Objective To develop an ultrasound‐guided technique for retrobulbar nerve block in horses, and to compare the distribution of three different volumes of injected contrast medium (CM) (4, 8 and 12 mL), with the hypothesis that successful placement of the needle within the retractor bulbi muscle cone would lead to the most effective dispersal of CM towards the nerves leaving the orbital fissure. Study design Prospective experimental cadaver study. Animals Twenty equine cadavers. Methods Ultrasound‐guided retrobulbar injections were performed in 40 cadaver orbits. Ultrasound visualization of needle placement within the retractor bulbi muscle cone and spread of injected CM towards the orbital fissure were scored. Needle position and destination of CM were then assessed using computerized tomography (CT), and comparisons performed between ultrasonographic visualization of orbital structures and success rate of injections (intraconal needle placement, CM reaching the orbital fissure). Results Higher scores for ultrasound visualization resulted in a higher success rate for intraconal CM injection, as documented on the CT images. Successful intraconal placement of the needle (22/34 orbits) resulted in CM always reaching the orbital fissure. CM also reached the orbital fissure in six orbits where needle placement was extraconal. With 4, 8 and 12 mL CM, the orbital fissure was reached in 16/34, 23/34 and 28/34 injections, respectively. Conclusion and clinical relevance The present study demonstrates the use of ultrasound for visualization of anatomical structures and needle placement during retrobulbar injections in equine orbits. However, this approach needs to be repeated in controlled clinical trials to assess practicability and effectiveness in clinical practice.  相似文献

In the mountain rainforest region of the South Ecuadorian Andes natural forests have often been converted to pastures by slash-and-burn practice. With advanced pasture age the pasture grasses are increasingly replaced by the tropical bracken leading to the abandonment of the sites. To improve pasture productivity a fertilisation experiment with urea was established. The effects of urea on soil organic matter (SOM) mineralisation and microbial community structure in top soil (0–5 cm depth) of an active and abandoned pasture site have been investigated in laboratory incubation experiments. Either 14C- or 15N-labelled urea (74 mg urea-N kg−1 dw soil) was added to track the fate of 14C into CO2 or microbial biomass and that of 15N into the KCl-extractable NH4-N or NO3-N or microbial biomass pool. The soil microbial community structure was assessed using phospholipid fatty acid analysis (PLFA). In a second experiment two levels of 14C-labelled urea (74 and 110 mg urea-N kg−1 dw soil) were added to soil from 5 to 10 cm depth of the respective sites. Urea fertilisation accelerated the mineralisation of SOC directly after addition up to 17% compared to the non-fertilised control after 14 days of incubation. The larger the amount of N potentially available per unit of microbial biomass N the larger was the positive priming effect. Since in average 80% of the urea-C had been mineralised already 1 day after amendment, the priming effect was strong enough to cause a net loss of soil C. Although the structure of the microbial community was significantly different between sites, urea fertilisation induced the same alteration in microbial community composition: towards a relative lower abundance of PLFA marker characteristic of Gram-positive bacteria and a higher one of those typical of Gram-negative bacteria and fungi. This change was positively correlated with the increase in NH4, NO3 and DON availability. In addition to the activation of different microbial groups the abolishment of energy limitation of the microbes seemed to be an important mechanism for the enhanced mineralisation of SOM.  相似文献

Soil moisture and gaseous N-flux (N2O, N2) dynamics in Costa Rican coffee plantations were successively simulated using a mechanistic model (PASTIS) and two process-based models (NGAS and NOE). Two fertilized (250 kg N ha−1 y−1) coffee plantations were considered, namely a monoculture and a system shaded by the N2 fixing legume species Inga densiflora. In situ N2O fluxes were previously measured in these plantations. NGAS and NOE used specific microbial activities for the soils. To parameterize NGAS, we estimated N mineralization via in situ incubations and the contribution of heterotrophic soil respiration to total soil respiration. Potential denitrification rates and the proportion of denitrified N emitted as N2O were measured in the laboratory to define the values of NOE parameters, as well as nitrification rates and related N2O production rates for parameterizing both models. Soil moisture and both NGAS and NOE N2O fluxes were best modelled on an hourly time step. Soil moisture dynamics were satisfactorily simulated by PASTIS. Simulated N2O fluxes by both NGAS and NOE (3.2 and 2.1 kg N ha−1 y−1 for NGAS; 7.1 and 3.7 kg N ha−1 y−1 for NOE, for the monoculture and shaded plantations respectively) were within a factor of about 2 of the observed annual fluxes (4.3 and 5.8 kg N ha−1 y−1, for the monoculture and shaded plantations respectively). Statistical indicators of association and coincidence between simulated and measured values were satisfactory for both models. Nevertheless, the two models differed greatly in describing the nitrification and denitrification processes. Some of the algorithms in the model NGAS were apparently not applicable to these tropical acidic Andosols. Therefore, more detailed information about microbial processes in different agroecosystems would be needed, notably if process-oriented models were to be used for testing strategies for mitigating N2O emissions.  相似文献

