The inflammatory bowel diseases Crohn's disease and ulcerative colitis are common, chronic disorders that cause abdominal pain, diarrhea, and gastrointestinal bleeding. To identify genetic factors that might contribute to these disorders, we performed a genome-wide association study. We found a highly significant association between Crohn's disease and the IL23R gene on chromosome 1p31, which encodes a subunit of the receptor for the proinflammatory cytokine interleukin-23. An uncommon coding variant (rs11209026, c.1142G>A, p.Arg381Gln) confers strong protection against Crohn's disease, and additional noncoding IL23R variants are independently associated. Replication studies confirmed IL23R associations in independent cohorts of patients with Crohn's disease or ulcerative colitis. These results and previous studies on the proinflammatory role of IL-23 prioritize this signaling pathway as a therapeutic target in inflammatory bowel disease.  相似文献

Organic dormancy, the inability to germinate under favorable conditions, is a common problem in many crop species and their wild relatives, leading to more variable emergence, plant density, and growth rates, thus increasing costs, and lowering yield. To overcome these problems, several different methods have been developed for various crop and model plants. However, in the emerging crop being bred from wild and semi-domesticated Silphium, no such method has been established thus far. The objective of this study was to identify a dormancy-breaking assay to increase the seed germination rate. Seeds of three different Silphium integrifolium Michx. genotypes were treated with five chemical and one cold treatment, and dried before or after the treatment at 27 °C or 40 °C. Untreated, dried seeds were taken as control. Seeds soaked for 24 h in a ethephon/potassium nitrate solution followed by a 72 h drying step at 40 °C showed an increase of germination to up to 90?±?2% compared to control seeds (3?±?0% and 5?±?1%). We identified the minimum time frame of cold stratification needed to enhance seed germination in Silphium, and found that cold stratification was nearly as good as the chemical treatment. Our results provide two alternate ways to treat S. integrifolium seeds for breaking organic dormancy, which will help to facilitate future research in the Silphium domestication community. The fact that our optimal treatments were similar to protocols developed for sunflower suggests that these methods may also be applicable to many related economically important Asteraceae species.  相似文献

The construction materials industry faces major challenges since 2013 when the European Construction Products Regulation was implemented, especially in the sector of environmentally friendly construction products. This study determined concentrations of leachable inorganic and organic compounds from microfine cement paste with and without superplasticiser addition. Furthermore, the leached superplasticiser amounts were detected via LC-MS. The multi-method approach was supplemented by ecotoxicological assays. Phytotoxicity was tested with white mustard (Sinapis alba) and cress (Lepidium sativum). The mutagenic and genotoxic potential of the leachates was tested with the Ames fluctuation assay and the umuC assay. As leaching protocol, the European horizontal dynamic surface leaching test was used. The cement paste samples with superplasticiser followed the wash-off effect with a total organic carbon release up to 43 mmol/m2, whereas the release of samples without superplasticiser was driven by diffusion. The ecotoxicological assays showed a clear time depending behaviour. No cytotoxicity and mutagenicity could be observed; anyhow some leachates show minor genotoxic potential. In all tests, a clear difference between the samples with and without superplasticiser could be detected.This study clearly demonstrates the importance of further studies in the field of leaching of construction products.  相似文献

This study aimed at quantifying the consequences of reduced precipitation and plant diversity on soil microbial community functioning in a Mediterranean shrubland of southern France. Across a natural gradient of shrub species diversity, we established a total of 92 plots (4 × 4 m) with and without a moderate rain exclusion treatment of about 12 % of total precipitation. Shrub diversity included all possible combinations of the four dominant species (Cistus albidus, Quercus coccifera, Rosmarinus officinalis, and Ulex parviflorus). Respective leaf litter mixtures of these species combinations were exposed in all plots over 2 years. We quantified how litter species richness and the reduction in precipitation affected the soil microbial substrate utilization (measured by CO2 evolution using the MicroResp method) on soil samples collected underneath each individual litter mixture after 1 and 2 years of decomposition. Moderate precipitation reduction had a minor impact, but litter species richness and the dissimilarity in phenolic concentrations (estimated using Rao’s quadratic entropy) showed a positive effect on the diversity of substrates metabolized by the microbial communities. Moreover, litter species richness increased soil microbial activity by increasing the catabolic diversity of the soil microbial community. These effects were mostly driven by the presence of Quercus and Ulex leaf litter, which at the same time reduced microbial metabolic dominance, while the presence of Rosmarinus had opposite effects. Our data suggest that plant species loss can have stronger effects on the functioning of soil microbial communities than moderate drought, with potentially important feedbacks on biogeochemical cycling in Mediterranean shrubland ecosystems.  相似文献

Setaria viridis (L.) P. Beauv. and its domesticated form, S. italica (L.) P. Beauv., have been developed over the past few years as model systems for C4 photosynthesis and for the analysis of bioenergy traits. S. viridis is native to Eurasia, but is now a ubiquitous weed. An analysis of the population structure of a set of 232 S. viridis lines, mostly from North America but also comprising some accessions from around the world, using 11 SSR markers, showed that S. viridis populations in the US largely separate by latitude and/or climatic zone. S. viridis populations from the Northern US and Canada (north of 44°N) group with accessions from Western Europe, while populations in the Mid and Southern US predominantly group with accessions from Turkey and Iran. We hypothesize that S. viridis in the US was most likely introduced from Europe, and that introductions were competitive only in regions that had climatic conditions that were similar to those in the regions of origins. This hypothesis is supported by the fact that Canadian S. viridis lines were fast cycling and undersized when grown in the Mid-Western and Southern US compared to their morphology in their native environment. A comparison of the population structure obtained with 11 SSR markers and ~40,000 single nucleotide polymorphisms (SNPs) in a common set of S. viridis germplasm showed that both methods essentially yielded the same groupings, although admixture was identified at a higher frequency in the SNP analysis. Small numbers of SSR markers can thus be used effectively to discern the population structure in this inbreeding species.  相似文献

