Prevention of isinglass on the chronic atrophic gastritis in rats and its mechanism

AIM: To evaluate the effect of isinglass on chronic atrophic gastritis(CAG) in rats and its mechanism. METHODS: An animal model of CAG in accordance with the previous experience of combined administration of 60% ethanol, 20 mmol/L sodium deoxycholate and 0.1% ammonia water was established in SD rats. Isinglass was used as preventive therapy while we were establishing CAG rat model. Finally all the rats were executed and pathologic changes of the gastric mucosa were studied by gross appearance and microscopy and serum epidermal growth factor (EFG) and growth hormone(GH) contents were tested. RESULTS: In each isinglass prevention group, inflammation grade of gastric antrum was less than that in model group (P<0.01) while the mean ratio of the thickness of gastric mucosal gland and muscularis mucosa (L1/L2), the number of gastric glands in 1 mm lengths of mucosal layer in longitudinal sections were much better than those in model group (P<0.01).They were very close to normal control group (P>0.05). The expression of proliferating cell nuclear antigen (PCNA) in gastric mucosa and serum EFG level were higher than those in model group (P<0.01, P<0.05), but serum GH content showed no different between isinglass prevention group and model group. CONCLUSION: Isinglass preventes the gastric mucosal atrophy in the CAG model. Its mechanism may be related to the effects of decreasing the gastric mucosal damage， promoting the cell proliferation and increasing of internal EFG secretion.  相似文献   

Effect of rhEGF activating extracellular signal-regulated kinase on the expression of Bcl-2 and p53 protein in human amniotic cells

AIM: To explore the effect of recombinant human epidermal growth factor (rhEGF) on the ERK1/2 pathway and its safety in use when the proliferation of cultured FL cells (human amnion epithelial cells) were promoted by rhEGF.METHODS: FL cells were stimulated by rhEGF at various concentrations.The proliferation rate of FL cells was evaluated by MTT assay.Western blotting was used to detect phosphorylation of ERK1/2 (p-ERK1/2) and the change of Bcl-2 and P53 protein.RESULTS: The proliferation rate FL cells reached the maximum when the concentration of rhEGF was 10 μg/L (42.4％，P<0.01).Western blotting showed that the activity of p-ERK1/2 increased significantly when rhEGF ranged in 10 μg/L to 60 μg/L.Level of Bcl-2 was unchanged in each groups.Level of P53 decreased significantly when the concentration of rhEGF was 60 μg/L.CONCLUSION: It is safe when rhEGF presents in optimization concentration of 10 μg/L.The capability of apoptosis may be depressed when rhEGF is in larger dose.  相似文献

HPLC法测定蒺参胶囊中人参皂苷Re的含量

目的建立蒺参胶囊中人参皂苷Re的含量测定方法。方法采用高效液相色谱法测定。结果人参皂苷Re在1.65～6.05μg之间与峰面积呈良好的线性关系(r=0.999 7),平均回收率为95.98%,RSD为1.35%(n=5)。结论方法简便、可靠、准确,可用于蒺参胶囊的质量控制。  相似文献

本试验对新生仔猪哺乳前初次进行猪瘟疫苗免疫，至７０日龄时进行第二次免疫，免疫持续至７０日龄和１５０日龄时两次应用Ｄｏｔ－ＥＬＩＳＡ方法进行检测，结果表明，阳性率分别为９２．３％和９１．２５％，说明此免疫方法是一较好的预防猪瘟的免疫程序。  相似文献

为验证牛分枝杆菌外膜蛋白A是否具有免疫活性,以牛分枝杆菌Vallee Ⅲ株基因组DNA为模板扩增牛分枝杆菌外膜蛋白基因ompA,采用DNA重组技术将此基因片段连于表达载体pGEX6p-1,构建重组质粒pGEX6p-1-ompA,并将其转化到大肠杆菌中BL21(DE3)中,IPTG诱导(终浓度为0.1mmol/L),表达产物进行SDS-PAGE分析.用glutathione sepharose 4B纯化蛋白和Western blot分析该蛋白的免疫学活性.结果表明:pGEX6p-1-ompA以可溶形式表达,蛋白分子量大小为60 kDa,与预计大小相符,纯化的蛋白具有良好的免疫学活性.  相似文献   