Peatlands typically exhibit significant spatial heterogeneity which can lead to large uncertainties when catchment scale greenhouse gas fluxes are extrapolated from chamber measurements (generally <1 m2). Here we examined the underlying environmental and vegetation characteristics which led to within-site variability in both CH4 and N2O emissions and the importance of such variability in up-scaling. We also consider within-site variation in the controls of temporal dynamics. Net annual emissions (and coefficients of variation) for CH4 and N2O were 1.06 kg ha−1 y−1 (300%) and 0.02 kg ha−1 y−1 (410%), respectively. The riparian zone was a significant CH4 hotspot contributing ∼12% of the total catchment emissions whilst covering only ∼0.5% of the catchment area. In contrast to many other studies we found smaller CH4 emissions and greater uptake in chambers containing either sedges or rushes. We also found clear differences in the drivers of temporal CH4 dynamics across the site, e.g. water table was important only in chambers which did not contain aerenchymous plants. We suggest that depending on the heterogeneity of the site, flux models could be improved by incorporating a number of spatially distinct sub-models, rather than a single model parameterized using whole-catchment averages.  相似文献

The aim of this study was to evaluate the effect of organic amendment on the germination and growth patterns of Jatropha curcas L. on completely barren and degraded land in the Sahelian area of Burkina Faso. Prior to the field trials, laboratory germination tests were undertaken to explore the impact of different pre-treatments on germination of Jatropha seeds. Seeds soaked in water for 24?h had the highest mean rate of germination (86%) while seeds that were pre-treated with sulphuric acid did not germinate. The results of the field experiment showed that plant growth and biomass development were significantly enhanced by organic amendment compared to the control. With direct seeding, 20% of the plants treated with organic manure survived after 2?years, while all seedlings of the control plot perished. In the plantations, 30% of the untreated seedlings remained alive whereas only 5% of the plants survived with amendment. The trials in unfenced plots were decimated by livestock grazing and trampling 2?months after the beginning of the experiment. This emphasizes the need to protect Jatropha plants at an early stage of their development from roaming animals. Organic amendment attracted humivorous termites, which were destructive to the seedlings. The use of pesticides may be necessary to control this problem. When directly seeded, plants of the control plots demonstrated poor growth and became rapidly diseased, further accelerating their decline. The low survival rates (5–30%) and meagre seedling performance, even for the amended plots, may be an indication that Jatropha is unsuited to severely degraded lands like the zippelé, and cannot be expected to give good yields and the claimed environmental and socio-economic benefits. However, we recommend that the performance of Jatropha on the zippelé should be further tested with other soil and water conservation techniques (half-moon, tillage, etc.) that have been shown to enhance crop production and yield on degraded lands in the Sahelo-Sudanian zone of West Africa. The impact of seed provenance on the outcome of this study is unknown. Therefore, further experiments should embrace seeds from different sources, including genotypes that are more adapted to dry conditions and might therefore show improved performance.  相似文献

Leaf specific conductivity (LSC; the ratio of stem conductivity (K(P)) to leaf area (A(L))), a measure of the hydraulic capacity of the stem to supply leaves with water, varies with soil water content. Empirical evidence for LSC responses to drought is ambiguous, because previously published results were subject to many confounding factors. We tested how LSC of similar-sized trees of the same population, under similar climatic conditions, responds to persistently wet or dry soil. Scots pine (Pinus sylvestris L.) and pubescent oak (Quercus pubescens Willd.) trees were compared between a dry site and a wet site in the Valais, an inner alpine valley in Switzerland. Soil water strongly influenced A(L) and K(P) and the plant components affecting K(P), such as conduit radius, conduit density and functional sapwood area. Trees at the dry site had lower LSC than trees with the same stem diameter at the wet site. Low LSC in trees at the dry site was associated with a smaller functional sapwood area and narrower conduits, resulting in a stronger reduction in K(P) than in A(L). These observations support the hypothesis that trees maintain a homeostatic water pressure gradient. An alternative hypothesis is that relatively high investments in leaves compared with sapwood contribute to carbon gain over an entire season by enabling rapid whole-plant photosynthesis during periods of high water availability (e.g., in spring, after rain events and during morning hours when leaf-to-air vapor pressure deficit is small). Dynamic data and a hydraulic plant growth model are needed to test how investments in leaves versus sapwood and roots contribute to transpiration and to maximizing carbon gain throughout entire growth seasons.  相似文献

