DNA ploidy analysis and the expression of TIMP-2 and E-cadherin in gastric carcinoma

AIM:To investigate DNA ploidy and the expression of TIMP-2 and E-cadherin in gastric carcinoma in order to understand its molecular basis and probable mechanism of invasion and metastasis. METHODS:Immunohistochemical methods were used to detect the expression for TIMP-2 and E-cadherin in 99 cases of gastric carcinoma, 16 cases of adjacent noncancerous mucosa, 16 cases of distant metastases and 25 cases of metastatic lymph nodes. Flow cytometry DNA ploidy and S-phase fraction (SPF) analysis was performed on 47 cases of gastric cancer, 6 cases of adjacent noncancerous mucosa and 4 cases of distant metastasis cancer with the use of formalin-fixed paraffin embedded specimens. RESULTS:The expression of TIMP-2 was significantly correlated with Borrmann’s classification, LN metastasis and the depth of invasion. The expression of E-cadherin was significantly correlated with tumor cell differentiation, Lauren’s classification, Borrmann’s classification, LN metastasis and the depth of invasion. There was a positive relationship between DNA aneuploid rate and differentiation and LN metastasis. There was a positive relationship between SPF that is higher than 15% and tumor size, differentiation and LN metastasis. And there was a significantly difference between carcinoma and noncarcinoma when the expression of E-cadherin, DNA aneuploid rate and SPF were analyzed. There was no correlation between TIMP-2 and E-cadherin. There was a positive relationship between DNA ploidy or SPF and the expression of E-cadherin. CONCLUSION:As the development of tumor progression and heterogeneity, the abnormal expression of TIMP-2 or E-cadherin or the rate of DNA aneupoid or higher SPF gradually correspondingly increases, suggesting that they play a crucial role in gastric carcinoma progression. Furthermore, each factor influences one another and further accelerates the process of tumor progression.  相似文献   

Significance of CD117 and CD34 expression in gastrointestinals tromal tumors

AIM:In this article,gastrointestinal tumo r-related historical ar chives of pathology in our hospital were reviewed in order to acknowledge gastro intestinal stromal tumors (GIST).METHODS:These cases were rediagnosed correctly ,these tissues were labeled with some antibodies such as CD117,CD34,α-SMA,S-100,and the ir clinical characteristics were explored.RESULTS:CD117,CD34 were expressed widely in GISTs ,the percen tage of positive expression was CD117 （50/56,89.3%）,CD34 （37/56,66.1% ）,respectively,α-SMA,S-100 were （17/56,30.4%）,（4/56,7.1%）,re spe ctively,desmin was negative in these cases.Among them,29 cases were benign a nd borderline,27 cases were malignant.The percentage of occurring in stomach a nd intestine was 91.1%.CONCLUSION:CD117,CD34 are expressed significantly in GIST,t hese might be assistant markers for GIST diagnosis.GISTs mostly occurred in sto mach and intestine.  相似文献

利用高分辨快原子轰击质谱(HR-FABMS),以(甘油 氯化钠)为基质测定了9个新的β-二氢沉香呋喃倍半萜多酯和生物碱的HR-FABMS。结果表明,所有该类化合物均可以精确地获得[M Na] ,[M H] ,[M H-H2O] 等特征离子的质量和元素组成,从而方便地获得原有该类化合物的分子量和分子式。  相似文献

哺乳动物体细胞核移植 (克隆 )是近年来迅速发展起来的一项新技术 ,对濒危动物保护、优良种畜快速扩群以及核质关系的研究、药物试验等诸多领域都具有重大意义。但体细胞核移植是一项程序繁杂的工作 ,每一步操作所采用的试剂、材料和方案等都会影响到核移植效率 ,因而其影响因素十分众多。文章根据目前已有资料 ,论述了卵母细胞来源、卵母细胞与供体细胞的胞质容量、卵母细胞的去核方案、体细胞供体动物的年龄、供体细胞的组织来源与传代次数及供体细胞的冷藏、冷冻和所处的细胞周期阶段对核移植效率的影响。另外 ,对重组胚的融合 -激活方案、核移植方案及重构胚的培养方案对核移植效率的影响也作了简要论述。  相似文献   

多层螺旋CT与核磁共振血管成像、数字减影对主动脉夹层诊断的比较研究

目的：比较多层螺旋CT(multi-slice spiral CT，MSCT)与核磁共振血管成像(MRA)、数字减影(DSA)对主动脉夹层(aortic dissection，AD)的诊断价值。方法：对17例AD患者行MSCT检查，并与MRA、DSA的检查结果进行比较。结果：对17例AD患者用MSCT、MRA、DSA都能作出诊断(按DeBakey分型，Ⅰ型有4例，Ⅱ型有1例，Ⅲ型有12例)，但只有MSCT能明确显示夹层的范围和分支的受累情况，以及剥离内膜瓣、裂口及主动脉的周围情况。而MRA对裂口的检出率只有21．74％，DSA可达87．0％。结论：MSCT对AD的分型以及破裂口的定位诊断更具准确性，对动脉分支显示更清晰。  相似文献

本文在卡普法的基础上，提出了木材干燥应力连续监测方法，并设计了所需的配套装置。该方法采用涡流传感器对干燥过程中卡普片的矢高进行连续测试，同时通过计算机自动采集、存储数据。结果表明：矢高与干燥时间存在精密的相关性，它们之间的相关系数大部分达到0．99以上，从而实现了木材干燥应力的自动与连续监测。  相似文献
