Response to thyrotropin (TSH) was evaluated in 2 groups of mixed-breed dogs. Thyrotropin (5 IU) was administered IV to dogs in group 1 (n = 15) and IM to dogs in group 2 (n = 15). Venous blood samples were collected immediately before administration of TSH and at 2-hour intervals for 12 hours thereafter. In group 1, the maximum mean concentration (+/- SD) of thyroxine (T4; 7.76 +/- 2.60 micrograms/dl) and 3,5,3'-triiodothyroxine (T3; 1.56 +/- 0.51 ng/ml) was attained at postinjection hours (PIH) 8 and 6, respectively. However, the mean concentration of T4 at PIH 6 (7.21 +/- 2.39 micrograms/dl) was not different (P greater than 0.05) from the mean concentration at PIH 8. The maximum mean concentration of T4 (10.10 +/- 3.50 micrograms/dl) and T3 (2.22 +/- 1.24 ng/ml) in group 2 was attained at PIH 12 and 10, respectively. Because dogs given TSH by the IM route manifested pain during injection, had variable serum concentrations of T3 after TSH administration, and may require 5 IU to achieve maximal increases in serum T4 concentrations, IV administration of TSH is recommended. The optimal sampling time to observe maximal increases in T3 and T4 after IV administration of TSH was 6 hours. Repeat IV administration of TSH may cause anaphylaxis and, therefore, is not recommended.  相似文献

The DNA fingerprint profiles of 126 isolants of Pasteurella multocida from 41 turkey farms in Missouri were analyzed after digestion with the restriction endonuclease HhaI and compared with their somatic antigenic type. The goal was to determine if the same isolant of P. multocida was reisolated from the the same farm during the same and consecutive years and after an interval of one or more years. Of the 37 pairs of P. multocida collected during the same year from the same turkey farms, the DNA fingerprint profiles were the same with 26 pairs (70.3%) and different with 11 pairs (29.7%). Of the 33 pairs of P. multocida collected during consecutive years from the same 22 turkey farms, 21 pairs (63.6%) were the same and 12 pairs (36.4%) were different. Of the 15 pairs of P. multocida collected with an interval of one or more years between them from the same 14 turkey farms, only four pairs (26.7%) were the same and 11 pairs (73.3%) were different. There did not appear to be any relationship between the DNA fingerprint profiles and the typing of their somatic antigens because, although 44 pairs of isolants had the same DNA fingerprint profile and somatic antigenic type, 42 pairs differed in these parameters when all pairs were combined.  相似文献