GPX4失活通过JNK通路缓解LPS诱导巨噬细胞炎症反应

【目的】 研究硒蛋白谷胱甘肽过氧化物酶4（glutathione peroxidases 4,GPX4）失活如何参与调控脂多糖（lipopolysaccharide,LPS）诱导的RAW264.7巨噬细胞炎症反应及其潜在的分子机制。【方法】 体外培养RAW264.7巨噬细胞,以DMSO为对照,使用0.1~5.0 μmol/L GPX4抑制剂FIN56处理,通过CCK-8法检测细胞活力和Western blotting检测GPX4蛋白表达水平,确定抑制剂最适浓度。将RAW264.7巨噬细胞分为4组：对照组,添加DMSO培养24 h;FIN56（GPX4抑制剂）组,添加0.5 μmol/L FIN56培养24 h;DMSO-LPS组,DMSO培养24 h后使用LPS （100 ng/mL）刺激3 h;FIN56-LPS组,FIN56培养24 h后使用LPS刺激3 h。各组细胞经培养后,利用荧光探针2',7'-二氯二氢荧光素二乙酸酯（2',7'-dichlorodi-hydrofluorescein diacetate,H₂DCFDA）检测细胞内活性氧（reactive oxygen species,ROS）水平,使用试剂盒法检测细胞内丙二醛（malondialdehyde,MDA）水平,分别使用ELISA和荧光定量PCR检测促炎细胞因子白细胞介素1β（interleukin-1β,IL-1β）、白细胞介素6（interleukin-6,IL-6）、肿瘤坏死因子α（tumor necrosis factor-α,TNF-α）的分泌水平和基因表达量,使用Western blotting法检测Toll样受体4（toll like receptor 4,TLR4）信号通路相关蛋白表达水平。【结果】 与DMSO处理相比,0.5 μmol/L FIN56处理巨噬细胞24 h对细胞活性无显著影响（P>0.05）,且显著降低GPX4蛋白表达水平（P<0.05）,故选用0.5 μmol/L FIN56用于后续实验。与对照组相比,FIN56和LPS均显著增加了ROS积累（P<0.05）,而与DMSO-LPS组相比,FIN56-LPS组ROS水平显著升高（P<0.05）。与对照组相比,LPS可显著增加小鼠巨噬细胞IL-1β、IL-6和TNF-α的mRNA表达量和IL-6含量,以及c-Jun氨基末端蛋白激酶（c-Jun N-terminal protein kinase,JNK）和c-Jun磷酸化水平（P<0.05）,而FIN56可显著下调LPS诱导的IL-6的mRNA表达量和含量及JNK和c-Jun磷酸化水平（P<0.05）,但对p38蛋白（p38 MAPK,p38）、细胞外调节蛋白激酶（phosphorylate extracellular regulated protein kinases1/2,Erk1/2）蛋白表达水平无显著影响（P>0.05）。【结论】 GPX4失活可有效阻断JNK和c-Jun磷酸化,从而特异性抑制LPS诱导的巨噬细胞的炎症反应。该结果可为以GPX4为靶点研发抗炎治疗策略提供理论依据。  相似文献

研究了核桃楸树皮丙酮-水提取物的水溶性萃取部位的化学成分.采用Sephadex LH-20柱色谱及薄层色谱等方法进行分离,经波谱分析及理化性质测定,鉴定了6个化合物:没食子酸(1)、鞣花酸(2)、1,2,6-三没食子酰葡萄糖(3)、1,3,6-三没食子酰葡萄糖(4)、1,2,4,6-四没食子酰葡萄糖(5)及1,2,3,4,6-五没食子酰葡萄糖(6).化合物2、4、6为首次从该植物中分得.  相似文献
