The new combination of isocratic high performance liquid chromatography (HPLC) with on line UV spectrum detection via a diode array configuration has been applied to the detection and identification of anabolics present in application sites of cattle. Combination of the characteristic retention time in the HPLC chromatogram and a comparison of the full spectrum between 190-400 nm of the anabolic components with that of a standard resulted in a very reliable identification. By means of this method 117 samples of application sites were investigated for the presence of anabolic residues. Of the xenobiotic anabolics , 19-nortestosterone (NT) was found most frequently (in 96 cases), whereas diethylstilbestrol (DES) was found in only 11 cases. In all samples the identification of NT and DES was confirmed by high resolution gas chromatography-mass spectrometry (GCMS).  相似文献

Staphylococcus aureus mastitis is an important cause of economic loss for the dairy industry. Control programs rely on the timely and accurate identification of positive quarters. The effects of sampling time and sample handling were examined in an attempt to improve the accuracy of detection of S. aureus. Premilking and postmilking milk samples were collected from 55 lactating quarters with subclinical S. aureus infection. Each sample was divided into 2 aliquots; one of which was cultured fresh, the other was frozen at -20 degrees C for 14 days before being cultured. Analysis of variance was used to determine the effect of sampling time (premilking vs postmilking) and sample handling (fresh vs frozen) on the detection of S. aureus, as measured by the mean category for colony-forming units per millilitre (cfu/mL). A stratified analysis was required, due to interaction between sampling time and sample handling. Only a fresh postmilking sample was inferior, yielding a lower mean category for cfu/mL (P < 0.05). The ability to detect S. aureus in quarters with subclinical intramammary infection, as measured by the mean category of cfu/mL, was maximized in fresh or frozen premilking samples and in frozen postmilking samples.  相似文献

A survey was carried out in The Netherlands to determine the prevalence of benzimidazole resistance in sheep nematodes. It was shown that 47 per cent of the 59 investigated flocks harboured trichostrongylid populations with resistance to benzimidazoles, as demonstrated by an LD50 value of 0.12 micrograms ml-1 thiabendazole or higher in an in vitro egg hatch assay. On these farms Haemonchus contortus was found to be the dominant species. In two flocks resistant Ostertagia and, or, Trichostrongylus species populations were found. Farms with resistance were revisited to determine the anthelmintic efficacy in vivo by means of treatment and egg count. In only four of the 19 flocks which showed resistance in vitro was an efficacy of less than 95 per cent in vivo achieved. This means that there is a discrepancy between the in vivo and in vitro results. Resistance was found more frequently on pure sheep farms than on mixed farms. It is suggested that mixed farms offer more refuge for susceptible H contortus populations.  相似文献

Markers flanking DNA regions, where quantitative trait loci (QTL) have been previously spotted, can be used to trace the common inheritance of major genes for a better definition of covariances among animals. A practical approach to the use of marker data to refine the additive relationship matrix used in the traditional best linear unbiased prediction (BLUP) methodology is presented. The technique allows the number of the mixed model equations to be kept to an animal level, blending polygenic pedigree data with marker haplotype information. The advantage of this marker-assisted selection (MAS) approach over BLUP selection has been assessed through a stochastic simulation. A finite locus model with 32 independent biallelic loci was generated with normally distributed allelic effects. The heritability of the trait, measured on both sexes and on females only, was set to 0.2 and 0.5. Five-allelic markers 2, 10 and 20 cM apart, bracketed the QTL with the largest effect on the trait, accounting for 17% of the genetic variance. The bracketed QTL had two or eight alleles and its position was undefined within the bracket. Results show a moderate 2% advantage of MAS over BLUP in terms of higher genetic response when trait was recorded on both sexes and heritability was 0.2. The benefit is in the short term, but it lasts longer with polyallelic QTL. When the trait was recorded on females only, MAS produced only a small and insignificant genetic gain, but reduced the overall inbreeding in the population. Rams have been immunized against an infection of their genitalia by Histophilus ovis. An alum-precipitated antigen and an antigen plus Freund's complete adjuvant proved equally effective. An injection of live H. ovis into the epididymal tissue proved to be a better method of challenging immunity than an injection into the vas deferens. It was shown that cell-mediated immunity, as evidenced by tests for lymphocyte transformation, the presence of a macrophage migration inhibition factor and a delayed hypersensitivity skin reaction did not play a role in the resistance, nor did specific IgG antibodies have any protective influence. It was shown that neutrophils play a cardinal role in the immunity against H. ovis infection in so far as they phagocytize and destroy the organisms and are attracted to them by chemotaxis in immune animals.  相似文献
