure the owners of the canal have consulted design professionals for its engineering needs. Early engineering in 1885 established canal alignments, delivery points and channel capacity. Until recently, the Buckeye Valley was a rural area relying on agricultural revenues to drive the local economy. Now, due to the recent housing market boom new homes started springing up like corn in September driving the existing open-channel laterals below grade into a patch work redesigned pipeline system. Because of these changes, professional construction managers are an enormous benefit to the Buckeye Water Conservation and Drainage District (BWCDD), the owner of the canal.As a result of this urbanization, contractors working for home developers are installing irrigation pipelines and systems with the focus of completing the project as quickly and economically as possible. Irrigation providers, on the other hand, focus on meeting the agricultural needs of their clients with consistent and uninterrupted water delivery. The services of a professional construction manager help both entities achieve their respective goals, shield BWCDD from construction of sub-par systems and risk, maintain uninterrupted service to growers and providing the technical expertise to communicate with both BWCDD and contractors.Construction managers provide a number of services to rural irrigation districts under-going urbanization. They team with an irrigation district by:
  • Understanding irrigation design concepts;
  • Understanding agricultural demands;
  • Providing a thorough knowledge of construction practices and standards;
  • Creating long-term design functionality;
  • Reducing stress on and increasing productivity of personnel;
  • Coordinating construction activities with water deliveries;
  • Verifying constructed facilities perform as designed, which reduces maintenance costs.
Construction managers represent an irrigation district in the field by:
  • Reviewing development plans for adherence to current construction standards and practices and coordinates construction schedules with water delivery needs;
  • Observing construction for proper installation;
  • Clarifying installers understanding of plans and specifications;
  • Coordinating interruptions in service;
  • Tracking multiple construction projects;
  • Explaining agricultural practices to contractor personnel;
  • Providing a single point of contact for construction related correspondence.
These “boots on the ground” create a local presence with a focus on the specific needs unique to an irrigation district. They provide a liaison while minimizing the impact to its growers.This paper will discuss the strain of urbanization on an irrigation district and the benefits of an engineering consulting firm to manage construction projects. The benefits include expertise, technical resources, design capability, coordination and inspection. Construction managers can help protect an irrigation district from being overwhelmed by rapid urbanization.  相似文献

Potato psyllid, Bactericera cockerelli (?ulc), causes economic damage to potato crops throughout the major potato growing regions of western North America. When cultivated crops are not available, potato psyllid often occurs on non-crop hosts. In the southern U.S. and northern Mexico, native species of Lycium (Solanaceae) are important non-crop hosts for the psyllid. We determined whether Old World species of Lycium now widespread in the Pacific Northwest are reservoirs of potato psyllid in this growing region. We examined Lycium spp. across a wide geographic region in Washington, Oregon, and Idaho at irregular intervals during three growing seasons. Potato psyllids were present at all locations. To determine whether Lycium is also a host during intervals of the year in which the potato crop is not available, we monitored a subset of these sites over the entire year. Six sites were monitored at 1- to 3-week intervals from June 2014 to June 2016. Psyllids were present on Lycium throughout the year at all sites, including during winter, indicating that Lycium is also a host when the potato crop is seasonally not available. Psyllid populations included a mixture of Northwestern and Western haplotypes. We observed well-defined spring and fall peaks in adult numbers, with peaks separated by long intervals in which psyllid numbers were very low. Seasonal patterns in psyllid numbers on these non-native Lycium hosts were very similar to what has been observed on native Lycium in the desert southwest region of the U.S. Our findings demonstrate that potato psyllid associates with Lycium across a broad geographic region within the Pacific Northwest. These results will assist in predicting sources of potato psyllid colonizing potatoes in this important growing region.  相似文献

Abstract. Experiments were set up at two sites to measure nitrogen (N) leaching loss from applications of separated pig/cattle slurry and cattle farmyard manure(FYM), during winters 1990/91–1993/94 (site A) and from broiler litter and FYM, during winters 1990/91–1992/93 (site B). The manures were applied at a target rate of 200 kg ha-1 total N during the autumn and winter to overwinter fallow or top dressed onto winter rye. The total N in leachate was calculated from leachate N concentrations, in samples collected using ceramic cups buried at 90 cm, and an estimate of drainage volume. Nitrogen losses were greatest following manure applications in September, October and November but losses following applications in December or January were not significantly elevated above those from untreated controls. Losses were consistently lower from FYM than from broiler litter or separated slurry. The presence of a cover crop (winter rye) significantly reduced overall N leaching compared with the fallow, but only reduced the manure N leaching losses at one site during one winter when a high proportion of drainage occurred late. The incorporation of a nitrification inhibitor (DCD) with manures applied in October did not significantly reduce the manure N leaching.  相似文献