BackgroundIn contrast to human medicine, only a small number of serum tumor markers are established in veterinary medicine even though they are a non-invasive diagnostic tool.ObjectivesThis study examined whether survivin could be suitable as a potential canine serum tumor marker.MethodsThis study measured the serum survivin concentrations of dogs with mammary tumors (n = 33), squamous cell carcinoma (n = 9), soft-tissue sarcoma (n = 18) and multicentric lymphoma (n = 22), using a commercially available, competitive immunoassay kit (BlueGene). The serum survivin concentrations were compared with those of a healthy control group (n = 20) and a control group of dogs with non-neoplastic diseases (n = 17).ResultsDogs with malignant tumors had serum survivin concentrations between 15 and 5,906 pg/mL (median, 72 pg/mL), those in the healthy group ranged from 7 to 99 pg/mL (median, 21 pg/mL) and those in the group of dogs suffering from non-neoplastic diseases from 15 to 93 pg/mL (median, 42 pg/mL). The differences in the survivin concentrations between the healthy dogs and dogs with malignant tumors and between the dogs with non-neoplastic diseases and those with malignant tumors were significant (p < 0.001 and p = 0.006, respectively).ConclusionsThe serum survivin concentrations in dogs with malignant tumors, with some exceptions, are higher than in dogs with benign tumors and dogs that do not suffer from a malignancy. Therefore, survivin can provide information on the presence of malignant tumors and be used as a tumor marker in dogs.  相似文献

Effective rooting depth, percolation water, and nitrate leaching in deeply developed loess soils of a water‐shortage area In 14 deeply developed loess soils, high amounts of mineral nitrogen (N) were measured within the first meter, whereas several nitrate depth profiles up to more than three meters resulted in low and medium nitrate values. The maximum depth of water uptake was measured in two years on four representative sites with regard to soil and crop properties. The maximum depth of water uptake was always considerably deeper than 200 cm, with a maximum of 290 cm (alfalfa). It is assumed that roots take up nitrogen even in this depth. The calculation of the effective rooting depth resulted in noticeably higher values (for wheat between 160 cm and 185 cm) than those given by the ”︁German Instructions for Soil Mapping” (AG Boden, 1994), the ”︁Regulations of the German Organisation for Water Management and Land Improvement” (DVWK, 1995) or the ”︁German Institute for Standardization” (DIN, 1998). As a result of low annual precipitation (normally less than 600 mm), only a minor part of the high amounts of nitrate within the root zone was leached into deeper soil layers. We conclude that it is not possible to predict the potential groundwater pollution with nitrate on the basis of the mineral N content in the first meter of the soil profile.  相似文献

Hepatitis E virus (HEV) is the causative agent of an acute and in most cases self‐limiting hepatitis. Of the four major HEV genotypes that infect humans, genotype 3 and 4 are zoonotic and have been identified in humans but predominantly in pigs and wild boar, which are considered the main reservoirs. However, the known host range of zoonotic HEV may be increasing to comprise additional species, including companion animals. Several studies have identified contact with dogs as a risk factor for HEV infection in humans, yet information on the occurrence of HEV in Swiss dogs is lacking. To examine a possible risk of exposure, this study was designed to assess the seroprevalence of HEV in 84 Swiss dogs. Serum and plasma samples collected from four veterinary clinics were screened for HEV‐specific antibodies by HEV‐antibody ELISA test kit. In addition, information of 22 dogs regarding the country of origin, the type of dog feed and any history of hunting was recorded. Samples from seropositive animals were also screened for the presence of HEV RNA by quantitative real‐time RT‐PCR (qRT‐PCR). Overall, 38% (32 of 84) of the dogs tested seropositive for anti‐HEV, indicating exposure to HEV. Among the 22 dogs for which information was available, HEV‐specific antibodies were detected in three of five dogs that were born abroad, in one of two dogs that were fed a raw meat‐based diet, and in one hunting dog. No viral RNA could be detected in any of the serum and plasma samples; thus, the genotype of the strains remained undetermined. This study provides further evidence for canine exposure and susceptibility to HEV and highlights the need to further assess the risks of HEV transmission to humans with contact to dogs.  相似文献

This paper examines the spatial-temporal evolution of urban spatial structure across 269 Chinese prefectural cities from 2002 to 2019. Our analysis identifies a consistent trend toward a more polycentric configuration in the 25 Chinese mega-cities during this period, primarily due to population growth and a supportive policy environment. However, the evolutionary pathways of small- and medium-sized cities unfolded in a rather complex and diverse manner, with some becoming more polycentric while the majority adhering to a monocentric trajectory. In these cases, population growth is usually associated with a more monocentric pattern, characterized by rapid expansion of the urban core, while polycentric development is primarily attributed to specific spatial policies that support the emergence of subcenters. We conclude that polycentric development, while potentially suitable for mega-cities to alleviate diseconomies of scale, may be less appropriate for small- and medium-sized cities as it may constrain growth associated with agglomeration economies. We suggest that the development and implementation of regional spatial policy should be considerate of local historical paths and contextual factors. Finally, we propose a stylized framework to more accurately reflect the diverse and complex nature of urban spatial structure evolution in Chinese prefectural cities.  相似文献
