The poor reputation of birch in Ireland is gradually changing, and the interest shown in it by foresters is growing, as is the recognition of the many advantages that this genus offers, especially from an afforestation and ecosystem development point of view. The potential of native birch species was investigated on industrial cutaway peatlands in the Irish midlands. Field experiments were established to evaluate differences (in terms of survival, growth attributes and form) between B. pendula and B. pubescens, between bare-root and container planting, and between small and medium size seedlings. After five growing seasons, B. pendula was found to be the superior species especially on well drained and shallow peat sites. Bare-root B. pendula will grow faster with reasonable form but containerised birch may be favoured on more difficult sites (with deeper and wetter peat). Larger seedlings performed better, regardless of species. This study demonstrated good growth potential for planted birch on cutaway peatlands and that this species should be integrated in further planting programmes.  相似文献

Sphaeropsis sapinea is a pathogen of many coniferous species and causes significant losses to the plantation forestry industries of many countries. New Zealand isolates of S. sapinea were examined for colony and morphological characteristics, and pathogenicity to Pinus radiata seedlings. Considerable variation was observed in colony growth form and colour, and in the growth rates on four agar media. The conidial dimensions, carbon and nitrogen utilization, and pathogenicity also varied among the isolates. The variation observed was such that the isolates could not be placed within previously described distinct morphotypes and the present results concur with previous studies cautioning the use of morphological criteria alone to partition the species. These results contrast markedly with previous reports and seemingly indicate a change in the New Zealand S. sapinea population. This difference may reflect the re‐introduction or spread of additional isolates or a shift in the structure of the resident population.  相似文献

Infection and sporulation of races of Phytophthora clandestina in susceptible and resistant cultivars of subterranean clover and the effect of the races on growth of the cultivars in pots were studied using a total of five races of the pathogen and eight host cultivars. The pathogen caused severe disease where races and cultivars were compatible, although a significant reduction in root and shoot dry weight occurred only on Woogenellup inoculated with race 2. There was little increase in disease severity after 1 wk from the time of inoculation. Sporangia and oospores were formed at high frequency in tap and lateral roots in all compatible combinations, except in Woogenellup in which sporangia were produced inconsistently. The pathogen caused negligible disease and produced very few sporangia in incompatible combinations. Plant age had a strong effect on disease development. Disease severity caused by virulent races on cultivars Gosse, Trikkala, Meteora and Woogenellup was less on older than on younger plants. A further experiment using aggressive isolates of races 0, 1, 2, 3 and 4 revealed that cultivar Gosse showed no reduction in root and shoot weight, despite it being susceptible to races 1, 3 and 4. This provided evidence of the existence of race-non-specific or horizontal resistance to P. clandestina in cultivar Gosse. Objective A real-time polymerase chain reaction (PCR)/high-resolution melt (HRM) curve analysis protocol was developed in our laboratory to differentiate infectious bronchitis (IB) virus reference strains. In the current study, this method was used to detect and classify IB viruses in field submissions. Procedure Over an 11-month period samples from 40 cases of suspected IB virus were received and 17 submissions were positive for IB virus by polymerase chain reaction. HRM curve analysis classified each strain as subgroup 1, 2 or 3 strain (12 submissions) or a strain that was unable to be classified (5 submissions). The 3′ untranslated region (UTR) and partial S1 gene nucleotide sequences for the 17 IB virus strains were determined and their identity with those of the relative reference strains compared to confirm the classifications generated using the HRM curve analysis. Results Of the 12 IB field viruses classified as subgroup 1, 2, or 3 using HRM curve analysis, the 3′UTR and S1 gene nucleotide sequences had identities ≥99% with the respective subgroup reference strain. Analysis of the 3′ UTR and S1 gene nucleotide sequences for the five IB virus strains that could not be classified indicated that four belonged to one of the subgroups, and one was a potential recombinant strain (between strains from subgroups 2 and 3). A novel recombinant strain was also detected. Conclusion HRM curve analysis can rapidly assign the majority of IB viruses present in field submissions to known subgroups. Importantly, HRM curve analysis also identified variant genotypes that require further investigation.  相似文献