Tumor induction in athymic nude mice can be used to detect dominant transforming genes in cellular DNA. Mouse NIH 3T3 cells freshly transfected with either cloned Moloney sarcoma proviral DNA or cellular DNA's derived from virally transformed cells induced tumors when injected into athymic nu/nu mice. Tumors were also induced by cells transfected with DNA from two tumor-derived and one chemically transformed human cell lines. The mouse tumors induced by human cell line DNA's contained human DNA sequences, and DNA derived from these tumors was capable of inducing both tumors and foci on subsequent transfection. Tumor induction in nude mice represents a useful new method for the detection and selection of cells transformed by cellular oncogenes.  相似文献

The jackass penguin Spheniscus demersus (L.) is confined to the coastal waters of southern Africa, and breeds on 18 islands predominantly within the region of the cold, nutrient rich Benguella current. Since the early 1900s the species' population has declined in numbers of unknown magnitude. The decline appears to be continuing, although the absence of reliable census data precludes an accurate measurement of the rate of change. The initial fall in numbers was undoubtedly due to severe disturbance of nesting birds and excessive commercial exploitation of eggs. The average annual egg crop for the period 1900-30 exceeded 450,000 eggs from Dassen Island alone. Egg collection has been suspended temporarily, but the species now appears to be threatened by oil pollution, and by competition with a pelagic shoal fish industry. Threats to breeding colonies are posed by harbour developments. These factors are discussed and evaluated. South African marine organisms are considered officially to be an economic resource and are heavily exploited. Legislation governing the control of this resource is the concern of the Ministry for Economic Affairs. There is no provision for any government body to investigate the status and biology of economically insignificant, but yet integral, components of the marine ecosystem. The major priority of conservations is to safeguard the jackass penguin population at its present level. This includes proclamation of several offshore islands and their surroundings as national nature reserves. We consider current efforts to rescue, clean and rehabilitate oiled seabirds as contributing little to conservation in real terms, even though the activity has an important humanitarian and educational function. Research priorities include a comprehensive census of the jackass penguin and other seabird populations in southern Africa; a study of mortality and recruitment, with particular attention to factors affecting the rates of these processes; a study of the dynamics of the penguin/prey interaction, with special reference to the possible effects of competition with the pelagic fishing industry; an evaluation of the degree of disturbance that nesting penguins can tolerate; an investigation into the temporal and spatial patterns of oil pollution incidents, oil slick occurrence and movement, and the development of methods of keeping birds away from oil slicks; and, lastly, an evaluation of the efficacy of rehabilitating oiled penguins as a contribution to maintaining or increasing the current jackass penguin population. Sorptive interactions between Escherichia coli (ATCC 11303B) and a purified allophane clay fraction were studied quantitatively using electronic particle counting and electrophoresis. Adsorption was influenced by pH and ions present at the allophane surface. These effects could be rationalized by considering changes in the net surface charge of the components. Electronic particle counting studies at pH 5.5 and above gave adsorption isotherms that obeyed single-term Langmuir kinetics. At pH 4.5, a more complex two-step isotherm was obtained, which was attributed to aggregation of unadsorbed cells by Al solubilized from the allophane surface.  相似文献

Drought is ubiquitous in rainfed cropping systems and often limits maize yields. The sensitivity of transpiration response early in progressive soil drying is a trait with potential to improve crop drought resistance. Simulation studies demonstrated that increased sensitivity to drying soil leading to restricted transpiration rates results in conservation of soil water during vegetative stages for possible use during grain filling. In contrast to other crops, there have been no studies characterizing genotypic variability for this trait in maize. Experiments in controlled environments were conducted to characterize the fraction of transpirable soil water (FTSW) threshold on drying soil for 36 hybrids selected for variation in the field for drought resistance, regions of adaptation and stay green. While FTSW thresholds varied among hybrids from 0.60 to 0.33, these thresholds were not uniformly associated with level of drought resistance in the field. Nevertheless, this study demonstrated a high FTSW threshold corresponded with drought resistance observed in some modern maize germplasm (hybrids #7, 17, 24, 27 and 32). This knowledge can enable breeding work seeking to exploit this adaptive trait to improved drought tolerance in low threshold FTSW germplasm.  相似文献