A goat flock grazing year-round on a meadow in a "TBE non-risk area" in Thuringia, Germany, with a history of only isolated human TBE cases was examined repeatedly for TBE virus-(TBEV)-specific antibodies and TBEV RNA between October 2008 and December 2009. Surprisingly, TBEV specific antibodies were detected in one goat, which had never left this area. To compare the results of a natural contact to TBEV with a defined contact to TBEV, two goats were immunized experimentally. Both animals developed TBEV-specific antibodies, one goat however in a delayed and reduced manner. In addition, 177 ticks were collected from the meadow in May and June 2009, and were examined by real-time RT-PCR. However, noTBEV RNA could be detected. The results suggest that goats can be used as TBEV sentinels in defined areas. To verify this observation further investigations with a large number of animals are recommended.  相似文献

Soil erosion is the main process leading to soil degradation on the Loess Plateau of China. The effects of soil‐erosion intensity (sheet, rill, and gully erosion) and different land use (140 y–old secondary forest site, 16 y–old bare site, 6 y–old succession site, and 43 y–old arable site) on gross and net N mineralization, soil organic‐carbon (SOC) turnover, the size and structure of the soil microbial community (phospholipid fatty acid analysis) were assessed. Erosion intensity in the bare plot increased from top slope (sheet erosion) to down slope (gully erosion). The more severe the soil erosion the stronger was the decline of SOC, total N, and microbial biomass (MB). The MBC/SOC ratio decreased whereas the metabolic quotient (qCO2) increased. Differences in nutrient turnover in the different erosion zones of the bare plot were not significant. The microbial community changed towards less Gram negative bacteria and relative more fungi in the gully‐erosion zone. In forest soils, qCO2 and the MBC/SOC ratio demonstrate a higher substrate‐use efficiency of the microbial biomass than in bare soils. Gross N mineralization and gross NH consumption clearly indicated a higher microbial activity in forest than in bare soils. Arable land use shifted the soil microbial community towards a higher relative abundance of fungi and a lower one of actinomycetes. During 6 y of natural succession on former bare plots, soil nutrient content and turnover as well as microbial biomass and structure developed towards forest conditions.  相似文献

Introduction

The European Water Framework Directive aims to achieve a good ecological and chemical status in surface water of European rivers by the year 2015. Since sediments and particulate matter act as secondary sources for pollutants, applied sediment toxicology is perceived to play a major role for obtaining new knowledge that can contribute to successful attainment of the goal. However, the existing bioassays for sediment toxicity analyses do not provide sufficient data concerning bioavailability of environmental pollutants. In this regard, there is an urgent need to combine sediment contact assays with gene expression analysis to investigate mechanism-specific sediment toxicity.

Purpose

The aim of the novel joint research project is to develop a eukaryotic test system, which can be used to investigate the ecotoxicological effects of contaminated sediments on gene expression level (DNA-array and RT-PCR). Current ecotoxicological research customarily involves a battery of bioassays to cover different toxicological endpoints (e.g., teratogenicity, genotoxicity, mutagenicity, Ah-receptor-mediated toxicity, neurotoxicity). In contrast, methods that detect alterations in gene expression offer deeper insight by elucidating how chemical exposure and/or environmental challenge affect multiple metabolic pathways leading to these particular kinds of toxic response. Gene expression profiles reflect the way cells and organisms adapt or respond to a changing environment.

Conclusion

The present project aspires to increase the fundamental molecular and physiological knowledge concerning the mode of action of environmental toxicants in zebrafish (Danio rerio). By working with partners from the academic and research institutions as well as from industry and waterway regulations, the success of this basic research-driven joint project in terms of development and implementation of novel sediment toxicity methods will be realized.  相似文献
