Conjugation allows bacteria to acquire genes for antibiotic resistance, novel virulence attributes, and alternative metabolic pathways. Using a fluorescent protein fusion, SeqA-YFP, we have visualized this process in real time and in single cells of Escherichia coli. We found that the F pilus mediates DNA transfer at considerable cell-to-cell distances. Integration of transferred DNA by recombination occurred in up to 96% of recipients; in the remaining cells, the transferred DNA was fully degraded by the RecBCD helicase/nuclease. The acquired integrated DNA was tracked through successive replication rounds and was found to occasionally split and segregate with different chromosomes, leading to the inheritance of different gene clusters within the cell lineage. The incidence of DNA splitting corresponds to about one crossover per cell generation. 相似文献

A 7-year 6-month-old, castrated male Shiba dog presented with a 1-month history of lethargy, anorexia, vomiting, and frequent watery diarrhea. Weight loss, hypoalbuminemia, anemia, and leukocytosis were detected at the first visit. The dog was diagnosed with non-responsive enteropathy (NRE) based on clinical and histopathological examinations. Since the dog did not respond to the immunosuppressive drugs, fecal microbiota transplantation (FMT) was performed during the treatment with chlorambucil. A single endoscopic FMT into the cecum and colon drastically recovered clinical signs and clinicopathological abnormalities and corrected dysbiosis in the dog. No recurrence or adverse events were observed. The present case report suggests that FMT, possibly together with chlorambucil, might be a treatment option for NRE in Shiba dogs that have poorer prognosis compared with other dog breeds. 相似文献

On the basis of a mechanistic model, the overall and liquid mass transfer coefficients of aroma compounds were estimated during aroma release when an inert gas diluted the static headspace over simple ethanol/water solutions (ethanol concentration = 120 mL x L(-1)). Studied for a range of 17 compounds, they were both increased in the ethanol/water solution compared to the water solution, showing a better mass transfer due to the presence of ethanol, additively to partition coefficient variation. Thermal imaging results showed differences in convection of the two systems (water and ethanol/water) arguing for ethanol convection enhancement inside the liquid. The effect of ethanol in the solution on mass transfer coefficients at different temperatures was minor. On the contrary, at different headspace dilution rates, the effect of ethanol in the solution helped to maintain the volatile headspace concentration close to equilibrium concentration, when the headspace was replenished 1-3 times per minute. 相似文献

The aim of this study was to evaluate the diagnostic imaging approach to identify the deficiency of boron in leaves of maize. The experiment was conducted in a greenhouse under a hydroponic system. The treatments were four levels of boron (B) in solution nutrition (zero, 0.12, 0.24 and 0.60 mg L?1), combined at V4, V7, and R1 growing stages. Plant parts sampled included index leaf and new leaf to chemical analysis and texture image analysis. Our proposal was to apply these texture analysis and pattern classification schemes to identify the levels of B. Texture methods achieved 98% of accuracy in differentiating between leaves properly fertilized with B, from leaves with deficiency, in V4. In all tests, with index leaf success rate was higher than 80%, and around 90% for the new leaf. The image analysis by texture techniques applied on maize leaves are able to identify boron deficiencies in younger plants. 相似文献
