Nitrous oxide (N2O) is a high‐impact greenhouse gas. Due to the scarcity of unmanaged forests in Central Europe, its long‐term natural background emission level is not entirely clear. We measured soil N2O emissions in an unmanaged, old‐growth beech forest in the Hainich National Park, Germany, at 15 plots over a 1‐year period. The average annual measured N2O flux rate was (0.49 ± 0.44) kg N ha–1 y–1. The N2O emissions showed background‐emission patterns with two N2O peaks. A correlation analysis shows that the distance between plots (up to 380 m) does not control flux correlations. Comparison of measured data with annual N2O flux rates obtained from a standard model (Forest‐DNDC) without site‐specific recalibration reveals that the model overestimates the actual measured N2O flux rates mainly in spring. Temporal variability of measured N2O flux was better depicted by the model at plots with high soil organic C (SOC) content. Modeled N2O flux rates were increased during freezing only when SOC was > 0.06 kg C kg–1. The results indicate that the natural background of N2O emissions may be lower than assumed by most approaches.  相似文献

The physical properties of a Luvisol derived from loess near Bonn, Germany, under different long‐term fertilization treatments were examined. For the investigation of the impact of farmyard manure (FYM) on soil strength at the mesoscale (100 to 300 cm3 soil cores), undisturbed samples were taken from two different depths (10 and 40 cm), either with no fertilization at all, with full mineral fertilization, with FYM only, and with both mineral and organic fertilization. We investigated hydraulic and mechanical parameters, namely precompression stress, pore‐size distribution, saturated hydraulic and air conductivity, and calculated pore connectivity. Long‐term organic fertilization resulted in significantly more and coarser pores which in addition were more conductant and mechanically stronger by trend. Mineral fertilization also increased pore volume by trend but not pore functionality. Mechanical strength generally increased with fertilization by trend, however, was reduced again when organic and mineral fertilization were combined. Nonetheless, FYM led to relatively higher soil strength as the FYM‐treated plots with lower bulk density attained similar soil strength as the unfertilized but denser plots and thus supported the soil‐improving impact of organic amendments. The subsoil physical properties were rather unaffected by fertilization, but were dominated by texture.  相似文献

We examined the design of a course that aims to ease the transition from pre-clinical learning into clinical work. This course is based on the premise that many of the difficulties with which students are confronted in this transition result from a lack of experience in applying knowledge in real practice situations. It is focused on the development of competence in solving clinical problems; uses an instructional model with alternating clinical practicals, demonstrations, and tutorials; and extends throughout the last pre-clinical year. We used a "proof-of-concept" approach to establish whether the core principles of the course design are feasible with regard to achieving the intended results. With the learning functions and processes as a frame of reference, retrospective analysis of the course's design features shows that this design matches the conditions from theories of the development of competence in solving clinical problems and instructional design. Three areas of uncertainty in the design are identified: the quality of the cases (information, openness), effective teaching (student and teacher roles), and adjustment to the development of competence (progress, coherence).  相似文献

ABSTRACT: In the rapidly changing context of research on animal health, INRA launched a collective discussion on the challenges facing the field, its distinguishing features, and synergies with biomedical research. As has been declared forcibly by the heads of WHO, FAO and OIE, the challenges facing animal health, beyond diseases transmissible to humans, are critically important and involve food security, agriculture economics, and the ensemble of economic activities associated with agriculture. There are in addition issues related to public health (zoonoses, xenobiotics, antimicrobial resistance), the environment, and animal welfare.Animal health research is distinguished by particular methodologies and scientific questions that stem from the specific biological features of domestic species and from animal husbandry practices. It generally does not explore the same scientific questions as research on human biology, even when the same pathogens are being studied, and the discipline is rooted in a very specific agricultural and economic context.Generic and methodological synergies nevertheless exist with biomedical research, particularly with regard to tools and biological models. Certain domestic species furthermore present more functional similarities with humans than laboratory rodents.The singularity of animal health research in relation to biomedical research should be taken into account in the organization, evaluation, and funding of the field through a policy that clearly recognizes the specific issues at stake. At the same time, the One Health approach should facilitate closer collaboration between biomedical and animal health research at the level of research teams and programmes.  相似文献

