Effects of water‐misting sprays with forced ventilation on post mortem glycolysis, adenosine monophosphate‐activated protein kinase (AMPK) and meat quality of broilers after transport during summer were investigated in the present paper. A total of 105 mixed‐sex Arbor Acres broilers were divided into three treatment groups: (i) 45 min transport without rest (T); (ii) 45 min transport with 1 h rest (TR); and (iii) 45 min transport with 15 min water‐misting sprays with forced ventilation and 45 min rest (TWFR). Each treatment consisted of five replicates with seven birds each. The results indicated that the water‐misting sprays with forced ventilation could mitigate the stress caused by transport under high temperature conditions during summer, which reduced the energy depletion in post mortem Pectoralis major (PM) muscle. This resulted in a higher energy status compared to the T group, which would decrease the expression of phosphorylation of AMPK (p‐AMPK). Furthermore, decreased the expression of p‐AMPK then slowed down the rate of glycolysis in post mortem PM muscle during the early post mortem period, which in turn lessened the negative effects caused by transport on meat quality. In conclusion, water‐misting sprays with forced ventilation may be a better method to control the incidence of the pale, soft and exudative meat in broilers.  相似文献

The epitopes of the capsid of foot-and-mouth disease virus (FMDV) play important roles in the construction of highly immunogenic subunit vaccines. However few epitopes have been found for FMDV serotype Asia1. In this study we screened for epitopes of the VP1 and VP2 proteins of FMDV serotype Asia1 isolate, YNBS/58. Fragments consisting of amino acids 133-163 of VP1 and amino acids 1-33 of VP2 contained epitopes, and both induced lymphoproliferation in guinea pigs. Only the VP1 fragment induced neutralizing antibodies but the VP2 peptide dramatically increased the neutralizing antibody response induced by the VP1 peptide.  相似文献

为解析二化螟Chilo suppressalis体内参与降解双链RNA(double-stranded RNA,dsRNA)的关键核酸酶的功能,克隆二化螟不同的非专一性核酸酶（non-specific nuclease,NUC）基因,并对这些基因进行生物信息学分析和组织定量表达分析,同时对dsRNA降解酶（dsRNA degrading nuclease,dsRNase）活力的组织分布进行研究。结果显示,共克隆获得5个NUC基因,其中有4个编码dsRNase亚家族基因（CsdsRNase1～CsdsRNase4）和1个编码Endonuclease G亚家族基因（CsEndoG）。5个NUC基因的开放阅读框核苷酸序列长度范围为828～1 338 bp,编码275～445个氨基酸残基,其分子量大小为31.68～49.57 kD,预测等电点为5.48～9.42。CsdsRNase1和CsdsRNase2含有信号肽序列,两者相似度极高,且与家蚕Bombyx mori和斜纹夜蛾Spodoptera litura中具有dsRNA降解酶活力的dsRNase同源聚类;CsdsRNase1和CsdsRN...  相似文献   

玉米黄花叶病毒运动蛋白的原核表达纯化与抗血清制备

为实现对玉米黄花叶病毒（maize yellow mosaic virus,MaYMV）的血清学检测,丰富该病毒的检测方法,将编码MaYMV运动蛋白（movement protein,MP）的基因连接到原核表达载体pDBHis-MBP上,将构建成功的原核表达质粒转化到大肠杆菌Escherichia coli中诱导表达融合蛋白,将纯化后的融合蛋白对新西兰大白兔Oryctolagus cuniculus进行免疫并制备MaYMV MP多克隆抗血清,并采用Western blot对抗血清的效价、灵敏度和特异性进行检测。结果显示,利用成功构建的原核表达载体经诱导表达获得分子量大小约为62 kD的融合蛋白,纯化后对新西兰大白兔进行免疫获得MaYMV MP抗血清,该抗血清的效价为1∶128 000,灵敏度为1∶32,且该抗血清能够特异性地检测到本氏烟Nicotiana benthamiana中瞬时表达的MaYMV MP,而不与马铃薯卷叶病毒属Polerovirus及黄症病毒属Luteovirus的其他病毒发生血清学交叉反应,证明该抗血清具有良好的特异性。表明本研究制备的MaYMV MP抗血清能特异性...  相似文献

本文研究了外源多胺对在低温胁迫下甜瓜幼苗体内DNA、RNA、可溶性蛋白质、超氧物歧化酶(SOD)及细胞质膜透性的影响.结果表明,多胺处理的甜瓜幼苗在低温(2±1℃)下处理10h后,RNA和DNA含量明显增多,蛋白质的图谱发生显著变化,SOD活性增强,细胞膜的破损渗漏较少,尤以0.4mmol的腐胺处理最为显著.多胺与核酸的相互作用调节了植物体内的代谢过程,与膜的相互作用增强了机体的抗冷性.  相似文献

The vendor selection directly relatives to the procurement quality and influences the competition of the corporation in the supply chains. It is important and practical to study the vendor selection method. The measuring criteria and the method for vendor selection are analyzed. The authors put forward an new method which is combined with both the advantage of analytic hierarchy process (AHP ) and random disposal of data envelopment analysis ( DEA) for vendor selection. It introduces a random variable which solves the disadvantage of the weigh value in DEA which convert subjective estimation to objective opinion in the course of the vendor selecting.
