Abstract. Urea and ammonium were compared with and without nitrification inhibitors (nitrapyrin and dicyandiamide) as a possible means of reducing the nitrate concentration in winter lettuce grown in the glasshouse. Neither inhibitor had a significant effect on nitrate concentrations at the lower rate of applied nitrogen (138 kg ha−1 N). At the higher rate of urea (414 kg ha−1 N) dicyandiamide significantly reduced the nitrate concentration but this was accompanied by an unacceptable loss in yield and quality thought to be due to phytotoxic effects of the high level of residual ammonium. Nitrapyrin had no effect on the nitrate concentrations in lettuce. 相似文献

SUMMARY An outbreak of chlamydiosis was Investigated in workers at a duck farm and processing plant during winter 1989. Chlamydia psittaci was isolated from ducks, but there was little evidence of clinical chlamydiosis in the ducks. Serological tests showed that 76% of workers had been exposed to Infection, 12% recently, 16% had not been exposed and 8% had inconclusive test results. There was a strong association (P < 0.05) between years of employment and exposure to Infection. Infection appeared to be contracted during the first year of employment. Measures were Implemented to reduce shed contamination and control rodents and wild birds. At the same time, ducks were medicated, the processing plant was modified, work practices were altered and workers were advised about chlamydiosis to reduce the risk of human infection. 相似文献

Polymerase chain reaction (PCR) from paraffin-embedded tissues provides a powerful tool to amplify DNA from a variety of recent and archival material. Because DNA from paraffin-embedded samples is more degraded than from fresh material, the amplification of reference genes is essential to exclude false-negative results. This study describes the use of the proliferative cell nuclear antigen (PCNA) gene as a reference gene in a range of animal species and in humans. The PCNA-PCR to amplify a fragment extending from exon 5 through exon 6 and including the intervening intron 6 gave a reproducible pattern, with a 280-base pair (bp) band from canine, equine, bovine, ovine, and caprine samples showing high sequence homology. Porcine, guinea pig, tiger, and lion samples, however, gave an additional fragment of approximately 197 bp. The whole intron 6 from these fragments is missing, possibly representing a pseudogene. In feline samples only the 197-bp fragment could be detected. This study shows that the PCNA gene is highly conserved across a broad range of animal species and is well suited as an internal control for PCR analysis in veterinary medicine. 相似文献

To facilitate transfer of bruchid resistance to azuki bean (Vigna angularis) from its relatives an interspecific mapping population was made between rice bean, V. umbellata, and the related wild species V. nakashimae. The V. umbellata parent is completely resistant and V. nakashimae is completely susceptible to the bruchid beetle pests, azuki bean weevil (Callosobruchus chinensis) and cowpea weevil (C. maculatus). There is very low cross compatibility between V. umbellata and azuki bean. Therefore, V. nakashimae, that crosses with both V. umbellata and V. angularis without the need for embryo rescue, is used as a bridging species. A genetic linkage map was constructed based on an interspecific F2 mapping population between V. umbellata and V. nakashimae consisting of 74 plants. A total of 175 DNA marker loci (74 RFLPs and 101 SSRs) were mapped on to 11 linkage groups spanning a total length of 652 cM. Segregation distortion was observed but only three markers were not linked to any linkage group due to severe segregation distortion. This interspecific genome map was compared with the genome map of azuki bean. Of 121 common markers on the two maps, 114 (94.2%) were located on the same linkage groups in both maps. The marker order was highly conserved between the two genome maps. Fifty F2 plants that produced sufficient seeds were used for quantitative trait locus (QTL) analysis and locating gene(s) for C. chinensis and C. maculatus resistance in V. umbellata. The resistance reaction of these F2 plants differed between C. chinensis and C. maculatus. Both resistances were quantitatively inherited with no F2 plants completely susceptible to C. chinensis or C. maculatus. One putative QTL for resistance to each of these bruchid species was located on different linkage groups. Other putative QTLs associated with resistance to both C. chinensis and C. maculatus were localized on the same linkage group 1. Linked markers associated with the bruchid‐resistant QTL will facilitate their transfer to azuki bean breeding lines. 相似文献