Blood was collected from 6 adult bitches (3 pregnant and 3 nonpregnant) daily for 59 days after the onset of diestrus, and serum progesterone concentrations were determined. The bitches were hysterectomized at various times during diestrus [diestrus day (DD) 5, 15, or 25] to determine whether the uterus has a role in maintaining luteal function in the pregnant or nonpregnant bitch. Concentrations of serum luteinizing hormone (LH) were also measured to determine whether LH concentrations decrease when luteal function abates (experiment 1). All 6 bitches in experiment 1 had progesterone concentrations greater than 1.0 ng/ml during the 2-month period, indicating that neither the pregnant nor nonpregnant uterus was necessary for maintaining luteal function in the bitch. Concentrations of LH appeared to increase in the serum of 5 of the 6 bitches when progesterone concentrations were declining, indicating that cessation of luteal function does not appear to result from a lack of LH. Blood was collected from the 6 bitches used in experiment 1 twice weekly during the 2nd diestrus after hysterectomy (experiment 2) to determine whether alterations in luteal function that might be observed during experiment 1 were due to some aspect of the surgical procedure, rather than to absence of a uterus. Progesterone concentrations in the serum were lower (P less than 0.05) during the 10 days after surgery in the 2 bitches hysterectomized on DD 5 than in the 4 bitches remaining intact during this period (experiment 1).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Strips of trachealis muscle were dissected from the mid-cervical portion of the trachea from horses that were free of respiratory tract disease. The epithelium and mucosa were removed from one group of tissues and were left intact in a second group of tissues. Each tissue was suspended in a bath filled with Krebs-bicarbonate solution that was aerated with 5% CO2 in oxygen and maintained at 37 degrees C. Isometric tension was continuously recorded. The contractile response to square-wave electrical stimulations increased as frequency (3, 5, 10, 15, 20, 25, and 30 Hz), voltage (10, 15, 18, and 25 V), and pulse duration (0.2, 0.5, 1.0, 1.5, and 2.0 ms) increased in tissues with the epithelium and mucosa intact. A stimulus of 18 V, 20 Hz, and 0.5 ms induced maximal contraction. Atropine (10(-6) M) abolished the response to 18 V and 0.5 ms at all frequencies. The increase in active isometric tension was concentration dependent when acetylcholine (10(-9) to 10(-4) M) was added to the baths in 0.5-logarithmic increments. Tissues that were contracted in response to acetylcholine (10(-5) M) had a concentration-dependent decrease in active isometric tension when isoproterenol was added to the baths in 0.5-logarithmic increments (10(-9) to 10(-4) M). The contraction and relaxation curves were qualitatively similar, but quantitatively different in tissues with and without the epithelium and mucosa. Removing the epithelium and mucosa increased the contractile response to acetylcholine at bath concentrations of 3.1 x 10(-7) M and 10(-6) M. Nitrification in a highly active Al horizon of a balsam fir forest floor soil can be greatly inhibited by an aqueous methanol extract of the forest floor. This extract was fractionated in an attempt to identify the compounds responsible for the inhibition. Condensed tannins, smaller molecular weight phenolics, and their distribution on particulate matter in the extract were the most important inhibiting components of the extract. When all phenolic material was removed from the extract, the remaining solution stimulated nitrification.  相似文献

Pseudorabies virus was inoculated by various routes into dogs to determine the relationship of pseudorabies virus to the development of heart lesions. Electrocardiograms and serum samples for lactate dehydrogenase isoenzymes were taken twice daily. Transitory and persistent arrhythmias were a consistent finding. Heart lesions were noted within each of the inoculated groups. These changes varied from severe multifocal areas of hemorrhage and myocardial degeneration to small zones of myolysis. A ganglioneuritis of the stellate ganglia and autonomic ganglia within the heart were also consistent findings. Herpes-like viral particles were found by electron microscopy in various autonomic ganglia and in myocardial endothelial cells. No viral particles were found in myocardial cells. Significant increases in lactate dehydrogenase-1 were noted. It was concluded that fatal arrhythmias resulted from pseudorabies virus infections in the dog after the occurrence of myocardial and ganglionic lesions.  相似文献

Four sheep inoculated with mixtures of bovine leukemia virus (BLV) and milk that was treated by a simulated high-temperature short-time pasteurization procedure did not become infected with BLV or develop tumor. Four sheep inoculated with unpasteurized BLV-mil mixtures became infected with BLV and 3 died with tumor at 15, 21, and 27 months, respectively, after inoculation. Fluid from a BLV-infected cell culture was heated to 56, 60, 65, and 73 C for 1/2 minute and 1 minute (in separate trials) and transferred to noninfected cells. Culture fluid heated to 60 C and higher did not infect the cells.  相似文献

A rapid and specific direct fluorescent antibody test was developed for the identification of group E Streptococci (GES). Tests for specificity included an inhibition test and application of the conjugate to smears of Staphylococcus aureus, Pasteurella multocida, and streptococci of Lancefield's groups, A, B, C, D, E (types I, IV and untypeable), F and G. With the inhibition test, a specific blocking reaction occurred and the conjugate cross-reacted only with group C Streptococci (GCS). Adsorption with GCS eliminated the cross-reaction and rendered the conjugate specific for GES.  相似文献