Lawsonia (L.) intracellularis, Brachyspira (B.) hyodysenteriae and B. pilosicoli are important pathogens in domestic pig production world-wide, responsible for porcine intestinal adenomatosis, swine dysentery, and porcine intestinal spirochetosis, respectively. Conventional PCR is the major diagnostic tool in the detection of the three pathogens, but the sole detection of bacterial DNA might lead to misinterpretations of results with respect to their clinical relevance, especially with mixed infections. Thus, the present study targeted the detection and quantification of the three pathogens in samples from herds with a case history of diarrhoea. Herds and samples were selected by the practitioners on a voluntary basis. Results were based on 1176 individual samples from 95 herds from Southern Germany. The pathogens were detected simultaneously by multiplex real-time PCR. The overall prevalence for L. intracellularis, B. hyodysenteriae and B. pilosicoli was 12.6%, 8.4% and 3.2% in faecal samples and 48.4%, 24.2% and 31.6% in herds, respectively. Sixty one percent, 82.6%, and 73.4% of herds positive for L. intracellularis, B. hyodysenteriae, and B. pilosicoli, respectively, had mixed infections. Median log values of DNA equivalents/g of faeces for L. intracellularis, B. hyodysenteriae and B. pilosicoli were 3.3, 5.9 and 3.2, with maxima of 8.3, 8.0 and 6.3, respectively. Within herd prevalence of B. hyodysenteriae and B. pilosicoli as well as the load of B. hyodysenteriae were significantly associated with the severity of diarrhoea.  相似文献

Staphylococcus aureus and coagulase negative staphylococci (CNS) were isolated from ovine and caprine mastitis milk samples originating from more than 40 Swiss farms. CNS dominated as causal microorganisms of mastitis in small ruminants. By restriction fragment length polymorphism (RFLP) analysis of the groEL gene and sequencing of 16S rDNA, various CNS species were identified, albeit certain of them predominated. For susceptibility testing of mastitis pathogens to selected antibiotics, minimal inhibitory concentrations were determined. Of the 67 S. aureus and 208 CNS strains, 31.3 % and 8.2 % were resistant to penicillin, 29.9 % and 1.0 % to ampicillin, 1.5 % and 10.6 % to erythromycin, and 3.0 % and 7.7 % to tetracycline, respectively. Moreover, 10 CNS strains (4.8 %) were resistant to oxacillin and one CNS strain to sulfamethoxazole/trimethoprim. The results obtained describe for the first time the resistance situation of mastitis pathogens from sheep and goats in Switzerland. However, accompanying and preventing measures are also of importance in mastitis control of small ruminants.  相似文献

In this project we report on the angiotensin I-converting enzyme (ACE)-inhibitory activity of a bovine gelatin hydrolysate (Bh2) that was submitted to further hydrolysis by different enzymes. The thermolysin hydrolysate (Bh2t) showed the highest in vitro ACE inhibitory activity, and interestingly a marked in vivo blood pressure-lowering effect was demonstrated in spontaneously hypertensive rats (SHR). In contrast, Bh2 showed no effect in SHR, confirming the need for the extra thermolysin hydrolysis. Hence, an angiotensin I-evoked contractile response in isolated rat aortic rings was inhibited by Bh2t, but not by Bh2, suggesting ACE inhibition as the underlying antihypertensive mechanism for Bh2t. Using mass spectrometry, seven small peptides, AG, AGP, VGP, PY, QY, DY and IY or LY or HO-PY were identified in Bh2t. As these peptides showed ACE inhibitory activity and were more prominent in Bh2t than in Bh2, the current data provide evidence that these contribute to the antihypertensive effect of Bh2t.  相似文献

Mining activities generate spoils and effluents with extremely high metal concentrations of heavy metals that might have adverse effects on ecosystems and human health. Therefore, information on soil and plant metal concentrations is needed to assess the severity of the pollution and develop a strategy for soil reclamation such as phytoremediation. Here, we studied soils and vegetation in three heavily contaminated sites with potential toxic metals and metalloids (Zn, Pb, Cd, As, TI) in the mining district of Les Malines in the Languedoc region (southern France). Extremely high concentrations were found at different places such as the Les Aviniéres tailing basins (up to 160,000 mg kg?C1 Zn, 90,000 mg kg?C1 Pb, 9,700 mg kg?C1 of As and 245 mg kg?C1 of Tl) near a former furnace. Metal contamination extended several kilometres away from the mine sites probably because of the transport of toxic mining residues by wind and water. Spontaneous vegetation growing on the three mine sites was highly diversified and included 116 plant species. The vegetation cover consisted of species also found in non-contaminated soils, some of which have been shown to be metal-tolerant ecotypes (Festuca arvernensis, Koeleria vallesiana and Armeria arenaria) and several Zn, Cd and Tl hyperaccumulators such as Anthyllis vulneraria, Thlaspi caerulescens, Iberis intermedia and Silene latifolia. This latter species was highlighted as a new thallium hyperaccumulator, accumulating nearly 1,500 mg kg?C1. These species represent a patrimonial interest for their potential use for the phytoremediation of toxic metal-polluted areas.  相似文献