Pure bred Holstein–Friesian (FR) and beef breed × FR male calves are used for beef production in Ireland. Beef breeds used for crossbreeding on FR cows include Aberdeen Angus (AA) and Belgian Blue (BB) which represent extremes in terms of maturity. The objective of this study was to compare spring-born steers of FR, AA and BB breed types, offered two feeding levels in their second winter, and subsequently finished on pasture or on a high concentrate diet indoors. Seventy-two steers (24 per breed type) were managed together to the end of their second grazing season. They were then blocked on weight within breed type and assigned to a 3 (FR, AA and BB breed types) × 2 (winter feeding levels) × 2 (finishing systems) factorial experiment. The two winter feeding levels were grass silage ad libitum plus mean daily concentrate levels of 0.91 (L) or 4.0 (H) kg dry matter for 113 days. The two finishing systems were pasture or concentrates ad libitum for a mean period of 94 days. Mean slaughter and carcass weights for FR, AA and BB were 634, 644 and 642 (s.e. 8.1), and 313, 326 and 340 (s.e. 4.7) kg, respectively. Other than bone proportion which was lower for AA, there were few differences in ribs joint composition or in m. longissimus chemical composition between FR and AA. BB had less fat and more muscle in the ribs joint, and more moisture and protein, and less lipid in m. longissimus than both FR and AA. Compared with L, the H winter feeding level increased slaughter weight and carcass weight by 24 and 15 kg, respectively. Indoor finished animals were 63 kg live weight and 39 kg carcass weight heavier than those finished at pasture. They also had more fat and less muscle and bone in the ribs joint and more lipid and less moisture in m. longissimus. It is concluded that except for BB finished at pasture all carcasses were commercially acceptable. Despite the excellent finishing performance on concentrates, this system is not profitable at current concentrate and beef prices.  相似文献

Genetic merit for growth rate, expressed as expected progeny difference for carcass weight (EPDCWT), is available for dairy and beef sires used in Ireland. The once predominantly Friesian (FR) dairy herd has experienced significant introgression of Holstein (HO) genes over the past two decades, and cross-breeding of dairy cows, not required to produce herd replacements, with beef bulls is common. The objective of this study was to compare growth rate, feed intake, live animal measurements and slaughter traits of progeny of Holstein–Friesian dairy cows and bulls of two contrasting maturity beef breeds namely Aberdeen Angus (AA) and Belgian Blue (BB), each selected for either high (H) or low (L) estimated genetic merit for carcass weight. Two dairy strains (FR and HO) were also included giving six genetic groups in total. A total of 170 male progeny from spring-calving cows were artificially reared indoors and subsequently managed together at pasture until the end of their second grazing season when they were assigned to one of two mean slaughter weights (i) 560 kg (Light) or (ii) 620 kg (Heavy). Daily feed intake was recorded during the first winter and during finishing. Body measurements were recorded four times during the animals' life, and linear scoring was carried out at 9 months of age and again at slaughter. Carcasses were graded for conformation and fatness (15 point scale). Slaughter and carcass weights per day of age for AAH, AAL, BBH, BBL, FR and HO were 782, 719, 795, 793, 804 and 783 (SE 12.9) g, and 415, 372, 438, 436, 413 and 401 (SE 5.8) g, respectively. Corresponding values for carcass weight, kill-out proportion, carcass conformation class (15 point scale) and carcass fat class (15 point scale) were 314, 283, 334, 333, 317 and 305 (SE 4.7) kg, 526, 518, 553, 550, 519 and 511 (SE 2.9) g/kg, 6.2, 5.4, 8.0, 7.9, 5.3 and 3.7 (SE 0.26), and 9.8, 9.3, 7.4, 7.2, 9.3 and 8.2 (SE 0.26). There were significant interactions between estimated genetic merit for carcass weight and beef breed with the differences between H and L mainly expressed for AA only. Feed intake differences between H and L animals were negligible and largely attributable to the differences in live weight. Following scaling for live weight, beef breeds of high estimated genetic merit for carcass weight had lower skeletal measurements, indicating greater compactness, with the effect more pronounced in AA. It is concluded that using beef sires of estimated high genetic merit for carcass weight on dairy cows increases growth rate and carcass weight of the progeny but the effect may not be similar for all breeds.  相似文献

This article describes the occurrence of bilateral dentigerous cysts in a 1‐year‐old entire male Standardbred horse. Computer tomography (CT) with a positive contrast CT fistulogram was used to accurately define the anatomy of the cysts preoperatively. Conventional surgical techniques were used to remove the cysts under general anaesthesia. The CT studies were able to precisely define the margins of the cysts and facilitated uncomplicated surgical excision. Follow‐up examination of the animal revealed excellent cosmesis at the surgical site. Histology was used to confirm the diagnosis of bilateral dentigerous cysts. Preoperative CT studies are invaluable to precisely define the anatomical locations of the dental anomalies associated with dentigerous cysts and thereby allow confident and uncomplicated excision. Dentigerous cysts may occur bilaterally in horses.  相似文献