Although water chemistry of precipitation and lakes in Nova Scotia is dominated by C1 from sea salt, correction for marine influence reveals that the dominant anion in acidified lakes is SO4. Atmospheric deposition of non-marine SO4 (SO4) and NO3- for the period 1977–1980 at 4 stations in southwest Nova Scotia averaged 47 meq SO4 * m?2 yr?1 and 21 meq NI3-m?2 yr?1 compared with 38 and 13 meq, respectively, for the average of 3 stations in the northeastern third of the province. Precipitation pH increased from 4.5 to 4.8 along the same axis. Almost 50% of the SO4 deposition occurred when storms came from the southwest, indicating low pressure tracks which pass south of major Canadian sources of S. SO4 * deposition in metropolitan Halifax (1982 bulk data) was 87 meq m?2 yr?1, due to local emissions of ca. 28 300 tonne S in the area, as well as LRTAP. Concurrent deposition of NO3-N was 15 meq m?2 yr?1 (2.1 kg ha?1 yr ?1). Loadings from SO4 deposition in the Halifax area amount to 42 kg ha?1 yr?1 and clearly exceed the federal guideline (M.O.I., 1983) of 20 kg ha?1 yr?1. Water chemistry of southwest, northeast, and Halifax area lakes show the same general SOI trends as observed for atmospheric deposition. In addition we find a positive relationship between SOI concentrations in the urban lakes and proximity to the center of the urban area. 相似文献

Thailand is a center of diversity for section Angulares of the Asian Vigna (genus Vigna subgenus Ceratotropis) and 4 Vigna species are cultivated in Thailand. Using newly collected wild and cultivated germplasm of Vigna from Thailand and outgroup accessions AFLP analysis was conducted to clarify genetic diversity and relationships. The results
suggest that cultivated V. umbellata and V. mungo evolved from wild relatives in a single domestication event. Vigna umbellata is poorly differentiated from its wild and weedy relatives compared to V. mungo. Results suggest northern Thailand and the neighboring Shan state, Myanmar, is the probable center of domestication for V. umbellata as wild accessions from this area and cultivated rice bean from a wide area in Asia are not greatly diverged. Vigna minima, V. tenuicaulis and V. exilis accessions in Thailand are well differentiated with considerable intra-specific variation. Vigna hirtella consists of two well differentiated subgroups, suggesting taxonomic revision may be necessary. Close genetic relationships
between V. radiata and V. grandiflora, and between V. mungo and V. trinervia are confirmed. Naturally growing V. mungo populations in northern Thailand appear to be true wild species as they are well differentiated from Indian wild and Thai
cultivated populations. The origin of naturally growing cowpea in Thailand needs to be further studied using a more comprehensive

A comparison is made of diatom remains in surficial lake sediments collected in 1971 and 1980. Changes of similar magnitude towards more acid tolerant assemblages are observed in four lakes studied in detail despite widely differing aquatic environments. No attempt is made to reconstruct past chemistry, the significant point being that at the most fundamental level, local lacustrine biological systems are being stressed to the point of alteration. The pattern of that alteration with emphasis on ‘simplification’ is discussed with the suggestion that rates of acidification may be as important as net changes. 相似文献

X-ray diffraction and diamond anvil techniques were used to measure the isothermal compressibility of K(3)C(60) and Rb(3)C(60), the superconducting, binary alkali-metal intercalation compounds of solid buckminsterfullerene. These results, combined with the pressure dependence of the superconducting onset temperature T(c) measured by other groups, establish a universal first-order relation between T(c) and the lattice parameter a over a broad range, between 13.9 and 14.5 angstroms. With the use of monopolar recordings for averaged evoked responses, detected signals in a vigilance task are associated with a late positive component which is absent for undetected signals as well as nonsignals. Bipolar recordings obscure the late positive component associated with detected signals.