Hyperspectral remote sensing for monitoring horticultural production systems requires the understanding of how plant physiology, canopy structure, management and solar elevation affect the retrieved canopy reflectance during different stages of the phenological cycle. Hence, the objective of this study was to set up and to interpret a hyperspectral time series for a mature and healthy citrus orchard in the Western Cape province of South Africa considering these effects. Based on the remotely sensed data, biophysical parameters at the canopy level were derived and related to known observed physiological and phenological changes at the leaf level and to orchard management. Fractions of mature fruit, flowers, and sunburnt leaves were considered, and indices related to canopy structure chlorophyll content and canopy water status were calculated.Results revealed small cover fractions of mature fruit, flowers and sunburnt leaves of respectively 2.1%, 3.1% and 7.0%, but the high spectral contrast between flowers and leaves allowed a successful classification of flowering intensity. Furthermore, it was shown that canopy level time series of vegetation indices were sensitive to changes in solar elevation and soil reflectance which could be reduced by applying an empirical soil line correction for the most affected indices. Most trends in vegetation indices at the canopy level could be explained by a combination of changes at the leaf level (chlorophyll, carotenoids, dry matter), changes in canopy structure (leaf area index and leaf angle distribution) and changes in cover fractions of vegetative flushes, flowers and sunburnt leaves. The transformed chlorophyll absorption ratio index over the optimised soil adjusted vegetation index (MCARI/OSAVI) was best related to leaf level trends in chlorophyll content. Seasonal changes in the photochemical reflectance index (PRI) were linked to inverse changes in the carotenoid-to-chlorophyll ratio. Canopy structure indices (the modified triangular vegetation index or MTVI2 and the standardized leaf area index determining index or sLAIDI) were sensitive to changes in leaf area index, average leaf angle as well to management interactions (pruning and harvest). Canopy water status was highly impacted during the spring flush due to expanding leaves that concealed trends in the underlying mature leaves. Seasonal trends in soil and weeds reflectance were related to changes in volumetric soil water content and to the earlier and reduced growth period of non-irrigated weeds.  相似文献

This study describes the diurnal and seasonal dynamics of the canopy reflectance, water use and water status of Midknight Valencia citrus trees under semi-arid conditions. Hyperspectral canopy reflectance data was collected on 30 trees at monthly intervals over a period of 16 months in a commercial orchard in South Africa. The mean canopy reflectance in the wavelength range 350-2500 nm followed a clear seasonal trend influenced by environmental conditions and tree phenology. Mean monthly reflectance peaked in summer (∼22%) while the lowest value (∼15%) was reached in winter with the seasonal changes in the sun's position accounting for a significant proportion of the variations. A sensitivity analysis of a Penman-Monteith transpiration model showed that water use by individual trees changed by up to 13% when the canopy reflectance was varied over the seasonal range of measured values. This suggested that the seasonal changes in tree water use influenced the seasonal trend of the canopy reflectance. Thus monitoring the canopy reflectance of citrus trees could offer information on the tree water status. To test this, sap flow data of water uptake and loss by the trees were compared with the canopy spectra. Sap flow data showed a heavy reliance by the citrus trees on the internally stored water with up to 25% of the daily total transpiration withdrawn from the trees’ internal water storage pools when soil water was limited. This depletion of internally stored water, and hence the change in tree water status, was detected using spectral indices based on the first order derivatives of the canopy reflectance centered at two and, at most, four spectral bands. We conclude that even if citrus trees are evergreen, their canopy reflectance changes significantly throughout the year with a considerable impact on tree energy balance and water use. In addition, the contribution of the internally stored water to daily transpiration is a possible indicator of drought stress for citrus trees detectable from changes in canopy reflectance and it has potential applications in irrigation scheduling using canopy level spectral information.  相似文献