The virulence spectrum of 112 isolates of Phytophthora clandestina collected from 56 sites in four subterranean clover-growing states in southern Australia was determined using differential cultivars of subterranean clover. Five races were detected, with race 0 in all states except New South Wales, race 1 in all states, race 2 only in Victoria, race 3 only in New South Wales, and race 4 in Victoria and Western Australia. The level of genotypic diversity among the different P. clandestina populations was investigated using five RAPD primers. Among 30 bands amplified, only two were polymorphic. This enabled identification of four multilocus RAPD genotypes. Three of the four genotypes occurred in all four states. Races 2 and 3 occurred with RAPD genotypes 1 and 2 only whereas races 0 and 1 occurred in all four multilocus RAPD genotypes. These results indicate that the pathogenicity spectrum of P. clandestina can change rapidly.  相似文献

Spermatogonial stem cells (SSC) have important applications in domestic animal reproduction and advanced biotechnologies. Because differential plating is one of the most common methods used for SSC enrichment, the goal of this study was to compare three differential plating methods for the enrichment of bovine SSC. To achieve this goal, testicular parenchyma from pre‐pubertal calves was minced and single cells were obtained after two enzymatic digestions. We compared three coating methods for differential plating: laminin (20 ng/ml), BSA (0.05 mg/ml) and PBS. Cells were incubated at 37°C, 5% CO2 in air for 15 min onto laminin‐coated dishes or 2 h onto BSA‐ or PBS‐coated dishes. Cell viability was assessed by trypan blue exclusion method. Recovered cells were analysed for the expression of SSC molecular markers by quantitative RT‐PCR (GFRA1, CXCR4, ITGA6, THY1) and flow cytometry (GFRA1, CXCR4 and ITGA6). Cells at time 0, adherent cells on laminin and non‐adherent cells from BSA and PBS groups had the same cell viability (p = 0.0655). GFRA1, CXCR4 and THY1 relative gene expression was higher (p = 0.0402, p = 0.0007, p = 0.0117, respectively) for non‐adherent cells selected in PBS group. Flow cytometry analysis revealed that the presence of GFRA‐positive (GFRA+) cells was higher in non‐adherent cells from BSA and PBS groups (p < 0.001). However, laminin‐adherent cells had higher number of ITGA6+ cells (p < 0.001) and lower presence of CXCR4+ cells (p = 0.0012). In conclusion, differential plating is an effective method for the enrichment of bovine undifferentiated spermatogonia and higher expression of SSC markers is obtained without laminin or BSA coating.  相似文献

Accuracy and bias of EBV are important measures of the quality of genetic evaluations. A sampling method that accounts for the uncertainty in the estimation of genetic group effects was used to calculate accuracy and bias of estimated effects. The method works by repeatedly simulating phenotypes for multiple traits for a defined data and pedigree structure. These simulated values are analyzed using BLUP with genetic groups in the relationship matrix. Accuracies and biases are then calculated as correlations among and differences between true and estimated values across all replicates, respectively. The method was applied to the Irish beef production data set for 15 traits and with 15 genetic groups to account for differences in breed means. Accuracy and bias of estimated genetic group effects, estimated comparisons between genetic group effects, EBV within genetic group, and EBV across genetic group were calculated. Small biases were detected for most estimated genetic group effects and most estimated comparisons between genetic group effects. Most of these were not important relative to the phenotypic SD of the traits involved. For example, a bias of 0.78% of the phenotypic SD was detected for carcass conformation in Aberdeen Angus. However, one trait, calf quality, which had few performance records in the data set, displayed larger bias, ranging from -10.31 to 5.85% of the phenotypic SD across the different estimated genetic group effects. Large differences were observed in the accuracies of genetic group effects, ranging from 0.02 for feed intake in Holstein, which had no data recorded, to >0.97 for carcass conformation, a trait with large amounts of data recorded in the different genetic groups. Large differences were also observed in the accuracies of the comparisons among genetic group effects. The accuracies of the EBV within genetic group and EBV across genetic group were sometimes different; for example, carcass conformation in Belgian Blue had an average accuracy within genetic group of 0.69 compared with an average accuracy across genetic group of 0.89. This suggests that the accuracy of genetic groups should be taken into account when publishing EBV across genetic groups.  相似文献
